The advancement of human pluripotent stem cell (hPSC) has provided a source of human neural cells for disease modeling and transplantation study. Here, we demonstrated the application of human induced pluripotent stem cell (hIPSC)-derived midbrain dopamine (mDA) neurons to study the mechanism of neurodegeneration in Parkinson’s disease (PD). Here, hIPSC-derived mDA neurons with PARK2 or PINK1 mutation were shown to recapitulate PD phenotypes, including pathogenic protein accumulation and abnormal neurotransmitter homeostasis. However, in the era of personalized medicine and considering the high degree of variance among PD cases, multiple hIPSC lines must be available to study. Unfortunately, the neural induction capacity of each hPSC lines are not always identical. Additionally, even though several successful cases had been reported in the field of cell therapy, improvement is still necessary for wider clinical application. To answer these issues, we also found that hPSCs can be differentiated into neural cells when co-cultured with human embryonic stem (hES)-derived NPCs, without leaving a trace of undifferentiated cells that may induce tumor formation. We further propose our findings as a new and efficient strategy to generate neurons from hPSCs, that works across hESC and hIPSC lines. Altogether, this study illustrates the promise of hPSC technology for disease modeling and transplantation study.
