Nevertheless, the heterogeneity of tumor cells and tumor environment induced recurrence, metastasis and the resistance of anticancer drugs tumor environment. Thus, this remained the difficult problem to solve. To develop the strategy for cancer treatment, the evaluation platform of in vitro and in vivo as a preclinical model has been improved, however, that is not enough to reproduce the properties of cancer patients. In this study, xenograft models were established to implement tumor microenvironment and heterogeneity in consideration of interaction between tumor cells and peripheral factors. Established xenograft models could increase the correlation of clinical data to improve the cancer treatment strategy. To this purpose, this study was described to three chapters. In Chapter 1, xenograft cancer model using ovariectomized mice mimicking post-menopausal women was used to evaluate the effects of estrogen on tumor cells. In Chapter 2, an improved cancer patient-derived organoid xenograft model was established by co-transplantation of endothelial colony-forming cells (ECFCs) that act on an angiogenesis. In Chapter 3, a metastasis-induced xenograft model by intravenous transplantation of cancer cells was produced, and this model was applied to an evaluation tool of anticancer efficacy. The first, tumor growth was slightly slowed but not significant in a xenograft model derived from gastric cancer cells (SNU-16) using ovariectomized mice. This is consistent with previous study that tumor growth was slowed in male xenograft models rather than in female xenograft model when SNU-16 cells were transplanted into male and female mice. Administration of estrogen in SUN-16-derived xenograft models using ovariectomized mice showed a significant increase in tumor proliferation. To elucidate this mechanism, total RNA sequencing of tumor tissues was analyzed. Increased expression genes only in ovariectomized xenograft model treated with estrogen were examined for protein expression in isolated tumor tissues and survival curve from clinical databases. In this results, metallothionein 3, artemin and JAK2 were found. These genes are suggested to be applied as prognosis biomarkers in female gastric cancer patients. Furthermore, these results suggest that hormone therapy should be cautious in postmenopausal gastric cancer patients. The second, based on the results of Chapter 1, the matched sex of animals with patients were used in establishing xenograft models. The xenograft model transplanted with colon cancer patient-derived organoids (OPDX) was confirmed to maintain the patient's genetic information and to be available as anticancer drug evaluation system by the test of 5-FU and oxaliplatin. Furthermore, OPDX model treated with 5-FU showed hematological toxicity, suggesting that this system could be utilized for the evaluation of chemotherapeutic efficacy and adverse effects. However, it is important to reduce the time required for the evaluation of chemotherapeutic efficacy because the screening result of anticancer drug susceptibility should be informed before chemotherapy in post-surgery patient. Thus, ECFCs was used to shorten the construction time of original OPDX. ECFCs that differentiates into vascular endothelial cells is an important peripheral factor for cancer cell proliferation. The colorectal cancer patients- derived organoid xenograft cancer model with co-implantation of ECFCs significantly promoted tumor formation and proliferation. Furthermore, the chemotherapeutic efficacy was different in OPDX models depending on patients treated with oxaliplatin. This improved OPDX model increased the rate of tumorigenesis and reduced the required time, suggesting this OPDX model could be applied for the screening test of chemotherapeutic efficacy. The third, circulating tumor cells isolated from the primary cancer tissues occurred metastasis via blood circulation system. To mimicking this condition, metastatic xenograft model made by intravenous injection of cancer cells was prepared. The chemotherapeutic efficacy of chrysin-nanoparticles (chrysin-NPs) was evaluated in triple-negative breast cancer (TNBC) MDA-MB-231 cells-derived metastasis model. In the previous study, it is reported that incorporation of insoluble chrysin into nanoparticles was enhanced to bioavailability. Thus, in this study, the result showed that chrysin-NPs significantly suppress the metastasis in metastasis model. Also, chrysin-NPs inhibited the tumor growth in MDA-MB-231-derived xenograft model and induced apoptosis in tumor tissues. Furthermore, chrysin-NPs inhibited NF-kB (p65) expression by GPER activation, suppressing the expression levels of proliferation-related proteins and invasive and metastatic factors (MMPs and TIMP-1). These results suggest that chrysin-NPs could be utilized as an anticancer adjuvant in the primary TNBC and metastasis. In conclusions, the xenograft tumor models constructed from this study improved the shortcomings of original xenograft models, suggesting that these models are useful as the chemotherapeutic efficacy evaluation system and provide valuable information for interpreting clinical data.
