A 균에 대한 항균활성을 측정한 결과 Paper disc method에서 S. aureus, S. epidermidis, P. aeruginosa 균의 생육을 저해하는 것으로 나타났으며 그 중 S. aureus에 대한 생육저해환은 0.5 g/mL농도에서 18.35 ± 1.5 mm로 대조군인 methyl paraben(16.83 ± 1.0 mm)보다 우수한 항균활성을 나타내었음을 확인하였다. MIC 측정을 통하여 와송 에틸아세테이트 분획물의 최소저해농도를 평가한 결과에서는 Candida. A를 제외한 나머지 S. aureus, S. epidermidis, E. coli P. aeruginosa 균주에서 17.5 mg/mL의 최소저해농도를 보였으며 화장품 에멀션 제형에 적용하여 Challenge test를 통해 방부 효능을 평가한 결과 S. aureus에 대하여 와송 에틸아세테이트 분획물을 0.3 % 첨가한 에멀션에서 7일 후 100.0 %균의 생장을 사멸시켜 화장품에 적용 시 우수한 방부효능을 나타내었다. 대체실험동물모델인 zebrafish를 이용한 독성실험결과에서는 모든 농도에서 100 %의 배아 생존율을 보여 응고 또는 부화지연을 관찰할 수 없었으며, UV-B조사 및 LPS에 의해 유도된 ROS생성은 3 μg/mL 농도에서 각각 양성대조군 대비 35.7, 60.8 %의 감소율을 보여 효과적으로 ROS생성을 억제하였음을 확인하였다. 각질형성세포인 HaCaT세포에 대한 와송 에틸아세테이트 분획물의 독성실험 결과에서는 1.0 μg/mL농도 이하에서 100 %이상의 세포 생존율을 나타내었으며, UV-B조사에 의한 세포 보호효과를 알아보기 위해 와송 에틸아세테이트 분획물을 처리하여 관찰한 결과 5.0 μg/mL 농도에서 양성 대조군 대비 48.48 %의 ROS 발현량 감소를 보여 UV-B에 의해 유도된 ROS생성을 우수하게 억제하였음을 확인하였다. 분획물을 함유한 안정성 평가에서는 대조군 대비 분획물의 농도가 증가할수록 pH, 경도 변화에 영향을 미치는 것으로 나타났으나 0, 25, 40℃ 온도별 안정성 평가에서 안정적인 입자거동을 확인 하였다. 이상의 결과로 미루어보아, 추후 와송 에틸아세테이트 분획물의 적절한 농도 설정을 통해 천연 항산화제, 방부제 및 항염 기능성 소재로서 화장품 분야에서 응용 가능성을 확인하였다.

Berger EtOAc fraction extracts to the emulsion. As the result, the total phenolic compounds and flavonoids of Orostachys japonica A. Berger EtOAc fraction extracts were 193.0 ± 1.0 mg GAE/g and 165.7 ± 8.3 mg QE/g. Antioxidant effect of Orostachys japonica A. Berger EtOAc fraction extract at 0.10 mg/mL showed a DPPH radical scavenging rate of 78.54 % and ABTS+ radical scavenging rate of 73.48 % and Ferric reducing ability of 0.39 and exhibited a dose-dependent increase in their antioxidant activity. Berger EtOAc fraction extracts using alternative experimental animal model zebrafish, confirmed a 100 % the survival of the zebrafish embryo was shown that there was no coagulation and no hatching delay at all concentrations.
