Organogels composed of poly(3-hydroxybutyrate) (PHB) and poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV) are fabricated in different compositions using non-solvent-induced phase separation (NIPS). Organogels composed of poly(3-hydroxybutyrate) (PHB) loaded with cellulose nanocrystals (CNC) were fabricated by non-solvent-induced phase separation (NIPS) using chloroform as solvent combined with two different non-solvents, tetrahydrofuran (THF) and 1, 4-dioxane (Diox). Applying sequential solvent exchange with methanol and water, followed by freeze-drying processes to the organogels allowed fabrication of PHB/PHBV nanofibrous hydrogels and foams with high porosity, PHB/CNC composite foams. Uniaxial mechanical behavior of hydrogels and foams was investigated by compressive testing, which indicates that the ductility increased but the modulus and strength decreased with increasing PHBV mass ratio. Hydrophobicity of PHB/PHBV gel networks resulted in unique mechanical and rheological behaviors of hydrogels originated from their weak bonding with water. Under a large compressive deformation, the hydrogels did not fracture, even at a strain of 95%, but contracted uniformly by expelling most of the water from hydrogels. Under a higher amplitude of dynamic shearing, hydrogels showed liquid-like behavior indicating that the shearing of water de-bonded from is dominant. Under a lower amplitude of dynamic shearing, solid-like behavior of typical hydrogels was recovered due to re-bonding of water with hydrophobic network exhibiting the unique thixotropic property of PHB/PHBV hydrogels. The drug loading and release behavior was investigated for PHB/PHBV foams using indomethacin as a hydrophobic model drug. With increasing PHBV content, IDM-loaded hydrogels exhibited more sustained drug release. It is found that the morphology and properties of NIPS-derived PHB/CNC nanocomposite foams greatly depended on the selection of non-solvent. Mechanical strength was much enhanced for NIPS-derived foams when THF was used for non-solvent compared to Diox. The CNC-reinforced PHB foams was successfully achieved only with Diox, whereas nanocomposite foams using THF as non-solvent for NIPS showed no mechanical improvement compared to pure PHB foam. CNC showed much improved solubility in Diox compared to chloroform and THF, which prevent the severe aggregation of CNC during phase separation resulting in the uniform dispersion of CNC in organogels and effective reinforcement of foams after the solvent evaporation. In vitro viability assays were performed using PC12 cell, and CNC-reinforced PHB foams demonstrated better cell adhesion and cell viability than pure PHB foam. Therefore, the nanofibrous PHB/CNC composite foams prepared by NIPS could find potential use as scaffolds for nerve tissue engineering.
