In previous studies, Curcumin was found to be effective anti-oxidant, anti-inflammatory, anti-viral, anti-cancer, cardiovascular disease, obesity and diabetes control, allergies, and degenerative neurological diseases. C. elegans, a useful model for the study of aging and PD, was used for evaluating potential suppression and neuroprotective effects on the signs of aging and PD. After ingesting curcumin with E. coli OP50, their effects of curcumin on lifespan and lipofuscin, expression of superoxide dismutase-3 (SOD-3) and catalase-1,2,3 (CTL-1,2,3) induced in the insulin/insulin like growth factor (IGF)-1 signaling (IIS) pathway were studied, C. elegans model of PD was tested for α-synuclein aggregation inhibition ability, lipid change, dopamine transport ability, locomotion rate, and a simulation of curcumin and α-synuclein binding was conducted. As a result, curcumin prolonged the lifespan of wild-type N2, dat-1p::GFP strain and α-synuclein expressing NL5901, daf-2 and daf-16 mutant respectively. Lipofuschin-related aging was also inhibited by curcumin, so it was confirmed that curcumin was involved in prolonging lifespan. SOD-3 and CTL-1,2,3, that an remove reactive oxygen species (ROS) in IIS pathway, a major cause of PD. Curcumin led to the increased nuclear localization of DAF-16 a key transcription factor that regulates lifespan, anti-oxidant, stress resistance, detoxification, fatty acid/lipid metabolism. Curcumin diet inhibited dopamine neuron degeneration and improved dopamine-related locomotion rate. The molecular docking simulation of α-synuclein with curcumin demonstrate that curcumin has a high affinity near β-hairpin (amino acid 36-55 region) of α-synuclein, suggesting an important role in α-synuclein aggregation. These results demonstrate a possibility of curcumin as anti-aging, neuroprotective effects and treatment in PD.
