DNFB (1-fluoro-2,4-dinitrobenzene)를 이용하여 Balb/c mice의 등 부분에 patching 방법과 painting 방법으로 아토피를 유발한 결과, painting법은 patching 방법에 비하여 임상평가 지수, 기관의 무게변화, 그리고 조직병리학적 평가에 있어서 아토피 피부염 개선을 더 효과적으로 확인할 수 있는 검증법으로 판단되었다. 또한, 시료의 적정 처리 농도의 경우 1% NaCl 처리 농도에서 아토피 피부염증 평가에 있어서 더 효과적이었고, 피부조직의 두께 변화와 호산구 수의 변화에서도 더 효과적이었다. 그리고, 시료의 적정 처리 방법에 있어서 bathing 방법이 painting 방법에 비해 모든 아토피 피부염증 평가 항목에서 더 효과적임을 확인하였다. 이어 소금의 종류(천일염, 일반소금, 죽염) 및 SS1의 아토피 피부염증 개선 효과를 검증하기 위해 SS1을(SS1군) 제외한 각 시료를 1% NaCl 함량으로 조제된 천일염(MRS군), 미네랄이 없는 시약(MDS군), 죽염(MBS군)을 아토피 피부염증 Balb/c mice에 bathing 방법으로 처리하였다. 그 결과, SS1군에서 임상지수의 유의적인 개선 효과를 보였으며 소금과 SS1를 처리한 모든 군에서 비장 및 림프절의 크기가 유의적으로 감소하였다. 또한, 대조군은 표피두께가 두꺼워진 반면에, 소금 및 응용제품(죽염과 SS1)을 처리한 군들은 그 표피두께가 상대적으로 엷음을 확인하였다. 특히, MBS처리군과 SS1처리군은 MDS처리군 보다 표피두께가 유의적으로 엷었다. 호산구 수는 SS1군이 유의적으로 가장 낮았으며, 비만세포 수의 경우 MBS군에서 가장 낮은 수준이었으며 SS1군과 MRS군도 MDS처리군에 비해 유의적으로 낮은 수준이었다. 또한, 혈중 IgE, IFN-γ, 그리고 IL-4의 농도를 조사한 결과, 소금처리군 및 SS1처리군에서 혈중 IgE 농도가 감소하였으며, IFN-γ은 SS1군에서 유의적으로 감소하였으나 혈중 IL-4의 농도는 처리군 간에 유의적인 차이를 보이지 않았다. These salts have been reported to cause less oxidative stress than mineral-deficient salt (MDS). In particular, bamboo salt(BS) has various health-beneficial effects, including anti-oxidative, anti-cancer, anti-inflammatory, and anti-metastatic activities. However, anti-atopic dermatitis (AD) activities of different dietary salts (MDS, MRS, and MBS) and the by product of sea salt(SS1) have not been reported. Therefore, in this study, the effect of MDS, MRS, MBS, and SS1 on AD using an animal model(Balb/c mice) of 2,4- dinitrofluorobenzene (DNFB)-induced AD was investigated. The DNFB-induced AD mice were bathed in the salt solutions containing 1% NaCl and SS1 solution every day for 10 min. The clinical observation scores of DNFB-induced AD mice were decreased by MBS and SS1 treatments. In particular, the SS1 group showed the lowest clinical observation score compared to other groups. All salt and SS1 treatments significantly reduced lymph nodes weights of DNFB-induced AD mice, but there was no significant difference among these groups. The epidermal thickness of the skin lesions was reduced in the salt and SS1 treated groups compared with the AD group. The MBS and SS1 groups were effective in the reduction of skin epidermal thickness induced by DNFB. Comparing to the AD group, the eosinophil number and mast cells were significantly reduced by the SS1 treatment (SS1 group). Additionally, the MRS and MBS treatment significantly reduced the mast cell number in the skin lesion of DNFB-induced AD mice. Serum IgE, IFN-γ, and IL-4 levels were increased by DNFB. The salt and SS1 treatments significantly reduced serum IgE levels increased by DNFB. The SS1 group significantly reduced serum IFN-γ levels as compared with the AD group, but there was no significant difference in serum IL-4 level between the salt and SS1 treatments. The mRNA expressions of IL-5 and IL-13 in the AD skin lesions were up-regulated by DNFB. The SS1 treatment significantly decreased the mRNA expression of IL-5 as compared to the AD and MDS groups. These results indicated that salt and SS1 treatment can diminished skin lesions in DNFB-induced AD mice. Especially, MBS and SS1 treatments were more effective in the AD suppression as compared to MDS. In addition, the present study was to examine whether the salt and SS1 treatment could control the microbiome in the AD skin lesions by using metagenomics analysis. The results showed no significant difference in diversity and uniformity of microbial communities among the salt and SS1 groups. In addition, soy sauce, a representative salt-based seasoning, has recently been reported to reduce oxidative stress. In the present study, the protective effect of processed salt (SMRS) made from MRS and soy sauce on oxidative stress in rats were examined. Sprague–Dawley rats were fed the diets containing various salts (8% NaCl, w/w) for 4 weeks. The SMRS groups showed significantly lower levels of lipid peroxidation (malondialdehyde, MDA) in the plasma and higher total protein carbonyl content (PCOs) in the liver, as compared to the MDS group. Comparing to the MDS group, the SMRS group was significantly higher expression levels of apurinic/apyrimidinic endonuclease 1 (APE1), flap endonuclease 1(FEN1)and proliferating cell nuclear antigen (PCNA) in the liver tissue. Additionally, the expression level of nuclear factor erythroid 2-related factor 2 (Nrf2) was down-regulated by the high salt diet but significantly increased by the SMRS diet. Expression levels of NF-κB p65, IL1-β, and TNF-α as inflammatory mediators in liver tissue were increased by the various salt diet, but the SMRS diet significantly lowered their levels compared to the MDS diet. However, there was no significant difference in the expression levels of TNF-α between the salt-fed groups. Moreover, the level of oxidative stress- and inflammation-related factors between the SMRS and CON groups were not different. These results suggest that SMRS could be consumed as alternative dietary salts that could reduce oxidative stress caused by the general salts.
