This study was conducted to figure out the efficient Agrobacterium- mediated transformation conditions using hypocotyl explants and to develop mutants resistant to sulfonylurea herbicides by inducing mutation in the acetolactate synthase (ALS) gene through Agrobacterium-mediated CRISPR/nCas9 base editing in Brassica napus ‘Westar’. In Agrobacterium- mediated transformation, the preculture period of hypocotyl explants before bacterial inoculation and the type and concentration of selection agents after inoculation were treated differently and the gene introduction efficiencies were compared through GUS histochemical analysis. It was confirmed that the preculture period had a great influence on gene introduction efficiency. In preculture 0 and 3 days, GUS expression rates were 84.8% and 8.4%, respectively. Preculture 0 day treatment was much more effective in gene introduction. In the comparison of the selection agents, phosphinothricin (PPT) and kanamycin (Km), high efficiency in GUS gene expression was obtained in the explants cultured in PPT containing medium. As a result, for the efficient Agrobacterium-mediated transformation of Brassica napus ‘Westar’, the explants should be inoculated without preculture and cultured on selection medium containing PPT. Based on this, CRISPR/nCas9 base editing vector pECO201_CDA1_BnALS19 containing 19 nucleotide sgRNA targeting Pro 173 of Brassica napus ALS gene was introduced into hypocotyl explants through Agrobacterium-mediated transformation and the transformed explants were cultured in a selection medium containing 5.0 mg/L PPT. After the culture and selection process, 5 lines of shoots were generated. PCR analysis using primers specific to each of the bar and nCas9 genes showed that the base editing system was stably integrated into 4 lines of transgenic shoots. The mutation patterns were assayed after Sanger sequencing, and base substitution(s) occurred in common at C6 of the protospacer in all four lines. One line had C-T substitution at the C6 and C7 positions, caused a change in the amino acid sequence from proline to serine (P173F). The base-edited mutants with P173S mutation of ALS are thought to show resistance to sulfonylurea herbicides.
