본 연구는 한국의 충북과 제주도에서 발견 된 야생화 꽃화분과 김치, 콩된장, 동치미 등 한국 발효식품에서 신규 미생물을 분리, 동정하였다. 분리 된 미생물은normal sequencing 분석을 통해 16S rRNA 유전자 염기 서열 분석을 수행하였다. 결과적으로 화장품 소재로 활용 가능한 4종의 야생 효모균주, 4종의 Lactobacillus sp. 및 2종의 Bacillus sp. 균주를 확보하였다. 천연물의 추출방식은 증포(JEUNG-PO) 추출공정을 통해 수득한 안토시아닌배당체를 함유한 블랙커런트 추출물을 기질로 하여 효모균 Saccharomycopsis fibuligera J2K-107과 유산균 Lactobacillus brevis J2K-55 균주의 이중발효를 통해 저분자 안토시아닌(delphinidin, cyanidin)과 젖산(lactic acid)을 함유한 블랙커런트 이중발효물을 생산하였고, γ-aminobutyric acid (GABA)을 생산하기 위해 글루탐산이 함유된 다시마추출물에 유산균 L. brevis J2K-55 균주의 발효배양을 통해서는 가바(GABA)를 생산하였다. 블랙커런트 이중발효물과 다시마추출발효물은 DPPH assay와 ABTS assay를 통해 농도의존적으로 항산화 활성을 나타내었으며, 두 시료 모두 100 µg/mL 농도에서 세포독성을 나타내지 않았다. LPS (lipopolysaccharide)로 유도된 RAW264.7 세포에 블랙커런트 이중발효물과 다시마추출발효물을 각각 처리하였을 때, 농도의존적으로 NO 생성이 감소하는 것을 확인하였고, 염증유도인자인 iNOS와 COX-2 단백질 발현을 Western blot으로 확인한 결과, 블랙커런트 이중발효물은 iNOS와 COX-2 단백질 발현을 저해하였으나, 다시마추출발효물의 경우 iNOS의 발현은 감소시켰지만 COX-2는 미미한 저해 활성을 확인하였다. 두 시료의 항주름 효과에서는 UVB에 조사된 피부 섬유아세포(CCD-986SK)에 각각의 시료를 처리한 결과, MMP-1 단백질 발현을 저해하는 것으로 나타났다. 또한, 두 시료를 각각 3% 적용한 화장품 제형을 인체적용시험을 수행한 결과, 두 제품 모두 피부 이상반응이 없었으며 피부결 개선, 탄력증진, 수분 및 피부 진정효과를 확인할 수 있었다.

In this study, the aqueous extracts of blackcurrant, containing high amount of anthocyanin glycosides through JEUNG-PO extraction, were co-fermented with Saccharomycopsis fibuligera J2K-107 and Lactobacillus brevis J2K-55 to produce low molecular anthocyanins (cyanidin and delphinidin) and lactic acid, respectively. In addition, the kelp extract with glutamic acid was cultured with L. brevis J2K-55 to produce γ-aminobutyric acid (GABA). Efficacy evaluation of fermented blackcurrant extract (FBE) and fermented kelp extract (FKE) were then studied. FBE and FKE had antioxidant activities and showed no cytotoxicity at a concentration of 100 µg/mL. The respective treatment of FBE and FKE in LPS-induced RAW264.7 cells resulted dose-dependent decreases of the NO production. The expression levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein in LPS-induced RAW264.7 cells were reduced by FBE treatment, according to Western blot analysis. FKE also decreased the expression of iNOS, however, it showed negligible activity on COX-2 protein expression inhibition. The respective treatment of FBE and FKE in UVB-irradiated CCD-986SK cells resulted a dose-dependent decrease of the expression level of matrix metalloproteinase-1 (MMP-1) protein, according to western blot analysis. The safety and efficacy of cosmetic compositions containing FBE and FKE were then investigated by clinical tests. According to the instrumental analysis results, cosmetic products containing FBE and FKE showed more positive skin effects. Especially, FBE containing product resulted on improved skin roughness, desquamation and elasticity compared to placebo. And, FKE containing product also showed higher moisturizing and soothing effect than placebo.
