2. 배추 검은썩음병에 대한 효과적인 병리검정법으로써 배추 종자를 파종하고 온실(25±5℃)에서 10일간 재배하여 2엽이 전개한 배추 유묘에, 병원균 밀도가 1.0×108 cfu/ml인 세균 현탁액을 배추 잎의 앞 뒷면에 골고루 분무하여 접종하고 30℃ 습실상에서 48시간 습실 처리한 후에, 항온항습실(25℃, 상대습도 80%)로 이동하여 하루에 12시간씩 광을 조사하면서 재배하는 것을 제안하고자 한다.
3. 배추 검은무늬병균에 대한 효과적인 병리검정법으로써 배추 종자를 파종하고 온실(25±5℃)에서 16일간 재배하여 2엽이 완전전개한 배추 유묘에 Alternaria brassicicola 포자 현탁액(3.0×105 spores/ml)을 배추 잎의 앞 뒷면에 골고루 분무 접종을 하고 25℃ 습실상에서 24시간 습실 처리한 후에, 항온항습실(25℃, 상대습도 80%)로 이동하여 하루에 12시간씩 광을 조사하면서 재배하는 것을 제안하고자 한다.
4. 실험한 무시들음병균 균주들의 병원력은 HN와 JHW 균주가 가장 강하였고, 다음은 NW1, 147A, 57A, 59A, 60A 순이었다. 그리고 이들 균주 중 중간 정도의 병원력을 나타내는 KR1 균주를 사용하여 시판 무 품종 60개의 시들음병 발생을 조사하였다. 이들 결과로부터 무의 시들음병균에 대한 저항성 특성 실험을 위하여 저항성을 나타내는 25개 품종을 선발하였다. KR1 균주를 제외한 7개 균주에 의한 선발한 25개 품종의 시들음병 발생을 조사한 결과, 무시들음병균 60A, 59A, 57A, 147A, NW1, HN, JHW에 의한 평균 발병도는 각각 0.5, 0.7, 1.2, 2.3, 2.8, 3.7, 3.9였으며, 저항성 품종의 수는 각각 21개, 20개, 13개, 7개, 2개, 1개 및 1개이었다. 그리고 각 품종에서의 시들음병 발생은 균주들의 병원력에 비례하였다.
5. 실험한 무무름병균 균주들의 병원력은 ATCC 12312 균주가 가장 강하였고, 다음은 ECC 301365, KACC 10421, KACC 10225, ATCC 15713, LY34 순이었다. 그리고 이들 균주 중 중간 정도의 병원력을 나타내는 KACC 10421 균주를 사용하여 시판 무 품종 60개의 무름병 발생을 조사하였다. 이로부터 무름병 저항성 특성 실험을 위하여, 무름병 발생이 적었던 24개 품종을 선발하였다. 무름병균 6개 균주에 의한 선발한 24개 품종의 무름병 발생을 조사한 결과, 무무름병균 LY34, ATCC 15713, KACC 10225, KACC 10421, ECC 301365, ATCC 12312에 의한 평균 발병도는 각각 8.3, 33.3, 43.6, 66.7, 90.5, 109.3이었으며, 각각에 대하여 저항성을 나타낸 품종의 수는 20개, 18개, 15개, 5개, 2개, 2개로 저항성 품종의 균주의 병원력에 반비례하여 증가하였다.
6. 실험한 배추검은썩음병균 균주들의 병원력은 KACC 19135 균주가 가장 강하였고, 다음은 KACC 19134, 19527, 17966, 19132, 19133, 10377, 19136 순이었다. 그리고 이들 균주 중 KACC 10377 균주를 사용하여 시판 배추 품종 88개 품종에 대해 검은썩음병 발생을 조사하였다. 이로부터 검은썩음병 저항성 특성 실험을 위하여 저항성을 나타내는 24개 품종을 선발하였다. 8개 검은썩음병균 균주들에 의한 선발한 23개 품종의 검은썩음병 발생을 조사한 결과, KACC 19136, 10377, 19133, 19132, 17966, 19527, 19134, 19135에 의한 평균 병반면적률은 각각 12.4, 19.9, 44.4, 51.2, 88.1, 88.0, 96.6, 97.7%였으며, 저항성 품종의 수는 각각 21개, 17개, 5개, 5개, 1개, 1개 였으며, KACC 19134와 KACC 19135 균주들에 대해서는 저항성을 나타낸 품종은 없었다. 그리고 각 품종에서의 검은썩음병 발생은 균주들의 병원력에 비례하였다.
7. 실험한 배추검은무늬병균 균주들의 병원력은 KACC 44877 균주가 가장 강하였고, 다음은 KACC 40857, 44415, 40036, 40034, 43923, 44878 순이었다. 그리고 이들 균주 중 중간 정도의 병원력을 나타내는 KACC 40036 균주를 사용하여 시판 배추 품종 86개 품종에 대해 검은무늬병 발생을 조사하였다. 이로부터 검은무늬병 저항성 특성 실험을 위하여 병 발생이 적었던 24개 품종을 선발하였다. 그리고 8개 시들음병균 균주들에 의한 선발한 24개 품종의 시들음병 발생을 조사한 결과, KACC 44878, 43923, 40034, 40036, 44415, 40857, 44877에 의한 평균 병반면적률은 각각 1.4, 9.2, 26.2, 53.2, 83.1, 90.2, 93.7%였으며, 저항성 품종의 수는 KACC 44878 균주에 대해서는 22개, KACC 43923 균주에 대해서는 9개였으며, KACC 140034, 40036, 44415, 40857, 44877 균주들에 대해서는 저항성을 나타낸 품종은 없었다. 그리고 각 품종에서의 검은무늬병 발생은 균주들의 병원력에 비례하였다.
8. 무무름병균, 배추검은썩음병균, 배추검은무늬병균, 무시들음병균에 대한 기주 식물의 저항성 양상은 질적 저항성에서 나타나는 품종과 균주 간의 특이적 관계는 존재하지 않았다. 그리고 모든 품종은 실험한 균주들의 병원력에 비례하여 병이 발생하는 양적 저항성 특성을 나타냈다. 따라서 이러한 양적 저항성을 나타내는 무 시들음병, 무 무름병, 배추 검은무늬병, 배추 검은썩음병에 대한 저항성 품종들은 이들의 저항성 정도에 차이가 존재하므로, 저항성 정도가 약한 품종은 포장에서 병이 심하게 발생할 경우 저항성 품종도 감수성 반응을 나타낼 수 있을 것이다.
9. 이러한 양적 저항성 특성을 나타내는 유전자원을 이용한 저항성 품종 개발 시, 저항성 계통을 만들기 위한 교배 집단의 병 저항성 개체 선발은 세대가 진전됨에 따라 저항성이 약해지는 점을 고려하여 초기 세대 선발에서는 강한 병원력을 가지는 균주를 사용하여 병리검정을 실시해야 하며, 후기 세대 선발이나 F1 품종의 저항성 정도를 확인하기 위해서는 중간 정도의 병원력을 나타내는 균주를 사용하여 병 저항성 검정을 실험하는 것이 효과적일 것이다.

This study was conducted to establish an efficient screening method for the development of resistant cruciferous crops against plant pathogens and to investigate the resistance of various cultivars of cruciferous crops. To develop the screening method for the resistant cultivars of radish (Raphanus sativus) to bacterial soft rot caused by Pectobacterium carotovorum subsp. carotovorum, resistance degrees of 60 commercial radish cultivars to the P. carotovorum subsp. carotovorum, KACC 10421 isolate were investigated. For further study, six radish cultivars (Awooriwoldong, YR Championyeolmu, Jeonmuhumu, Bitgoeunyeolmu, Sunbongaltari, and Housecheongok) showing different level of resistance to the bacterium were selected. Disease severity of bacterial soft rot on various cultivars was tested with different conditions, such as incubation temperature/period after inoculation, seedling stage of radish, and inoculum concentration. On the basis of the results, we suggest that an efficient screening method for the resistant radish to aginst P. carotovorum subsp. carotovorum: 1) inoculate 20−day−old seedlings with a bacterial suspension of P. carotovorum subsp. carotovorum (8×105 cfu/ml), 2) to incubate the inoculated plants in a growth room at 25℃ (80% RH) with 12 h light/dark cycle, and 3) to investigate disease severity 4 days after inoculation. For the development of screening methods for resistant Chinese cabbage to black rot caused by Xanthomonas campestris pv. campestris, the resistance degree of 88 commercial cultivars of Chinese cabbage against X. campestris pv. campestris KACC 10377 was examined. Sequencially, we selected three resistant cultivars (Mansumugang, Woldongcheonha, and CR Anshim) and three susceptible cultivars (Bulam3ho, Chunkwang, and CR Nongshim). According to the growth stages of seedlings, inoculation density, and incubation temperature/period after inoculation, the development of black rot on the six cultivars was investigated. As an efficient method for the selection of resistant Chinese cabbage seedlings to X. campestris pv. campestris, we suggest the following method: 1) to inoculates cell suspension of X. campestris pv. campestris (1.0×108 cfu/ml) on 10−day−old Chinese cabbage seedlings using the spray method, 2) to incubate the inoculated plants in a dew chamber at 30°C for 48 h and then transfer to a growth chamber at 25°C (80% RH) under a 12 h light/dark cycle, 3) to measure disease severity of the black rot six days after inoculation. Finally, we also established an efficient screening method for Chinse cabbage cultivars that are resistant to black spot, which is caused by Alternaria brassicicola. When the resistance of 86 commercial Chinese cabbage cultivars was investigated with A. brassicicola KACC 40036, we found that there were no highly resistant Chinese cabbage cultivars; however, some cultivars, such as ‘cheonhajangkun’ and ‘Mansumukang’, showed weak resistance to A. brassicicola. For further study, we selected eight Chinese cabbage cultivars that showed different disease responses to A. brassicicola KACC 40036. According to the growth stage of the Chinese cabbage seedlings, inoculum concentration, and incubation temperature/period after inoculation, development of black spot on eight cultivars was investigated. The results showed that younger seedlings were more sensitive to A. brassicicola than older seedlings, and the disease severity was dependent on the concentration of inoculum. The disease severity of plants incubated in humidity chamber at 25°C was greater than that of plants grown at 20°C or 30°C. Taken together, we suggest the following method for screening for Chinses cabbage plants that are resistant to A. brassicicola: 1) to inoculate 16−day−old seedlings with an A. brassicicola spore suspension (3.0×105 spores/ml) using the spray method, 2) to incubate the inoculated plants in a humidity chamber at 25°C for 24 h and then transfer the plants to a growth chamber at 25°C with 80% relative humidity under a 12 h light/dark cycle, and 3) to assess the disease severity of the plants two days after inoculation. To determine the resistance of radish cultivars against Fusarium oxysporum f. sp. raphani, we examined the virulence of 8 F. oxysporum f. sp. raphani isolates in a susceptible radish cultivar ‘Backchun’. Area under disease progress curve (AUDPC) of Fusarium wilt on the cultivar caused by F. oxysporum f. sp. raphani 60A, 59A, 57A, 147A, KR1, NW1, HN, and JHW isolates was 6.7, 14, 16, 27, 27, 30, 35, and 35, respectively. Thus, virulence of F. oxysporum f. sp. raphani HN and JHW was the strongest, followed by F. oxysporum f. sp. raphani NW1, KR1, 147A, 57A, 59A, and 60A isolates. With a medium virulent isolate F. oxysporum f. sp. raphani KR1, we investigated the resistance of 60 commercial radish cultivars, and consequently 25 radish cultivars were selected for the investigation of race differentiation. When 25 cultivars were inoculated by each of seven F. oxysporum f. sp. raphaniisolates except for KR1, the means of disease index by F. oxysporum f. sp. raphani 60A, 59A, 57A, 147A, NW1, HN, and JHW isolates were 0.5, 0.7, 1.2, 2.3, 2.8, 3.7 and 3.9, respectively. The number of resistant radish cultivars to F. oxysporum f. sp. raphani 60A, 59A, 57A, 147A, NW1, HN, and JHW isolates were 21, 20, 13, 7, 2, 1, and 1, respectively. Consequently, I found that the development of Fusarium wilt in each cultivar was positively correlated with the virulence of F. oxysporum f. sp. raphani isolates, suggesting that. Resistance of radish to F. oxysporum f. sp. raphani isolates is likely affected by the virulence, but not resulted from race differentiation. In terms of resitance of cruciferous crops, I determined the resistance of radish and Chinese cabbage cultivars by six P. carotovorum subsp. carotovorum isolates, eight X. campestris pv. campestris isolates, and seven A. brassicicola isolates. AUDPC of bacterial soft rot on the susceptible cultivars caused by P. carotovorum subsp. carotovorum LY34, ATCC15713, KACC 10225, KACC 10421, ECC301365 and ATCC12312 was 63.0, 72.0, 87.5, 107.7, 224.8 and 233.1, respectively. among the all tested P. carotovorum subsp. carotovorum isolates, ATCC 12312 showed the strongest virulence, followed by ECC301365, KACC 10421, KACC 10225, ATCC15713 and LY34 isolates. When the cultivars were inoculated by six P. carotovorum subsp. carotovorum isolates, the mean of disease index by LY34, ATCC15713, KACC 10225, KACC 10421, ECC301365, and ATCC12312 isolates was 0.3, 0.8, 1.1, 2.0, 2.2, and 2.3, respectively. The number of resistant radish cultivars to LY34, ATCC15713, KACC 10225, KACC 10421, ECC301365, and ATCC12312 were 20, 18, 15, 5, 2, and 2, respectively. Infected leaf area (%) of black rot on the susceptible Chinese cabbage cultivar caused by X. campestris pv. campestris KACC 19136, 10377, 19133, 17966, 19134, 19132, 19527, and 19135 isoates were 16.0, 16.5, 21.5, 23.5, 76.5, 97.0, 97.0, and 100.0, respectively. When the cultivars were inoculated with eight X. campestris pv. campestris isolates, the mean of disease severity (%) by KACC 19136, 10377, 19133, 19132, 17966, 19527, 19134, and 19135 isolates were 12.4, 19.9, 44.4, 51.2, 88.0, 88.1, 96.6, and 97.4, respectively. The number of resistant Chinese cabbage cultivars to KACC 19136, 10377, 19133, 19132, 17966, and 19527 isolates were 21, 17, 5, 5, 1, and 1, respectivly. When the cultivars were inoculated with the seven A. brassicicola isolates, the means of disease severity (%) of the cultivars by A. brassicicola KACC 44878, 43923, 40034, 40036, 44415, 40857 and 44877 isolates were 1.4, 9.2, 26.2, 53.2, 83.1, 90.2, and 93.7, respectively. The number of resistant Chinese cabbage cultivars to A. brassicicola KACC 44878 and KACC 43923 were 22 and 9, respectively. Taken together, we found that the disease development of Fusarium wilt, bacterial soft rot, black rot, and black spot on tested cultivars was positively correlated with the virulence of each pathogen, suggesting that the resistance of radish and Chinese cabbage are likely affected by the virulence of pathogens, but not resulted from race differentiation.
This study was conducted to establish an efficient screening method for the development of resistant cruciferous crops against plant pathogens and to investigate the resistance of various cultivars of cruciferous crops. To develop the screening method for the resistant cultivars of radish (Raphanus sativus) to bacterial soft rot caused by Pectobacterium carotovorum subsp. carotovorum, resistance degrees of 60 commercial radish cultivars to the P. carotovorum subsp. carotovorum, KACC 10421 isolate were investigated. For further study, six radish cultivars (Awooriwoldong, YR Championyeolmu, Jeonmuhumu, Bitgoeunyeolmu, Sunbongaltari, and Housecheongok) showing different level of resistance to the bacterium were selected. Disease severity of bacterial soft rot on various cultivars was tested with different conditions, such as incubation temperature/period after inoculation, seedling stage of radish, and inoculum concentration. On the basis of the results, we suggest that an efficient screening method for the resistant radish to aginst P. carotovorum subsp. carotovorum: 1) inoculate 20−day−old seedlings with a bacterial suspension of P. carotovorum subsp. carotovorum (8×105 cfu/ml), 2) to incubate the inoculated plants in a growth room at 25℃ (80% RH) with 12 h light/dark cycle, and 3) to investigate disease severity 4 days after inoculation. For the development of screening methods for resistant Chinese cabbage to black rot caused by Xanthomonas campestris pv. campestris, the resistance degree of 88 commercial cultivars of Chinese cabbage against X. campestris pv. campestris KACC 10377 was examined. Sequencially, we selected three resistant cultivars (Mansumugang, Woldongcheonha, and CR Anshim) and three susceptible cultivars (Bulam3ho, Chunkwang, and CR Nongshim). According to the growth stages of seedlings, inoculation density, and incubation temperature/period after inoculation, the development of black rot on the six cultivars was investigated. As an efficient method for the selection of resistant Chinese cabbage seedlings to X. campestris pv. campestris, we suggest the following method: 1) to inoculates cell suspension of X. campestris pv. campestris (1.0×108 cfu/ml) on 10−day−old Chinese cabbage seedlings using the spray method, 2) to incubate the inoculated plants in a dew chamber at 30°C for 48 h and then transfer to a growth chamber at 25°C (80% RH) under a 12 h light/dark cycle, 3) to measure disease severity of the black rot six days after inoculation. Finally, we also established an efficient screening method for Chinse cabbage cultivars that are resistant to black spot, which is caused by Alternaria brassicicola. When the resistance of 86 commercial Chinese cabbage cultivars was investigated with A. brassicicola KACC 40036, we found that there were no highly resistant Chinese cabbage cultivars; however, some cultivars, such as ‘cheonhajangkun’ and ‘Mansumukang’, showed weak resistance to A. brassicicola. For further study, we selected eight Chinese cabbage cultivars that showed different disease responses to A. brassicicola KACC 40036. According to the growth stage of the Chinese cabbage seedlings, inoculum concentration, and incubation temperature/period after inoculation, development of black spot on eight cultivars was investigated. The results showed that younger seedlings were more sensitive to A. brassicicola than older seedlings, and the disease severity was dependent on the concentration of inoculum. The disease severity of plants incubated in humidity chamber at 25°C was greater than that of plants grown at 20°C or 30°C. Taken together, we suggest the following method for screening for Chinses cabbage plants that are resistant to A. brassicicola: 1) to inoculate 16−day−old seedlings with an A. brassicicola spore suspension (3.0×105 spores/ml) using the spray method, 2) to incubate the inoculated plants in a humidity chamber at 25°C for 24 h and then transfer the plants to a growth chamber at 25°C with 80% relative humidity under a 12 h light/dark cycle, and 3) to assess the disease severity of the plants two days after inoculation. To determine the resistance of radish cultivars against Fusarium oxysporum f. sp. raphani, we examined the virulence of 8 F. oxysporum f. sp. raphani isolates in a susceptible radish cultivar ‘Backchun’. Area under disease progress curve (AUDPC) of Fusarium wilt on the cultivar caused by F. oxysporum f. sp. raphani 60A, 59A, 57A, 147A, KR1, NW1, HN, and JHW isolates was 6.7, 14, 16, 27, 27, 30, 35, and 35, respectively. Thus, virulence of F. oxysporum f. sp. raphani HN and JHW was the strongest, followed by F. oxysporum f. sp. raphani NW1, KR1, 147A, 57A, 59A, and 60A isolates. With a medium virulent isolate F. oxysporum f. sp. raphani KR1, we investigated the resistance of 60 commercial radish cultivars, and consequently 25 radish cultivars were selected for the investigation of race differentiation. When 25 cultivars were inoculated by each of seven F. oxysporum f. sp. raphaniisolates except for KR1, the means of disease index by F. oxysporum f. sp. raphani 60A, 59A, 57A, 147A, NW1, HN, and JHW isolates were 0.5, 0.7, 1.2, 2.3, 2.8, 3.7 and 3.9, respectively. When the cultivars were inoculated by six P. carotovorum subsp. carotovorum isolates, the mean of disease index by LY34, ATCC15713, KACC 10225, KACC 10421, ECC301365, and ATCC12312 isolates was 0.3, 0.8, 1.1, 2.0, 2.2, and 2.3, respectively.
