Ginsenosides 처리 후 백혈병세포의 세포사멸을 annexin-V 과 propidium iodide를 이용하여 유세포분석기로 분석하였고, propidium iodide dye과 R nase 사용한 유세포분석을 사용하여 세포주기 분포를 정량 하였다. 세포 사멸 관련 분자 경로를 찾기 위해 5 개의 항체 (TNF-α, caspase 9/3, poly (ADP-ribose) polymerase, c-myc)을 사용하여 immunoblots 을 수행하였다. 말초 혈액 유래 조혈 줄기 세포 (PBSC)를 이용하여 정상적인 혈액 세포 구획에서의 세포 독성의 유무를 확인하였다.
결과: Rh2 및 compound K는 정상인 유래 말초혈액 유래 조혈모세포에는 세포 독성이 없음을 확인하였다. 그러나 K562, THP-1, Molt-4 사람 백혈병 세포에 대해 선택적 억제 효과가 있음을 입증하였다. MTT 분석에서 Rh2 및 compound K (20 uM ~ 50 uM)로 처리 (4, 8, 12 및 24 시간) 후 백혈병 세포주의 생존율이 시간 및 농도 의존적으로 감소됨을 확인하였다. Rh2 및 compound K의 IC50 값은 백혈병 세포의 유형에 따라 다르게 확인되었다. K562, THP-1, Molt4 세포에서 Rh2 와 compound K 의 IC50 값은 50 uM이었다. Rh2 와 compound K가 세포주기에서 Pro G0 정지를 유도함을 확인하였다. Whereas a significant progress has been made in the molecular pathobiology, the prospect for elderly patients has remained dismal, with median survival times of only several months. In this context, safe and alternative anticancer drugs are urgently needed for aged AML patients. Ginsenosides and their derivatives have been reported to show anti-oxidant, anti-inflammatory, anti-obesity, angiogenic, and anticancer activities. However, their anti-leukemic properties have not been fully explored. Therefore, This study aimed to identify ginsenosides having anti-leukemic effect and their molecular mechanism. Methods: This study screened dozens of ginsenoside species for searching cytotoxic activity on several human leukemia cell lines, such as KG-1, K562, Molt-4, and THP-1 using MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay, which is a colorimetric assay for assessing cell metabolic activity. Apoptosis was determined using the flow cytometric assay with Annexin-V and propidium iodide dye. Cell cycle distribution was quantified via the flow cytometric analysis with propidium iodide dye and Rnase. For the investigation of apoptosis- related molecular pathways, immunoblots were performed with 5 antibodies (TNF-α, caspase 9/3, poly (ADP-ribose) polymerase, c-myc). Peripheral blood-derived hematopoietic stem cell (PBSC) was used for the cytotoxicity of ginsenosides on normal blood cell compartment. Results: The results demonstrated that Rh2 and compound K have a selective inhibitory effect on the K562, THP-1, Molt-4 human leukemia cells without serious cytotoxic effects on primary healthy PBSCs. The MTT assay showed that the viability of the leukemia cell lines after the treatment (4, 8, 12 and 24h) with Rh2 and compound K (20 uM to 50 uM) was reduced in a time- and dose-dependent manner. The IC50 value of ginsenoside Rh2 and compound K varied according to the type of leukemia cells. Representatively, the IC50 value of compound K and ginsenoside Rh2 in the K562, THP-1, Molt4 cells was 50 uM. Expression of cell cycle distribution demonstrated that Rh2 and compound K induced pro G0 Phase arrest. Immunoblot experiments for investigating apoptosis signaling pathway revealed that the killing effect of Rh2 and compound K was induced via TNF-α – mediated extrinsic and common pathway: the expression of c-myc was decreased; however, those of cleaved caspase 3, cleaved poly (ADP-ribose) polymerase, and TNF-α increased.
Conclusion: This study identified compound K and Rh2 ginsenoside as a novel natural anti-leukemic agents, especially for AML cell lines. These natural compounds might be effectively used with safe and alternative antileukemic drugs for the treatment of elderly AML patients.
