The purpose of this study is to find out the physicochemical properties of the formulation by preparing niosomes, which are nonionic surfactant vesicles, and to evaluate the permeability and antioxidant through cell experiments. Rosmarinic acid, a poorly soluble antioxidant, is made into a niosomal formulation to increase solubility and permeability, and to evaluate the possibility of ocular and oral administration. The proniosomal method, which is known to be the simplest of the niosomes, is applied with a slight modification, In this study, including drug molecules (rosmarinic acid), surfactants, DC-Chol, DCP, and Maltodextrin were used as materials for niosome preparations. By comparing the physicochemical properties of formulation with compositions of the materials in various ratios, the optimal drug delivery system was established. The niosomal particles showed a size range of 150-400 nm on average and showed a positive zeta potential charge. The physicochemical properties of niosomes were measured by DSC, XRD, FT-IR, SEM, and TEM, and the permeability and antioxidant efficacy of drugs were evaluated by cell permeation and antioxidant experiments with ocular epithelial cells. In results, the nanoformulation of niosomes showed improved cell permeability and antioxidant efficacy in cell models by efficiently delivering the rosmarinic acid into cells. In conclusion, this study proved that the niosomal formulation could be an efficient drug vehicle system for oral or ocular delivery of poorly water soluble drugs. Ultimately, our formulation with rosmarinic acid could be useful to prevent or treat ocular diseases related with tissue damages by reactive oxygen species.
