본 연구는 세포 재생능력을 갖는 Centella asiatica (병풀) 잎 추출물 및 병풀에 함유된 단일물질인 아시아티코사이드(asiaticoside)가 human dermal fibroblast (HDF) 세포에 미치는 영향을 조사하였다. C. asiatica 추출물, 에틸아세티이트 분획물 및 아시아티코사이드가 H2O2에 의해 유도된 산화 스트레스에서 HDF 세포의 항산화 효과 및 미토콘드리아 활성조절, TNF-α 및 IL-6 수준을 조절하고, 세포주기 재활성화 및 웨스턴블롯에 의한 단백질 수준 변화를 확인하고자 하였다. C. asiatica의 에틸 아세테이트 분획 및 아시아티코사이드는 H2O2유도 산화 스트레스에 의해 야기된 G1 세포주기 정지 및 apoptosis 효과를 약화시켰고, C. asiatica의 에틸 아세테이트 분획 및 아시아티코사이드로 처리시 H2O2 단독처리군 대비 낮은 수준의 TNF-α 및 IL-6을 분비하였다. 아시아티코사이드의 항산화 효과는 세포주기 및 염증성 사이토카인 수준에서 조절된 바와 같이 항산화 효과를 높이고, 미토콘드리아 막 전위(JC-1 염색을 통해 관찰됨)를 증가시켰으며, 정상적인 미토콘드리아 형태로 변화시켰다. 스트레스 유도 후 미토콘드리아 산소 소비율이 증가하였지만, C. asiatica, 에틸 아세테이트 분획 및 아시아티코사이드로 처리된 세포에서는 산소 소비율이 정상으로 회복되었고, TLR4-MyD88-TRAF6-p65 경로가 단백질 수준에서 다시 감소하였다. 이러한 결과는 C. asiatica, 에틸 아세테이트 분획 및 아시아티코사이드가 항산화 및 항염증 효과를 가질 뿐 아니라 HDF 세포의 미토콘드리아 활성을 조절할 수 있음을 시사한다. 따라서 향후 병풀을 이용한 HDF의 항산화 및 미토콘드리아 조절에 대한 연구의 기초 자료로 항산화를 조절하는 기전에 대한 추가연구 시 정보를 제시하고자하며, 병풀을 이용한 응용기술의 개발과 타겟 범위를 확장시켜 보다 넓은 화장품 시장진출이 가능할 것으로 사료된다.

The antioxidant effect of asiaticoside was higher than that of C. asiatica extract and its ethyl acetate fraction. Moreover, treatment with C. asiatica extract, its ethyl acetate fraction, and asiaticoside increased the mitochondrial membrane potential and returned normal mitochondrial morphology. Following stress- induction, the mitochondrial oxygen consumption rate increased, and the TLR4-MyD88-TRAF6-p65 pathway re-decreased in cells treated with C. asiatica extract, its ethyl acetate fraction, and asiaticoside. These findings suggest that C. asiatica extract, its ethyl acetate fraction, and asiaticoside have antioxidant and anti- inflammatory effects as well as control the mitochondrial activities of HDF cells.
