This study was conducted to evaluate the potential as a general foods and health functional foods material by comparing the functional characteristics and immunological functions of dead cells treated with Lactic acid bacteria (LAB). For the development of dead cells, one Lactiplantibacillus and four Lacticaseibacillus species were selected from kimchi and heat-treated at 60℃ and 100℃ for each hours. As a result of the heat treatment, it was confirmed that the bacteria were killed under the conditions of 60℃ for 30 minutes and 100℃ for 5 minutes or more. Lipoteichoic acid (LTA) present in lysate during heat-treatment confirmed that the LTA expression rate increased as the heat treatment time increased. Also, the protein content was higher at 60℃ than at 100℃, where the LTA expression rate was high through protein content measurement. This means that the protein content decreases due to degradation of protein during heat treatment at 100℃, so the final heat treatment condition was set to 60℃ for 60 minutes. As a result of the antioxidant experiment under the heat treatment conditions, Lacticaseibacillus paracasei DK121 and Lacticaseibacillus casei DK207 showed superior antioxidant activity. In the in vitro cell experiment, neither DK121 nor DK207 was cytotoxic, and as the concentration increased, nitric oxide production was significantly inhibited. In addition, all three inflammatory cytokines (TNF-α, IL-1α and IL-6) were confirmed to have significantly lower than lipopolysaccharide (+) group, and this result shows the possibility of reducing the inflammatory response in dead cells. Based on this study, DK121 and DK207, which showed remarkable activity, were selected and treated with commercialized enzymes (Amylase, pepsin and oxgall) to indirectly confirm whether dead cells in the human body were decomposed through SDS-PAGE measurement. As a result, it was confirmed that there is no decomposition of dead cells under all conditions, and 5 minutes of mastication during human digestion, 30 minutes of passing through the stomach and 6 hours of intestinal passage were set as enzyme treatment times. In the antioxidant experiment of the enzymatically decomposed dead cell (EDDC), the EDDC showed lower or similar activity than the dead cell. All of the EDDCs showed non-cytotoxicity through in vitro cell experiments, and as the concentration increased, nitric oxide production was significantly inhibited. In addition, all three inflammatory cytokines (TNF-α, IL-1α and IL-6) were confirmed to have significantly lower than lipopolysaccharide (+) group, and this result shows the possibility of reducing the inflammatory response in EDDC.
