Low energy, minimal generation of inhibitor compounds, and improved process speed during the bio conversion process from lignocellulosic biomass are the main challenges in the biofuel production process. Usually, pretreatment and enzyme production steps contribute majorly ( approximately 50-70%) in the overall production cost of bioconversion process. In the present study, magnetic Cerium-doped ironoxide (CeFeO) nanomaterial possessing exceptional laccase enzyme mimicking properties, have been exploited for the delignification of raw corn cob substrate. Also, cellulase and hemicellulase enzymes required for biomass hydrolysis have been produced in-house using cheaper carbon substrates. Hence, the inclusion of nanomaterials for delignification (as pretreatment) and in-house enzyme preparation would help in reducing the process cost at some extent. Here, we developed a strategy for the conversion of corncob biomass into simple sugars in one step by addition of CeFeO and cellulase-hemicellulase enzymes. The simultaneous pretreatment and saccharification (SPS) process exhibited generation of cellulose-hemicellulose rich fraction after degradation of lignin by laccase-mimicking CeFeO which is simultaneously hydrolyzed into fermentable sugars due to action of cellulase and hemicellulase enzymes in the broth. Predominantly, SPS experiments were performed under mild conditions, i.e. at 55°C generating fermentable sugars without formation of any inhibitor compounds. The maximum 18.76 g/L, 23.45 g/L, and 43.12 g/L concentrations of glucose, xylose and reducing sugar were obtained respectively, after 24 hours of SPS reaction at 55°C from 200 g/L corncob. These results indirectly indicate that there is no inhibitory effect of enzymes on the action of CeFeO nanoparticles during lignin degradation. In addition, since nanoparticles have magnetic properties, they can be easily removed using a bar magnet after simultaneous pretreatment and saccharification and can be reused for several cycles. After recovery of nanoparticles about 85% and 70% efficiency were maintained during second and third recycling, respectively. Therefore, the fermentation process can be carried out almost immediately after hydrolysis, greatly shortening the overall process time. We succeeded in converting the hydrolyzate containing 24.5 g/L of glucose and 22.7 g/L of xylose into 22.2 g/L of ethanol by Saccharomyces cerevisiae yeast strain in 12 hours. During ethanol fermentation, the yeast strain showed the reduction in carbon catabolite repression by utilizing glucose and xylose almost simultaneously which has shortened the fermentation and improved productivity. In this study, we tried to develop zero waste process by utilizing each component in one step through magnetic nanoparticles and enzymes is presented which can ultimately reduce the cost of whole process. To our knowledge, this is the first study on claiming laccase and hemicellulase mimicking activity of CeFeO and their application in raw corn cob conversion into bioethanol production, which can negate the need of pretreatment process and costly laccase enzymes. Hence, CeFeO-associated biomass to bioethanol process may pave the way toward development of green sustainable biofuel technology.
