2. 각 최적 추출 조건 하에서 얻은 punicalagin 및 ellagic acid 추출물을 같은 조건에서 추출한 물 추출물과 비교하고자 이들의 기능성 성분 함량을 측정한 결과, 총 폴리페놀 함량은 PE-DES가 88.15 mg GAE/g, 252.36 mg CAE/g, 총 플라보노이드 함량은 EE-DES가 22.76 mg QE/g으로 나타나 동일한 조건일 때의 물 추출물보다 유의적으로 높은 함량을 보였다. 페놀 화합물 분석 결과, 물 추출물보다 DES 추출물에서 다양한 폴리페놀이 검출되었으며, 특히 PE-DES의 총 페놀 화합물 함량이 가장 높았다.

This study was conducted to select the optimal extraction conditions of deep eutectic solvent (DES)-based ultrasound-assisted extraction (UAE) for extracting functional ingredients from pomegranate peel (PP) using response surface methodology through Box-Behnken design. In addition, functional components and physiological activity of DES extracts obtained under optimal extraction conditions were compared with water extracts obtained using water as solvent under the same conditions.
1. The extraction efficiency for polyphenol, punicalagin, and ellagic acid according to DES types was compared. As a result, DES composed of choline chloride and urea, which is capable of extracting polyphenols including punicalagin and ellagic acid, was selected as a solvent.
2. A Box-Behnken design was used to investigate the effects of four independent variables (water contents of DES, liquid to solid ratio, ultrasonic power, extraction time) on the responses such as total polyphenol content (Y1), punicalagin content (Y2), and ellagic acid content (Y3).
3. The optimal extraction conditions are as follows: total polyphenol content (water contents of DES, 29.30%; liquid to solid ratio, 53.50 mL/g; ultrasonic power, 238.20 W; extraction time, 29.50 min), punicalagin content (water contents of DES, 25.65%; liquid to solid ratio, 44.20 mL/g; ultrasonic power, 120 W; extraction time, 20 min), and ellagic acid content (water contents of DES, 33.13%; liquid to solid ratio, 60 mL/g; ultrasonic power, 300 W; extraction time, 20 min). Under the optimal extraction conditions, the experimental values for total polyphenol content, punicalagin content, and ellagic acid content were 67.05 mg GAE/g, 130.65 mg/g, and 2.64 mg/g, respectively.

Punicalagin extract obtained using UAE with DES (PE-DES) presented the highest total polyphenol content (88.15 mg GAE/g, 252.36 mg GAE/g), and ellagic acid extract obtained using UAE with DES (EE-DES) showed the highest total flavonoid content (22.76 mg QE/g). The eleven types of phenolic compounds were detected in the extracts obtained using DES, and PE-DES extract showed the highest phenolic compounds content.
5. DPPH radical scavenging, ABTS radical scavenging, and metal-ion chelating activity were measured and IC50 values were calculated. DES extracts showed lower IC50 values than water extracts. In particular, PE-DES extact exhibited the lowest IC50 values in DPPH radical scavnenging activity (65.69 µg/mL), ABTS radical scavenging activity (0.77 µg/mL), and metal-ion chelating activity (114.23 µg/mL).
6. To evaluate the antidiabetic activity of PP extracts, α-glucosidase inhibitory activity was measured and the IC50 values were calculated. DES extracts showed lower IC50 values than water extracts. All PP extracts exhibited lower IC50 values than the Acarbose, and PE-DES had a significantly the lowest IC50 value (6.49 µg/mL) among all extarcts.
7. DES extracts showed higher pancreatic lipase (PL) inhibitory activity than water extracts. PE-DES showed the highest PL inhibitory activity of 45.07%, 55.63%, 72.69%, 82.25% in 10 µg/mL, 20 µg/mL, 30 µg/mL, and 40 µg/mL, respectively.
