It has been shown that brain-derived neurotrophic factor (BDNF) promoted survival of the dopaminergic neurons, and improved motor function in the models of PD suggesting BDNF is one of the promising therapeutic agents for PD treatment. Administering BDNF per se would not be a viable method as it cannot cross the blood-brain barrier. Hence, one of the strategies for the therapeutic application of BDNF to the neurodegenerative disease is to upregulate the BDNF level in the brain. Inosine is a metabolite of purinergic nucleosides and activates adenosine receptors which in turn elevates the level of BDNF. In this study, we evaluated the neuroprotective effects of inosine and elucidated the underlying mechanisms for the neuroprotective action. We checked the neuroprotective effects of inosine using the SH-SY5Y neuroblastoma cells and the results revealed that inosine protected the neuroblastoma cells from MPP+ in a dose-dependent manner. We found that the neuroprotective effects of inosine were associated with the induction of BDNF and activation of its downstream signaling pathway, as Trk-B receptor inhibitor, K252a or BDNF gene silencing using siRNA significantly attenuated the protective effect of inosine in MPP+ treated cells. In addition, we found that the A2a receptor antagonist SCH58261 mitigated inosine-induced BDNF induction and consequent protection of the SH-SY5Y cells, implying the significant role of adenosine A2a receptor in BDNF induction by inosine. Next, we assessed the dopaminergic neuroprotective effects of inosine in C57BL/6 mice. Pretreatment of inosine for 3 weeks clearly upregulated BDNF and activated its downstream targets. Importantly, inosine reduced the dopaminergic neuronal loss caused by MPTP in the substantia nigra. Furthermore, inosine ameliorated depletion of the striatal dopamine and its metabolites after MPTP injection. Overall, this thesis clearly demonstrated that inosine could spare the neuron from the neurotoxicants by inducing BDNF via activating the A2a receptor. These findings highlight the potential of inosine to prevent or intervene in the dopaminergic neurodegeneration in the PD brains.
