The aims of this study were to optimize fermentation conditions for probiotic Lactobacillus plantarum 200655 and develop synbiotic fermented soymilk. L. plantarum 200655 has been reported to provide antioxidant and neuroprotective effects against oxidative stress. Optimization was conducted using one-factor-at-a-time (OFAT) method, Plackett-Burman design (PBD), and response surface methodology (RSM). The optimized medium composition was 31.29 g/L maltose, 30.27 g/L yeast extract, and 39.43 g/L soytone, with the maximal biomass of 3.845 g/L. The optimal culture conditions were determined to be pH 6.5 and 30°C. The batch fermentation was conducted under optimized conditions with a jar fermentor. As a result, the highest biomass (5.866 g/L) was obtained after 18 h of fermentation, which was 2.34-fold higher compared to MRS medium. The effect of fructooligosaccharides (FOS) on soymilk fermentation and neuroprotective effects of fermented soymilk (FS) were also investigated. FOS supplement increased β-glucosidase activity of L. plantarum 200655 by 4.26−20.74% and total phenol content in FS by 52.95−55.94%, resulting in enhanced antioxidant activities. In addition, FS with 3% and 5% FOS (FS3 and FS5) exerted neuroprotective effects on SH-SY5Y cells against H2O2-induced oxidative stress. FS3 and FS5 increased cell viability and decreased LDH leakage of SH-SY5Y cells. Pretreatment of FS3 and FS5 attenuated neuronal apoptosis by regulating the gene expression of neuronal biomarkers and apoptosis-related factors. This study suggested the optimized conditions for enhanced biomass of L. plantarum 200655 and the potential use of synbiotic fermented soymilk as a functional food with neuroprotective effects against oxidative stress.
