Hemp seed is rich in omega-3 polyunsaturated fatty acids and protein, and hemp seed oil consists mainly of essential fatty acids, linoleic acid, α-linolenic acid and oleic acid. The main psychoactive components of cannabis are Δ9-tetrahydrocannabinol (THC), cannabidiol (CBD) and cannabinol (CBN). The purpose of this study is to analyze the cannabinoid concentrations of hemp seeds and hemp seed oil distributed in Korea and determine whether they meet the THC and CBD standards of the Korean Ministry of Food and Drug Safety (MFDS). As an analytical method, an internal standard was added using homogenized hemp seeds or a hemp seed oil sample diluted 10-fold in methanol, and then methanol was added, followed by shaking, sonication and centrifugation. Then, C18 200 mg + primary secondary amine (PSA) 100 mg and refrigeration storage time were used for purification. Finally, the filtered supernatant was injected into LC-MS/MS. In this study, THC concentrations ranged from 0.022 to 3.153 (mean 0.416 μg/mg) and CBD concentrations ranged from 0.053 to 3.044 (mean 0.671 μg/mg) in hemp seed. THC concentrations ranged from 0.103 to 1.527 (average 0.642 μg/mL) and CBD concentrations ranged from 1.153 to 11.518 (average 4.320 μg/mL) in hemp seed oil. In addition, through the THC/CBD, (THC+CBN)/CBD, CBN/THC and log(THC/CBD) ratios, drug-type marijuana and fibre-type hemp are distinguished, and three Chemotypes can be classified and the freshness of hemp seed and hemp seed oil can be evaluated. THC/CBD, (THC+CBN)/CBD, CBN/THC, and log10(THC/CBD) ratios are as follows: THC/CBD ranged from 0.039-2.046, (THC+CBN)/CBD ranged from 0.039-2.148 in hemp seed. THC/CBD ranged from 0.073-0.464, (THC+CBN)/CBD ranged from 0.073-0.563 in hemp seed oil. Through the method established in this study, the cannabinoid contents in hemp seed and hemp seed oil can be continuously monitored and the results in this study could provide basic data for regulatory agencies.
