괭이신나무는 단풍나무과 식물로, 잎은 난상 타원형으로 대생하며 꽃 은 황백색으로 복산방화서 형태로 달린다. 나무껍질은 거칠며 회갈색을 띄나, 열매는 다른 신나무 종들에 비하여 시과가 넓게 벌어지는 것이 특징이다. 괭 이신나무(Acer tataricum subsp. ginnala or Acer ginnala for. divaricatum),(AG) 는 항염 및 항산화 등 다양한 생리활성효과가 있다고 알려져 이전부터 쓰이 던 약재이나, 피부 광노화에 대한 보호 효과에 대해서는 보고된 바가 없다. 따라서 이번 연구에서는 인간 진피 섬유아세포를 이용하여 AG의 광노화 억제 효능 및 작용기전을 규명하고자 하였다. 본 연구에서, AGE는 UVB 조사에 의해 유도된 콜라겐 분해효소인 MMP1의 증가 및 Type 1 procollagen의 감소를 농도의존적으로 억제하였다. 콜라겐 분해에 관여되는 pathway에는 MAPK/AP-1과 NF-κB pathway가 있다. MAPK/AP-1 pathway의 경우, UVB조사 시 MAP kinase (ERK, JNK, p38) 인산화 의 증가로, AP-1의 활성을 높임으로써 MMP1 발현을 증가시킨다. AGE는 MAP kinase (ERK, JNK, p38)의 인산화를 억제하여, AP-1의 활성을 낮춤으로써 콜라 겐 분해를 억제하는 효과를 보였다. 또다른 콜라겐 분해경로인 NF-κB pathway에서, AGE는 UVB에 의한 IκB의 발현 저해 및 NF-κB의 핵내 이동을 모두 억제 시킴으로써, MMP1의 발 현을 억제하였다. NF-κB pathway는 염증 유발인자를 발현 시키는 것으로도 알려져 있으며, 본 연구에서도 AGE는 콜라겐 분해를 유도하는 TNF-α, IL-1β, IL-6 과 같은 염증성 사이토카인의 발현을 억제시켰다. 콜라겐 생성 관련 기전인 TGFβ/Smad pathway의 경우, AGE는 UVB에 의해 저해된 TGFβ Receptor Ⅱ의 발현 증가 및 하위인자인 Smad3의 인산화 를 회복시키고, Smad4와 함께 핵 내로 이동하여 콜라겐 합성을 증가시켰다. 또한 TGFβ Receptor에 결합하여 Smad3 와의 경쟁적 억제를 일으키는 Smad7 의 발현을 AGE가 억제시키는 것도 확인하였다. 이는 AGE가 콜라겐 합성에 중요하게 작용하는 TGFβ/Smad pathway에도 작용하여 광노화를 억제하는 것 을 의미한다. 따라서, 본 연구는 AGE가 UVB로 인한 콜라겐 분해를 억제하고, 콜라 겐 합성은 증가시킴으로써, 광노화를 억제하는 기능성 물질로 활용될 수 있음 을 시사한다.

Skin, the organ to protect the human body from external factors, maintains structure and tensile by containing many fibril collagen, particularly type 1 procollagen, in the extracellular matrix (ECM). However, oxidative stress by ultraviolet (UV) exposure causes skin photoaging by activating collagen breakdown and inhibiting collagen synthesis. Acer tataricum subsp. ginnala (AG) is an herbal medicine with anti-oxidative and anti-inflammatory effect, but there is no report about the protective effect against skin photoaging. Therefore, we conducted the research concentrating on the anti-photoaging effect in human dermal fibroblasts (HDF). In this study, we primarily found that AGE downregulates the expression of matrix metalloproteinase1 (MMP1) and recovers the expression of type 1 procollagen damaged by UVB, which are the main factors to influence skin photoaging. Then, we investigated mitogen-activated protein kinases/activator protein 1 (MAPK/AP-1) and nuclear factor-κB (NF-κB) pathway which induce collagen degradation by promoting MMP1 level and pro-inflammatory cytokines. The results indicated that AGE downregulates the expression of MAPK/AP-1 pathway, leading toMMP1 reduction. AGE inhibits nuclear translocation of NF-κB and inhibitor of nuclear factor-κB (IκB) degradation. Therefore, it downregulates the expression of MMP1 and proinflammatory cytokines, TNF-α, IL-6, and IL-1β increased by UVB. Besides, TGFβ/Smad pathway which is mainly responsible for the collagen synthesis in the skin was also analyzed. AGE decreases the expression of Smad7 and increases TGFβRⅡ expression and Smad3 phosphorylation. This means that AGE stimulates TGFβ/Smad pathway that plays a critical role in promoting collagen synthesis. Thus, this study suggests that AGE can be a functional material with the anti-photoaging property.
