DJ(Dioscoria japonica tunb.) is a perennial vines herb of the Dioscorea family with medical ingredients such as saponin, mucin, and arginine. No research has been reported on the anti-inflammatory and skin protection effects of particulate matter using yam extract. Therefore, this study confirmed whether DJ extract inhibits skin damage and inflammation caused by particulate matter. The total polyphenol content was 4 ± 0.08 mg GAE/100g, and the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability was evaluated, and the scavenging ability was 84 ± 1.05% at 100 mg/mL concentration. In order to identify the anti-inflammatory effect, LPS stimulation was given to RAW 264.7 cells, and NO production was evaluated, and NO production was significantly decreased by DJ extract treatment. The production of IL-6 and TNF-α, known as inflammatory cytokines, was measured, and the expression was suppressed in a concentration- dependent manner by DJ extract treatment. In order to identify the cellular protection effect for UV-B in Detroit 551 cells, UV-B was irradiated, DJ extract was treated, and RT-PCR was performed, and MMP-1, MMP-2, MMP-9 were performed. The amount of mRNA expression was decreased in a concentration-dependent manner. In the model of inflammatory mice induced by particulate matter, DJ extracts did not affect the colon length, and decreased the number of intestinal bacteria. Therefore, DJ extracts are considered to be available in cosmetics and various fields as natural materials with effectiveness such as antioxidation, anti-inflammation, and anti-aging.
