Solubilization of high-concentrated adenosine using mixed hydrotrope and the application to anti-wrinkle cosmetics원문보기

Adenosine is a typical active ingredient used for cosmetics to improve wrinkle problems. However, because of the low solubility of adenosine in an aqueous solution, the amount of adenosine in cosmetics is limited with a very small amount. From the previous research, it was confirmed that the solubility of caffeine, which is structurally similar to adenosine, in water was improved by using hydrotrope. Niacinamide and vanillin were used for hydrotrope agents. When dissolving adenosine alone in the water, only 0.4% of adenosine was dissolved at 20℃, and only 0.5% at 25℃. When adenosine and niacinamide were dissolved together in water, adenosine and niacinamide were dissolved in the ratio of 1% to 3.4%, and 2% to 8% at 20℃. At 25℃, they were in the ratio of 1% to 2.7%, and 2% to 6.8%. The availability of vanillin as a hydrotrope agent was also verified, but the result was different from what was expected, as adenosine and vanillin together were not able to dissolve in water. Moreover, the amount of niacinamide required for dissolving 2% adenosine exceeds the amount of 2~5%, the prescribed amount used in wrinkle improvement functional cosmetics, the excessive amount that can lead to skin irritation. Therefore utilizing mixed hydrotrope was necessary to solve these problems. Optimizing the content ratio of niacinamide and vanillin of mixed hydrotrope to dissolve 1% and 2% of adenosine at 20℃ and 25℃ respectively were tried through this experiment. The content ratio of adenosine and niacinamide for the experiment was set based on the results of previous research using niacinamide as a hydrotrope agent to dissolve adenosine. Optimizing the ratio of niacinamide and vanillin of mixed hydrotrope was done by making difference in the content ratio of vanillin used in each experiment. As a result, using mixed hydrotrope with vanillin, less niacinamide was required for dissolving adenosine. Reducing the proportion of niacinamide increased the vanillin content to dissolve the same amount of adenosine, but resultingly this lead to using less amount of total hydrotrope agents. In higher temperature condition, less amount of mixed hydrotrope in total, and also less amount of niacinamide and vanillin respectively were required to dissolve adenosine. Based on these results, cosmetics of skin-softener, lotion and cream containing 2% of adenosine were invented and went through clinical trials. Periorbital wrinkle, skin melanin content, skin brightness, skin moisture, and transepidermal water loss were evaluated. These clinical trials went for four weeks and showed significant results of these cosmetics with 2% adenosine are effective in wrinkle improvement and also improving overall skin conditions. Therefore these results are expected to be utilized widely in the constantly growing anti-aging market and cosmetics.
