The main carcinogenic pathways of CRC that have been clarified at present include the accumulation of mutations leading to oncogene (KRAS) activation and tumor suppressor gene (DCC, APC, SMAD4, TP53) inactivation, mutations in DNA mismatch repair genes (MSH2, MLH1, MSH6, PMS2, MLH3, MSH3, PMS1, Exo1), and aberrant methylation that are mechanisms that silence gene function. Another mechanism was to activate EGFR kinase and reduce phosphorylation of proteins downstream of EGFR. Physiological active substances induced apoptosis by external exposure of phosphatidylserine to cells, cell cycle arrest, depolarization of mitochondrial membrane potential, and Multi-caspase activation. Western blot experiments confirmed that physiological active substances induce apoptosis by regulating ER stress, apoptosis, and cell cycle-related proteins. In conclusion, naturally product-derived bioactive substances have shown molecular mechanisms for ROS-induced cell proliferation, cell cycle arrest and apoptosis induction in digestive system cancer cells. Physiological active substances of nature products induced apoptosis by activating the JNK / p38 MAPK signaling pathway by increasing ROS, which is an oxidative stress in cancer cells. Therefore, physiological active substances derived from natural products have shown potential as new anticancer adjuvants and anticancer agents in digestive system cancer.
