OsHY5.2 유전자는 target1에서 46bp, 40bp의 염기가, target2에서 14bp의 염기가 결실되었다. 세 패턴 모두 단백질의 비정상적 번역에 의해 early stop codon이 생성된 것을 확인했다. OsHY5.3 유전자는 아데닌(A)과 티민(T)이 삽입된 두 개의 패턴과 6bp가 삭제된 한 개의 패턴으로 총 3개의 패턴을 확인했다. 세 패턴 모두 종자의 크기가 왜소하고 뿌리 발육이 좋지 못했으며 발아율이 현저하게 낮은 것을 볼 수 있었다. 또한, 티민이 삽입된 패턴의 경우 야생형과 비교하여 엽색이 뚜렷하게 옅은 것을 확인할 수 있었다. 아데닌이 삽입된 패턴은 단백질의 비정상적 번역에 의해 종결 코돈이 생성되지 않는 것을 확인했으며, 티민이 삽입된 패턴은 그 위치에 바로 종결 코돈이 생성돼 early stop codon을 확인했다. 6bp가 삭제된 패턴은 도메인의 앞쪽 단백질 2개가 결실된 것을 확인하였다. SlHY5 유전자는 2bp, 49bp의 염기가 삭제된 두 패턴을 확인했다. 두 패턴 모두 단백질의 비정상적 번역에 의한 early stop codon을 확인할 수 있었다. 엽색이 녹색인 야생형과 비교하여 돌연변이 식물체에서는 옅은 연두색을 띠었고 잎의 끝에서부터 하얗게 말라가다가 결과적으로 회복이 되지 않는 표현형을 보였다. 본 연구에서는 벼와 토마토에서 CRISPR/Cas9 기술을 이용하여 벼에서 총 11개 패턴의 순계 계통 식물체와 토마토에서 2개 패턴의 순계 계통 식물체를 각각 확보하였고 토마토 야생형 식물체에서 조직별로 HY5 유전자의 발현량을 확인하였으며 염기의 삭제 또는 삽입에 의한 단백질의 이상 번역, 이에 따른 표현형을 확인했다. 또한, 토마토에서 HY5의 과발현 식물체 2개 계통을 확보하였다. 추후 연구를 통해 앞서 확보한 돌연변이 식물체와 함께 과발현 식물체를 확보하여 벼와 토마토에서 HY5 유전자의 기능을 밝히는 연구가 진행되어야 할 것으로 생각된다.

The HY5 gene is a transcription factor comprising a bZIP domain family with asn-arg/lys-leu motif. It has been found to play a variety of roles in plant developmental stage, biosynthesis, and several signaling pathways in Arabidopsis thaliana. However, there are few studies of the HY5 gene in rice (Oryza sativa) and tomato (Solanum lycopersicum). Through the orthologues analysis of Arabidopsis HY5 gene, three of OsHY5.1, OsHY5.2, OsHY5.3, and single SlHY5 genes are isolated in Rice and Tomato, respectively. Cloning of OsHY5 and SlHY5 was conducted based on the separated genes, and genes were edited using the CRISPR/Cas9 system. Editing of the target gene was confirmed through deep-sequencing and sanger-sequencing. The OsHY5.1 gene had 36bp, 39bp, and 25bp bases deleted from target1 and 159bp bases deleted from target2. Protein sequences were analyzed based on the amino acid sequence of the edited plants, and 25bp of target1 and 159bp of target2 were found to have been generated early stop codons by abnormal translation of proteins in the deleted plants. The OsHY5.2 gene was deleted 46 bp and 40 bp bases from target1 and 14 bp bases from target2. In all three patterns, it was confirmed that early stop codon was produced by abnormal translation of proteins. The OsHY5.3 gene identified a total of three patterns: two patterns in which adenine (A) and thymine (T) were inserted and one pattern in which 6bp was deleted. In all three patterns, it was confirmed that the seed size was small, the root showed an impaired phenotype, and the germination rate was significantly low. In addition, in the case of a pattern in which thymine was inserted, it was confirmed that the leaf colour was clearly pale green compared to the wild type. It was confirmed that the pattern in which adenine was inserted did not produce a termination codon by abnormal translation of the protein, and the pattern in which thymine was inserted created immediately at that location, confirming the early stop codon. In the pattern in which 6bp was deleted, it was confirmed that two proteins in the front of the domain were lost. The SlHY5 gene identified two patterns in which 2bp and 49bp bases were deleted. Compared to the wild type, the leaf colour turn pale green, and it showed blight phenomenon from the end of the leaf and eventually died. In both patterns, early stop codon due to abnormal translation of proteins were confirmed. In this study, CRISPR/Cas9 technology was used in rice and tomato to obtain a total of 11 patterns in rice and 2 patterns in tomato, the expression level of the HY5 gene by WT tissue in tomato was confirmed, and abnormal translation of proteins by deletion or insertion of bases, and phenotypes accordingly were confirmed. In addition, two lines of overexpressed plants of HY5 gene were secured in tomato. Through further research, it is thought that research should be conducted to reveal the function of the HY5 gene in rice and tomato by securing overexpression plants along with mutant plants previously secured.
