The genome editing tool consists of a guide RNA that guides to a target sequence and a Cas protein that cleavages the targeted region. To achieve sequence-specific cleavage, the Cas protein must identify a protospacer adjacent modif (PAM) sequence prior to protospacer-spacer base pairing. Each Cas protein has a distinct preference for PAM sequence. For instance, Cas9 protein recognizes 5'-NGG-3', Cas12a recognizes 5'-TTTV-3' (V=A/C/G), and Cas12f1 recognizes 5'-TTTR-3' (R=A/G). Given the PAM preference of each CRISPR-Cas system, the limited recognition could be problematic due to the lack or unavailability of targetable sites within a certain region. In this study, we aim to develop PAM variants of Cas12f1 so that they can expand the targetable sites using the hypercompact CRISPR system. For this aim, various candidates were selected by several amino acid sequences the are assumed to be involved in the recognition of the PAM sequence through the structural analysis of Cas12f1, and the cleavage pattern of the PAM variants were tested following the production in E. coli cells. During the selection of candidates, any variants with the cleavage for TNTN PAM were screened in in vitro system. Thereafter, the candidate variants were investigated in HEK293T cells with repect to the indel formation activity. As a results, we could develop several Cas12f1 PAM variants will contribute to expanding the opportunites for the development of various gene-editing therapeutics by overcoming the PAM limitation of our hypercompact genome-editing platform.
