CO2 레이저가공 및 후속 다층 접합 공정은 비용이 효율적이며 제작이 간단하다는 면에서 위에 언급한 다른 가공기술들에 비해 두드러진 장점을 가지고 있다. 또한, 공정에서 이용되는 대부분의 기판 재료인 열가소성 플라스틱은 우수한 생체 적합성과 용제 상용성(solvent compatibility)을 가지고 있어 핵산분석에 적합하다. 원형 미소 챔버를 가지고 있는 미소유체 장치는 쉬운 세포 용해와 DNA 추출을 위해 사용된다. 미소유체 장치는 CO2레이저 가공과 용제 보조 열 접합 (solvent-assisted thermal bonding)을 통해 빠르게 제작된다 (~25분). 먼저, 레이저 가공 파라미터가 원하는 치수(CAD 디자인)로의 가공과 레이저로 절단된 형상의 품질에 어떠한 영향을 미치는지 조사하였다. 이를 위해 흔히 사용되는 열가소성 플라스틱인 PMMA (polymethyl methacrylate)을 간단한 디자인(수직 마이크로 채널)으로 가공하였다. PMMA는 비용이 저렴하고 레이저 가공 중 PC (polycarbonate)와 같은 다른 열가소성 플라스틱에 비해 표면 거칠기가 적고 버 (절단가장자리를 따라 PMMA가 녹아 응고된 돌출된 형상의 파형)의 높이가 낮다고 알려져 있다. CO2레이저가공은 미소 형상(microfeature)의 패터닝에 널리 이용되어 왔지만, 높은 열 에너지를 열가소성기판에 가하여 패터닝하기 때문에 형상해상도 (feature resolution)와 절단면의 표면 거칠기를 저하시킨다. 본연구에서는 (1) 보다 더 미세한 형상해상도를 얻기 위해 레이저 절단 파라미터의 실험적이지만 체계적인 최적화를 위한 DOE(design of experiments)를 제안한다. (2) 절단면의 표면 거칠기와 버의 형성에 관한 레이저 파라미터가 어떠한 영향을 미치는지 조사한다. (3) 우수한 치수 무결성 (dimensional integrity)과 용제의 누출 없이 레이저로 가공된 PMMA 층을 접합하기 위해 용제 노출 (solvent exposure) 및 열 접합 파라미터가 실험적으로 선정된다. 선정된 가공 파라미터를 이용해 부드러운 절단면 (조도 프로파일의 산술평균 Ra=0.07 μm)과 최소화된 버의 높이 (0.34 µm) 및 높은 종횡비 (4.2:1)를 가진 세계 최초 100 µm이하의 채널 폭 (56 µm)을 얻었다. 또한 채널 내부 표면 거칠기는 PMMA를 관통 가공하고 커버슬립을 아래위로 접합했을 때 감소하였다. 이 결과는 채널을 가공하는데 있어 레이저 미세가공에서 많이 이용하는 engraving방식과 극명한 대비를 보인다. Engraving방식으로 가공했을 때 채널의 바닥 표면기가 Ra=~21.4 μm로 상당히 거칠었으며 이는 광학적 분석 (optical analysis)을 어렵게 하고 거친 표면에 생체분자의 흡착을 촉진한다. 가장 작은 채널폭을 가진 미소유체 장치를 제작하기 위해 50-μm 두께의 PMMA를 이용하였으며 가공 파라미터를 정밀하게 조정하였다. 그 결과 거의 수직인 종횡비 (0.9:1)와 46.3-μm 채널 폭을 가진 미소유체 장치를 제작하였다. 46.3-μm 채널 폭은 지금까지 CO2레이저로 관통 가공한 채널 폭 중 가장 작은 폭이다. 이후 미소 챔버 기반 미소유체 장치는 이전에 직선 마이크로채널을 가공하는데 사용했던 기술(레이저가공, 용제보조 열 접합)을 이용하여 제작된다. 폴리카보네이트는 (polycarbonate)는 에세이 관찰 (assay observation)을 위한 광학적 투명도 (optical clarity), 용제 상용성 (세포용해 프로토콜 동안 에탄올이 사용됨), 생체 적합성에 기인해 기판으로 사용된다. 레이저 절단 조건은 생체분자가 흡착되어도 그 정도가 무시할 만한 깨끗한 절단면과 접합에 지장을 주지 않을 정도로 최소화된 버를 얻기 위해 DOE를 사용하지 않고 실험적으로 결정된다. 용제 노출과 열 접합 조건 역시 누수가 없을 뿐만 아니라 우수한 치수 무결성을 가진 장치 제작을 위해 실험적으로 선정하였다. 화학적 세포용해와 DNA 추출은 미소유체 포맷에서 수행되었지만 낮은 레이놀즈 수에 기인한 비효율적인 물질전달로 인해 성능이 제한된다. 본 연구에서는 비효율적인 물질전달을 개선하여 화학적 세포용해와 DNA의 자성입자 기반 동적 고상 추출 (magnetic-bead-based dynamic solid phase extraction)을 향상시키기 위한 방법으로 공동 마이크로스트리밍을 제안한다. 공동 마이크로스트리밍 조건은 전기 임피던스 분광법을 통해 측정된 미소유체 칩의 굴곡 공진 주파수 (flexural resonance frequency) (fr)에서 칩을 가진하여 최적화된다. 굴곡 공진 주파수에서 가진된 버블은 그 주변에 강한 순환 유동 (circulatory flows)을 발생시킨다. 이러한 강한 순환 유동은 격렬한 교반을 일으켜 빠른 세포용해와 DNA의 추출 및 정제를 가능하게 한다. 우리의 세포용해 및 DNA 추출 방식은 완전히 최적화되지는 않았지만 상업용 키트 (commercial kit)나 이전의 미소유체 포맷에서 수행된 방식에 비해 우수한 DNA 추출 성능을 보여준다. 짧은 분석 시간 (~25분)동안 높은 추출 효율 (76.9%)과 순도 (A/260/A280=1.85)를 보인다. 순도는 260 nm와 280 nm의 파장에서 UV흡광도 (UV absorbance)의 비(A260/A280)로 측정된다. 특히 고희석 세포 샘플 (0.18 cells/μl)에서 단지 18개의 세포로만으로 DNA가 성공적으로 추출될 수 있다. PCR 및 전기영동법을 통해 추출된 DNA의 우수한 품질을 확인하였다. 이러한 괄목할 만한 성능을 고려하여 우리의 DNA 추출방식이 미소유체 핵산 분석 장치에 널리 사용되기를 기대한다. 또한 빠르고 클린룸에서 자유로운 제작 방식과 비용 효율적인 압전 변환기 (PZT transducer)를 통해 생성되는 간단한 미소 혼합(micromixing) 방식을 채택한 우리의 장치는 다양한 POC NAT 어플리케이션 중 시료 전처리에 있어 충분한 잠재성을 지니고 있다고 판단된다. 마지막으로 설계변경을 거의 하지 않고 고해상도 (high resolution)의 마이크로 채널을 가진 미소유체 장치 또한 우리가 제안한 세포용해 및 DNA 추출 방식을 기반한 핵산 분석을 위한 용도로 사용될 수 있다.

The ever-increasing danger of infectious diseases on human lives has recently been amplified by the highly-pathogenic viral disease, COVID-19. The large-scale spread of such kinds of infectious diseases can be curbed using point-of-care (POC) testing, which has been essential for quick diagnosis as it is conducted near or at the site of the patient. Especially, POC nucleic acid testing (NAT) is appreciated due to its high sensitivity and specificity compared to diagnostic methods that are based on phenotypic expression of antigens. POC NAT uses microfluidic devices to perform nucleic acid (NA) analysis. However, the initial and most critical step in NA analysis, cell lysis and DNA extraction, is significantly challenging in microfluidic devices due to the difficulties of translating the conventional macroscale methods to the microscale. In this work, a simple and cost-effective microfluidic lysis and DNA extraction method is proposed. Furthermore, the microfluidic device is made using a straightforward and affordable microfabrication technique. Fabrication techniques including conventional clean-room-based lithography, soft lithography, 3D printing, and laser machining of a thermoplastic substrate followed by multilayer bonding have been used to develop microfluidic devices for POC NAT. CO2-laser machining and subsequent multilayer bonding stands out from the rest owing to the cost-effective and straightforward fabrication. Moreover, most of the substrates used for the fabrication, thermoplastic sheets, are suitable for NA analysis as they have superior biocompatibility and solvent compatibility. A microfluidic device with a circular microchamber is used for the facile lysis and DNA extraction. The device is rapidly fabricated (~25 min) using CO2-laser machining and solvent-assisted thermal bonding. However, initially, the effect of fabrication parameters on the desired dimension and quality of a laser-cut feature is studied using a simpler design (i.e., a straight microchannel) and commonly used thermoplastic, PMMA. PMMA is known for its low cost, and minimal surface roughness and reduced burr (ridge-shaped waveform of solidified molten PMMA along the edge of the cut) during laser ablation, compared to other common thermoplastics such as polycarbonate (PC). CO2-laser machining has been widely used for patterning microfeatures, but the high heat deposition on thermoplastic substrates generates poor feature resolution and surface quality of cut surfaces. In this work, the use of a design of experiments (DOE) is proposed for experimental yet systematic optimization of laser-cutting parameters to obtain finer feature resolution. The effect of the parameters on the formation of cut-sidewall surface roughness and burr is also investigated. Solvent exposure and thermal bonding parameters are also experimentally determined to ensure the bonding of laser-cut PMMA layers with excellent dimensional integrity and no leakage. Using the proposed method, an unprecedented sub-100-µm channel width (56 µm) with a high aspect ratio (AR) of 4.2:1 having smooth sidewall surfaces (i.e., estimated using the arithmetic average of the roughness profile Ra=0.07 µm) and minimized burr height (0.34 µm) is achieved. Furthermore, smoothness of the inner surfaces of the channel is enhanced by cutting through (i.e., penetrating) the PMMA sheet and enclosing it using fresh coverslips on the top and bottom. The result was a stark contrast to the many laser micromachining works that rely on engraving to create a channel, which leads to significantly rough bottom surfaces (Ra=~21.4 µm) that inhibit accurate optical analysis and promote nonspecific adsorption of biomolecules. In addition, a microfluidic device with the narrowest (46.3 µm) CO2-laser cut channel (cut-through) yet having a near-vertical sidewall and AR of 0.9 is achieved by employing thoroughly fine-tuned fabrication parameters and an ultra-thin 50-µm-thick channel layer. Later, the microchamber-based microfluidic device is fabricated based on the technique used to make the straight microchannel. Polycarbonate was used as a substrate due to its optical clarity for assay observation, solvent compatibility (i.e., ethanol is used during the lysis protocol), and biocompatibility. Laser-cutting conditions are experimentally determined without the use of DOE to achieve a clean sidewall for negligible nonspecific binding and minimal burrs for unobstructed bonding. Solvent exposure and thermal bonding conditions are also experimentally determined for a leakage-free device with excellent dimensional integrity. Chemical lysis and DNA extraction have been performed in a microfluidic format but the performance is limited by ineffective mass transport due to low Reynolds number. In this work, cavitation microstreaming is proposed for enhancing chemical lysis and magnetic-bead-based dynamic solid phase extraction (dSPE) of DNA by improving mass transport. Cavitation microstreaming condition is optimized by exciting a microfluidic chip at its flexural resonance frequency (fr) measured via electromechanical impedance spectroscopy (EMIS). Strong circulatory flows around bubbles excited at fr yields vigorous agitation, allowing fast lysis, and DNA extraction and purification. Our lysis and DNA-extraction method, although not fully optimized, exhibits an excellent DNA extraction performance, compared to a commercial kit and previous microfluidic methods. High extraction efficiency (76.9%) and purity (measured as a ratio of the UV absorbance at a wavelength of 260 nm and 280 nm; A260/A280=1.85) are achieved for a relatively short assay time (~25 min). Notably, DNA from as few as 18 cells is successfully extracted even from a highly diluted cell sample (0.18 cells/l). PCR and electrophoresis results confirm the excellent quality of the extracted DNA. Considering these notable performances, we anticipate our DNA extraction method will be widely used in microfluidic nucleic-acid analysis devices. Furthermore, the cleanroom-free and rapid fabrication, and straightforward micromixing generated by a cost-effective PZT transducer make our device promising for sample preparation in POC NAT applications. Finally, with few design modifications, the microfluidic device with a high-resolution microchannel can also be used for NA analysis based on the proposed method of lysis and DNA extraction.

CO2-laser machining has been widely used for patterning microfeatures, but the high heat deposition on thermoplastic substrates generates poor feature resolution and surface quality of cut surfaces. In this work, the use of a design of experiments (DOE) is proposed for experimental yet systematic optimization of laser-cutting parameters to obtain finer feature resolution. The effect of the parameters on the formation of cut-sidewall surface roughness and burr is also investigated. Solvent exposure and thermal bonding parameters are also experimentally determined to ensure the bonding of laser-cut PMMA layers with excellent dimensional integrity and no leakage. Using the proposed method, an unprecedented sub-100-µm channel width (56 µm) with a high aspect ratio (AR) of 4.2:1 having smooth sidewall surfaces (i.e., estimated using the arithmetic average of the roughness profile Ra=0.07 µm) and minimized burr height (0.34 µm) is achieved. Furthermore, smoothness of the inner surfaces of the channel is enhanced by cutting through (i.e., penetrating) the PMMA sheet and enclosing it using fresh coverslips on the top and bottom. The result was a stark contrast to the many laser micromachining works that rely on engraving to create a channel, which leads to significantly rough bottom surfaces (Ra=~21.4 µm) that inhibit accurate optical analysis and promote nonspecific adsorption of biomolecules. In addition, a microfluidic device with the narrowest (46.3 µm) CO2-laser cut channel (cut-through) yet having a near-vertical sidewall and AR of 0.9 is achieved by employing thoroughly fine-tuned fabrication parameters and an ultra-thin 50-µm-thick channel layer. Later, the microchamber-based microfluidic device is fabricated based on the technique used to make the straight microchannel. Polycarbonate was used as a substrate due to its optical clarity for assay observation, solvent compatibility (i.e., ethanol is used during the lysis protocol), and biocompatibility. Laser-cutting conditions are experimentally determined without the use of DOE to achieve a clean sidewall for negligible nonspecific binding and minimal burrs for unobstructed bonding. Solvent exposure and thermal bonding conditions are also experimentally determined for a leakage-free device with excellent dimensional integrity. Chemical lysis and DNA extraction have been performed in a microfluidic format but the performance is limited by ineffective mass transport due to low Reynolds number. In this work, cavitation microstreaming is proposed for enhancing chemical lysis and magnetic-bead-based dynamic solid phase extraction (dSPE) of DNA by improving mass transport. Cavitation microstreaming condition is optimized by exciting a microfluidic chip at its flexural resonance frequency (fr) measured via electromechanical impedance spectroscopy (EMIS). Strong circulatory flows around bubbles excited at fr yields vigorous agitation, allowing fast lysis, and DNA extraction and purification. Our lysis and DNA-extraction method, although not fully optimized, exhibits an excellent DNA extraction performance, compared to a commercial kit and previous microfluidic methods. High extraction efficiency (76.9%) and purity (measured as a ratio of the UV absorbance at a wavelength of 260 nm and 280 nm; A260/A280=1.85) are achieved for a relatively short assay time (~25 min). Notably, DNA from as few as 18 cells is successfully extracted even from a highly diluted cell sample (0.18 cells/l). PCR and electrophoresis results confirm the excellent quality of the extracted DNA. Considering these notable performances, we anticipate our DNA extraction method will be widely used in microfluidic nucleic-acid analysis devices. Furthermore, the cleanroom-free and rapid fabrication, and straightforward micromixing generated by a cost-effective PZT transducer make our device promising for sample preparation in POC NAT applications. Finally, with few design modifications, the microfluidic device with a high-resolution microchannel can also be used for NA analysis based on the proposed method of lysis and DNA extraction.
