Alzheimer's disease (AD) is a chronic neurodegenerative disease. The formation of neurofibrillary tangles, caused by hyperphosphorylation of tau protein, is one of the most prominent pathological events in the appearance and development of Alzheimer’s disease. O-GlcNAcylation, a post-translational modification, could protect tau protein from pathological tau-induced abnormal hyperphosphorylation in Alzheimer's disease (AD). O-GlcNAcase (OGA) catalyzes the removal of O-linked β-N-acetylglucosaminoglucose (O-GlcNAc) from tau. Therefore, using small molecule inhibitors to inhibit OGA would be a feasible way to increase O-GlcNAcylated tau levels to reduce tau hyperphosphorylation. We designed a novel non-saccharide scaffold for OGA inhibitors based on the structure of Thiamet-G. A series of target compounds were synthesized. Among them, compound 33e exhibited an IC50 value of 65 nM in vitro, no cytotoxicity, potential BBB permeability, and no inhibition on human lysosomal β-aminohexosidase A. In addition, molecular modeling studies showed that 33e bind to OGA via strong H-bonds and hydrophobic interactions. Overall, these findings suggest that 33e might be used as a potential AD therapeutic compound for the treatment of tau-related AD.
