Therefore, this study was initiated to solve environmental pollution associated with synthetic plastic waste using a microorganism biodegradation system via the digestive tract of mealworm beetle larvae, Tenebrio molitor. In this study, mealworms could live on a diet of synthetic plastics after receiving a steady diet of polystyrene, polyethylene, and polyurethane for up to 27 days. Subsequently, the process of biodegradation of microplastics in the digestive tract after ingestion through the oral cavity was analyzed from histological and microstructural aspects. Mealworm larvae showed advantages, compared with adults, since they had higher survival and plastic consumption rates. In addition, larvae plastic consumption rates increased with lower hardness, for example from polystyrene and polyethylene to polyurethane. Moreover, the plastic-fed group had lower survival rates than the bran-fed group. Histological and scanning electron microscopic observation suggested that plastic particles adversely affect the digestive tract activity, since histological damage to the digestive tract was observed, and the thickness of the muscular and epithelial layers was reduced. The original polystyrene surface structure remained within the hindgut, but it was found to be lost in the finally excreted frass. Conversely, polyethylene lost its original surface structure within the hindgut. In the case of polyurethane, some plastic particles that did not degrade in excreted frass, and polyurethane was estimated to be difficult to biodegrade. This study also demonstrated the fine structural analyses of the entire biodegradation process of the frass production from the originally intact plastics through the digestive tract of the larva. It was confirmed that the microplastic particles of an appropriate size for passage through the digestive tract were formed by the 'primary mechanical digestion process' through the mouth of T. molitor, and chemical degradation by microorganisms or enzymes occurred during passage through the larval digestive tract. Accordingly, this study could visualize the plastic biodegradation process in the mealworm digestive tract, showing that plastics are easily biodegraded in the order of simple molecular structure from polyethylene and polystyrene to polyurethane. Finally, in order to understand the plastic biodegradation mechanism of insects, reduction of variables and additional verification are required but this study is expected to be used as basic biological data for plastic waste biodegradation using the digestive system of arthorpod animals.
