Huntington's disease (HD) is an autosomal dominant genetic disorder caused by polyglutamine expansion of the Huntingtin (HTT) gene. HD is translated into the huntingtin (HTT) protein through a trinucleotide CAG repeat expansion. Also, loss of medium spiny neurons (MSNs) in the striatum is associated with motor deficits, resulting in cognitive deficits and signs of motor impairment in early-stage patients. A perfect therapeutic agent for treating HD has not yet been developed, and only drugs for symptom relief have been developed. We investigated the effect of HLA-homozygous induced pluripotent stem cell (iPSC)-derived neural precursor cells (iPSC-NPCs) as a therapeutic agent for HD treatment in 9-month-old YAC128 transgenic mice. In HD, there is much evidence for abnormalities in the corticostriatal circuit. A recent study have shown that dysfunction of the dopaminergic system in YAC128 transgenic mice results in behavioral and emotional abnormalities. Also, there is a study result showing that the mRNA expression level of dopaminergic receptors 1 and 2 decreased by MSN degeneration in old YAC128 mice. This is related to the direct pathway and the indirect pathway, which is closely related to the pathological features of HD. In particular, the dopaminergic R2(DRD2) is strongly associated with disease-wide aberrant changes in dopamine levels in HD patients and animal models, and thus restoring function for receptor expression related to this dopamine signaling will be an important role in the treatment of HD. The results of our study show that the transplanted cells exhibit various functions by transplanting (HLA)-homozygous iPSC-NPCs into the striatum.
