This study aimed to evaluate the osteogenic differentiation potential of human mesenchymal stem cells (hMSCs) by ultraviolet and plasma treatment on dental implants of resorbable blasting media (RBM) and sandblasted, large grit, acid-etched (SLA) treated surface.
Materials and Methods: Ultraviolet and plasma were treated on RBM and SLA titanium implants. Contect angle and wettability were evaluated using purified water. Human mesenchymal stem cells (hMSCs) were attached to the surface of each implant and the cell adhesion ability was evaluated using a scanning electron microscope. Cytotoxicity was analyzed by Live/Dead assay, and cell proliferation was (3-(4,5- dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) (MTS) assay was performed. Osteogenic differentiation was assessed by alkaline phosphatase (ALP) activity and alizarin red-sulfate (AR-S) staining.
Results: There was no difference between the RBM and SLA treatment methods, but when plasma was treated, the contact angle and wettability increased compared to ultraviolet treatment, and cell adhesion also increased. No cytotoxicity was observed due to ultraviolet and plasma treatment. In addition, cell proliferation and ALP activity showed similar levels in ultraviolet and plasma treatment, but a significant increase was shown in the evaluation of calcium accumulation in plasma treatment.
Conclusions: The application of plasma to dental implants improves the hydrophilicity of implants more than ultraviolet, promotes osteogenic differentiation of hMSCs. These results suggest a possibility as a method for successful implant placement by improving osseointegration of implant.
