Aedes albopictus, commonly known as the “Asian tiger mosquito”, is native to the tropical and subtropical regions of Southeast Asia. But the species has also been located from United States and Europe, establishing its heritage as an indigenous species with increasing global health concern. The arrival of Ae. albopictus in non-native habitats has been correlated to the competitive exclusion of Ae, aegyptii. Ae. albopictus is a blood-feeding insect that requires human or animal blood for growth and reproduction. It is a competent vector of more than 30 viruses, including the dengue virus, zika virus, and chikungunya virus that affect humans. The vector competency of Ae. albopictus and other mosquitoes refer to their intrinsic ability to acquire, host and transmit the viruses. With the advent of next-generation sequencing (NGS) and significant advances in genomics, novel insights have been revealed on host-pathogen interactions, immunogenetics, and viral evolution in mosquitoes. This has inspired innovative treatment and control strategies. A large proportion of the Korean mosquito community is accounted for by Ae. albopictus, although autochthonous clinical cases have not been reported. However, with the reported geographical genetic variations of Ae. albopictus in Korea, it is important to secure the molecular resources and different genetic markers that could be used to closely monitor the vector population and its competence for hosting viruses, thereby deciding on efficient vector surveillance and control. In order to conduct genome and transcriptome analysis, Ae. albopictus mosquitoes from were collected using BG sentinel traps and reared in laboratory environment. The DNA and cDNA library (from extracted total RNA) was sequenced using the Illumina HiSeq 4000 platform. About 39 Gb and 8.5 Gb of raw data was obtained from the genome and transcriptome of Korean Ae. albopictus. The contig assembly predicted genes that were subsequently annotated using PANM database and other public databases, The functional predictions were performed using the KOG database. Moreover, the unigenes obtained from the transcriptome were also annotated using the said databases as the genome, while functional predictions were performed using KOG, GO, KEGG, and IPS analysis. An attempt was also made to screen the polymorphic SSRs from the unigene information derived from the transcriptome. Further, comparative transcriptome of Ae. albopictus with or without the presence of DENV-like ssRNA have been studied., 0.4 Gb of raw data was obtained from both the groups of mosquitoes. Contigs were used to annotate public DB and local DB that formed by previous genome and transcriptome analysis. Afterwards, the functions of genes were classified, genes that do not cross each other between the experimental group and the control group were identified in each condition. Furthermore, DENV2 oral infection test was performed in an ABSL3 environment to understand the vector competency of Korean Ae. albopictus. The results confirmed the weaker disease mediating ability of Korean Ae. albopictus as a source of dengue virus. A basic dataset is ready for analyzing the genome, transcriptome, comparative analysis and vector competence of Korean Ae. albopictus. This result can be used as reference resource for conducting additional domestic vector research in the future.
