GFP mRNA를 포획한 지질 나노입자가 세포 내에서 단백질 번역을 유도할 수 있는지 확인하기 위하여 RAW 264.7, CT26 세포를 이용해 공초점 현미경과 유세포 분석기로 형광의 발현 여부를 관찰한 결과, DMKD-PS 지질 나노입자를 사용하였을 때 가장 우수한 GFP 발현 여부를 보였다. 종양연관항원인 MAGE-A3 mRNA를 이용하여 암 백신을 개발하였고 16주간 냉장 보관을 하여도 항원이 발현되는 것을 확인하였다. 마지막으로, 1) 암 백신을 접종한 후 피하에 종양세포 이식(종양의 예방), 2) 암 백신을 접종한 후 혈관 내로 종양세포 이식(종양 전이의 예방), 3) 종양을 형성한 마우스에 암 백신을 투여(종양의 치료) 하여 실험을 진행하였다. 결과적으로, 종양 예방 실험에서는 DMKD-PS mRNA 암 백신의 종양 크기 변화가 가장 적었으며 종양 치료 실험에서는 항암제 독소루비신과 비슷한 종양의 크기 변화를 보였으며 종양 전이 모델에서도 크게 개선된 생존율을 보여주었다. 암백신을 접종한 마우스의 생체 내 사이토카인과 분리한 비장의 배양액에서 IL-4, IFN-γ의 수준을 측정한 결과, 체액성 면역을 의미하는 Th2 우세 면역임을 확인하였다. 이러한 결과를 바탕으로, DMKD와 PS를 이용하여 제조된 MAGE-A3 mRNA 항암 백신은 종양 발생 예방, 종양 치료, 종양 전이의 예방 및 치료에 이용될 수 있을 것으로 보이며, 기타 mRNA를 이용하여 암 뿐만 아니라 다양한 감염 질환의 치료 및 예방에도 활용될 수 있을 것으로 기대된다.

Cancer, a global scourge that claims more than 10 million lives yearly, can be battled through various approaches, including surgery, radiotherapy, chemotherapy, and immunotherapy. Cancer vaccines, a key component of immunotherapy, target unique markers on cancer cells, known as tumor-specific or tumor-associated antigens. They mobilize the body's own immune system to fight cancer. One of the most advanced types of these vaccines is the mRNA vaccine. Similar to the technology used in COVID-19 vaccines, mRNA vaccines deliver genetic instructions to our cells to produce proteins that trigger an immune response against cancer. This approach has shown promising results, indicating a potential breakthrough in cancer treatment. However, the carriers employed in existing mRNA vaccines are structurally fragile and necessitate a cold-chain system, leading to safety and stability concerns. This study made a notable effort to enhance stability by leveraging a cationic lipid, O,O′-dimyristyl-N-lysyl aspartate (DMKD), to bind readily to mRNA. Furthermore, the strategy aimed to optimize antigen presenting by immune cells, utilizing phosphatidylserine (PS) to stimulate phagocytosis. The synthesized DMKD and PS-based particles (DMKD-PS) demonstrated their structural stability through little change in particle size despite the long-term refrigeration and presence of RNase, all while successfully containing mRNA. In vitro mRNA transfection of green fluorescence protein (GFP) revealed superior protein expression in immune and non-immune cells compared to lipid nanoparticles (LNPs) of other compositions. The In vivo biodistribution of DMKD-PS also showed that LNPs were safely eliminated from the body within 48 hr. Subsequently, MAGE-A3 mRNA was produced and encapsulated in various LNPs to assess the potential for preventing and treating colorectal cancer. The preventive and therapeutic efficacy of DMKD-PS, as evidenced by growth inhibition in tumor size, weight, along with its substantial impact against metastatic cancer, was confirmed. Furthermore, to verify the generation of immunity against tumors, an increase in immunoglobulins and specific cytokines was observed. Despite the administration of the DMKD-PS carrying MAGE-A3 mRNA, no specific histotoxicity was identified in the liver, lungs, heart, kidneys, spleen, and testicles. When the tumor cross-section underwent immunohistochemical staining for MAGE-A3, a reduction in MAGE-A3 expression was observed following inoculation with the DMKD-PS cancer vaccine. In addition, Through ELISA, the levels of IL-4 and IFN- were quantified in both the serum of DMKD-PS immunized mice and the culture media from primarily cultured spleen. While no significant increase in either cytokine was observed in the serum, both cytokines showed a marked elevation in the primary spleen culture media than naïve mice. The results imply that the MAGE-A3 mRNA cancer vaccine, developed with DMKD-PS lipid nanoparticles, holds promising potential for not only preventing tumor development but also treating existing and metastasized tumors, thereby marking a significant breakthrough in cancer therapeutics.
