Although the immune-enhancing activity of plants in the genus Hibiscus has been reported, there are no studies on the immune-enhancing activity of Hibiscus syriacus, except for its roots. Therefore, in this study, we investigated the immune-enhancing activity of Hibiscus leaves (HSL) in RAW264.7 cells, mouse macrophages, and immunosuppressed mice. HSL increased immunostimulatory factors such as nitric oxide (NO), inducible nitric oxide synthase (iNOS), interleukin-1β (IL-1β), and tumor necrosis factor-α (TNF-α) and activated phagocytosis in RAW264.7 cells. The production of immunostimulatory factors by HSL was activated by toll-like receptor 4 (TLR4) in RAW264.7 cells, and the signal transduction pathway is dependent on p38, JNK. Furthermore, HSL increased the spleen index, cytokine levels, and lymphocyte, neutrophil, and monocyte counts in mice in an immunosuppressive mouse model. In addition, autophagy is typically thought of only as cell recycling, macrophage autophagy also contributes to strengthening the innate and adaptive immune system against foreign pathogens. Therefore, macrophage autophagy has been utilized as a key marker in the development of vaccine adjuvants to increase immune response. In this study, HSL activated the p38, JNK, and NF-κB signaling pathways while increasing the expression of the autophagosomal cargo protein p62/SQSTM1. Inhibition of TLR4 and the p38, JNK, and NF-κB signaling pathways blocked the HSL-induced increase in p62/SQSTM1. Furthermore, HSL increased the expression of Nrf2, and inhibiting TLR4, p38, JNK, and NF-κB pathways suppressed Nrf2 expression. In conclusion, HSL-induced autophagic activity is mediated by activation of p62/SQSTM1 and Nrf2 through the TLR4 receptor and p38, JNK, and NF-κB signaling pathways on macrophages. These results suggest that HSL has immunostimulatory properties and could potentially be utilized in the development of immune enhancers or vaccine adjuvants.
