The measurement of antioxidant activity and analysis of the components of spent coffee ground was conducted. Through in vitro experimants, the mechanism for lipogenesis and adipogenesis inhibition were evaluated to examine the possibility as a functional material. As a result of measuring the antioxidant activity through total polyphenol content and DPPH radical scavenging assay, it was found that spent coffee ground(SCG) had higher antioxidant activity than fermented spent coffee ground(FSCG) The analysis of chlorogenic acid and caffeic acid to confirm the change in the composition of SCG. FSCG indicated that SCG had a higher content of chlorogenic acid and lower content of caffeic acid than FSCG. It is thought that the chlorogenic acid was decomposed into caffeic acid through fermentation. As a result of MTT assay in 3T3-L1 cell, both SCG and FSCG showed no significant difference compared to the control group showing no toxicity. In subsequent cell experiments, SCG showed ability to inhibit fat accumulation through oil red o staining. Additionally, SCG was found to inhibit differentiation, accumulation, and synthesis of lipid through RT-PCR and western blot. As a result of TLC analysis to confirm the change in composition between SCG and FSCG, it was found that SCG had a higher content of chlorogenic acid and a lower content of caffeic acid than FSCG.
