In this study, Saccharomyces cerevisiae was engineered to produce heme because it is widely used in the food industry due to its GRAS (generally recognized as safe) status. In S. cerevisiae the heme production pathway is divided into two parts: (1) the cytoplasmic pathway consisting of HEM2, HEM3, HEM4, HEM12, and HEM13 enzymes and (2) the mitochondrial pathway consisting of HEM1, HEM13, HEM14, and HEM15. The heme production efficiency in S. cerevisiae is low because the heme biosynthesis pathway in S. cerevisiae is divided into two compartments. To overcome this limitation, this study aimed to fractionate the all heme biosynthesis pathways into mitochondria. The engineered S. cerevisiae with mitochondria compartmentation of the heme biosynthetic pathway SKSK369produced 4.6 mg/L heme in 72 h. This value was 48% higher than that produced by the control strain overexpressing the same enzymes without mitochondria compartmentation. Two main pathways for the biosynthesis of heme are protoporphyrindependent (PPD) and coproporphyrin-dependent (CPD) pathways. In S.cerevisiae, heme is synthesized via the PPD pathway. However, it was previously reported that the CPD pathway is thermodynamically more favorable compared with the PPD pathway. Accordingly, this study sought to enhance heme production in S. cerevisiae by introducing the CPD pathway into S. cerevisiae. To do so, hemQ gene encoding copro heme derived from Corynebacterium glutamicum or Bacillus subtillis were introduced into the SKSC339 strain. Of two different hemQs, the introduction of hemQ from C. glutamicum resulted in the highest production titer of heme (6.78 mg/L), which was 65% higher than that produced by the SKCK339 strain without the CPD pathway.
