LOD와 LOQ는 각각 32.57 µg/g, 98.68 µg/g, 정확도는 99~104 %, 반복성과 중간정밀성은 각각 100.4~102.2 %, 100.4~103.6 %로 나타났다. 두 개의 레시틴 샘플에 대한 측정 불확도를 추정한 결과, 대두 레시틴의 경우 (120.1 ± 12.0) mg/g, 난황 레시틴의 경우 (406.6 ± 37.0) mg/g였으며 모두 CODEX 기준을 충족하였다. 분석법 비교를 위해 HPLC-RID 시험법 검증을 수행한 결과, 크로마토그램에서 특이성을 확보하였고, 직선성은 1 000~10 000 µg/mL에서 대두 PC와 난황 PC 각각 R2 > 0.998 4, R2 > 0.997 9였다. The samples were chosen soy lecithin, which is a vegetable, and egg yolk lecithin, which is an animal, depending on the raw materials as functional foods. Phosphatidylcholine was extracted using a chloroform and methanol (5:5, v/v) solution and then analyzed with gradient elution consisting of chloroform, chloroform: methanol (7:3, v/v), and chloroform: methanol: water: ammonia (4.5:4.5:0.95:0.05, v/v) through the analytical silica column by HPLC with Evaporative Light-Scattering Detector. The flow rate was 0.8 mL/min, injection volume was 10 L. The evaporator temperature and nebulizer of the ELS detector were set at 40 °C, and the gas flow was 60 - 100 psi. For the method verification for PC (from soy), the specificity was satisfied, and linearity for the calibration curve was 25 – 500 µg/mL, and R2 was 0.994 9. The LOD and LOQ were 22.64 µg/g, and 68.60 µg/g respectively. The average recovery rate for accuracy was 98 ~104 %, repeatability, and inter-precision were 98.1 ~ 104.0 %, and 99.3 ~ 104.3 % respectively. For the method verification for PC (from egg yolk), the specificity was satisfied as well, and the linearity for the calibration curve was 50 – 1000 µg/mL, and R2 was 0.997 0. LOD and LOQ were 32.57 µg/g, and 98.68 µg/g respectively. The average recovery rate for accuracy was 99 ~ 104 %, repeatability, and inter-precision were 100.4 ~ 102.2 % and 100.4 ~ 103.6 %. The estimated measurement uncertainty for two lecithin samples was (120.1 ± 12.0) mg/g for soy lecithin, and (406.6 ± 37.0) mg/g for egg lecithin which all met the CODEX standard. The method validation of HPLC-RID for comparison to the ELS detector was conducted. The calibration curve was 1000 ~ 10 000 µg/mL, and the R2 for linearity was 0.998 4, and 0.997 9 for each soy and egg yolk respectively. LOD and LOQ were 226.31, 685.78 µg/g for PC from soy, and 112.40, 340.61 µg/g for PC from egg yolk. Accuracy was 99 ~103 % (soy PC), and 99 ~ 104 % (egg yolk PC). The repeatability and intermediate precision were 100.3 % - 102.8 % (soy PC), and 100.4 % - 101.7 % (egg yolk PC). The result compared to the two HPLC analysis methods to determine the phosphatidylcholine, through the ELS detector was able to obtain concentration values in which the detection limit and quantitation limit were lower by about 3 - 10 %. Ultimately, this study has shown to detect lower concentrations significantly than the RI detector and increase the selectivity of the chromatogram when the HPLC-ELS detector was used. Thus, it was expected to be expanded to quantify other components of phospholipids in the future in this method as much as the test specifications for phosphatidylcholine among lecithin are diverse.
