Prion은 양의 scrapie, 소의 bovine spongiform encephalopathy와 사람의 CJD와 같은 다양한 신경 퇴행성 질환을 유발시키는 단백질 병원체이다. 정상 prion 단백질인 PrPC가 병원성 PrPSc로 바뀌는 과정에 대해서는 많은 연구가 진행되었고, PrPSc로의 단백질 구조 변화가 다양한 환경적 요소에 의해서 영향 받는 것으로 추측된다. 바다조류로부터 분리된 MAAs는 다양한 스트레스 환경에서 조류를 보호해주는 것으로 알려져 있다. 이와 같은 사실에 기초하여 mycosporineglycine, porphyra-334와 shinorine 3종의 MAAs로 처리한 prion 감염 신경세포 주에서 prion 단백질 축적의 변화를 평가하였다. Although the detailed process, regarding the abnormal conversion of prion proteins (PrP), remains to be fully elucidated, a number of environmental factors appear to affect the formation of misfolded PrP, termed PrPSc. Because oceanic algae contain mycosporine-like amino acids (MAAs), which exhibit cellular defensive activities under a variety of stress conditions, we investigated the level of PrPSc in prion-infected neuroblastoma cells using mycosporine-glycine, porphyra-334 and shinorine. 본 연구에서는 자외선에 의해서 발생되는 세포 내 손상을 억제하는데 있어 매우 우수한 효과를 보이는 세 종류의 MAA인, mycosporine-glycine, porphyra-334와 shinorine을 이용하여 prion의 생화학적 요체인 PrPSc의 축적에 대한 변화를 prion이 영구 감염된 neuroblastoma 세포 주에서 확인하였다.
제안 방법
5% CO2와 포화습도 상태의 세포배양기조건에서 세포배양을 실시하였으며, 세포배양 후, 90%정도의 confluent state에서 세포배양 plate로부터 각각 5%의 세포를 새로운 세포배양 plate로 옮겼다. 세포가 plate표면에 부착된 후, 여러 농도의 MAA (0.1, 0.3, 1, 3, 10 μM)를 세포 배양액에 첨가하였다. 3일에 한번씩 동일 MAA가 포함된 세포배양액으로 교체하였으며, 총 7일간 배양 후에 세포파쇄액을 수집하였다.

7일간의 배양 후, MAA 분리를 위해 C. hedleyi를 수집하였다.
이론/모형
PrPSc의 축적을 확인하기 위하여 western blot을 이용하였다. 7일간 MAA 처리된 세포를 lysis buffer (20 mM Tris, pH 8.
성능/효과
Mycosporine-glycine은 고리 구조를 가지고 있는데, 이를 통해 빛을 흡수함으로써 강력한 빛에 의한 해양 생물의 세포 내 미토콘드리아의 손상 혹은 세포를 둘러싸고 있는 지질 막의 산화를 억제하는 물질로 알려져 있다[23]. Prion 질병에 있어, 세포에서 PrPC가 PrPSc로 전환되는 과정에 mycosporine-glycine의 영향을 확인하기 위해서 여러 농도의 mycosporine-glycine를 처리한 결과, prion 감염된 ScN2a 세포에서 PrPSc 단백질의 감소는 확인되지 않았으며, 대조구에 비교하였을 때 PrPSc 수준은 변화하지 않았다(Fig. 2).
동시에 피부세포의 노화를 유도하는 활성산소인 reactive oxygen species (ROS)를 제거함과 동시에 matrix metalloproteinase (MMP)과 extracellular matrix (ECM)의 발현을 촉진시킴에 따라 피부의 노화를 억제하는 것으로 알려져 있다[21]. Mycosporine-glycinec 처리 조건과 동일하게 ScN2a 세포에 porphyra-334를 처리한 결과 세포에서 PrPC가 PrPSc로 변환하는 작용이 촉진되는 것이 관찰되었다(Fig. 3).

이와 같은 MAA 처리에 있어 prion 질병의 원인인 PrPSc가 MAA에 의한 세포 내 환경 변화에 따라 응집이 증가되는 것이 관찰되는데, 이는 여러 이유에서 기인할 것으로 추론된다. 그 중 하나는 MAA의 경우 prion 단백질이 PrPC에서 PrPSc로 전환하는데 있어 직접적으로 전혀 영향을 미치지 않는 것으로 사료된다.

또한 MAA를 처리 할 경우, PrPSc 축적에 의해 발생되는, 세포 내 활성 산소가 MAA의 존재로 인해 감소될 것으로 추측되며, 이 같은 결과는 MAA가 존재하지 않은 경우에 비해, PrPC 전사와 번역이 상대적으로 활성화 되어, PrPSc의 기질인 PrPC의 양이 상대적으로 동일 세포 내 많이 존재할 수 있게 한다. 따라서 이로 인해 PrPSc에 의한 PrPC의 변환이 MAA를 처리하지 않은 세포에 비해 상대적으로 많이 진행된 것으로 사료되며, 이로 인해 MAA를 처리한 세포에서 PrPSc 축적이 보다 더 활성화 되는 것으로 추측된다. 실제 세포 내PrPC의 발현이 감소되면, PrPSc의 증식이 감소되면서 prion 질병 발병 시기를 늦추는 것으로 확인되었다[5].
이전 연구에서 다양한 아미노산 화합물을 이용하여 ScN2a를 처리할 경우, plasminogen과 아미노산 화합물이 결합하면서 PrPC의 PrPSc 변환을 억제하는 것으로 관찰되는데, 이러한 화합물의 경우 일반적으로 10개 미만의 단량체에서는 효과가 최소화되는 반면에, 30개 이상으로 구성된 화합물에서 prion억제 활성을 보이는 것으로 보고되었다[20]. 따라서 현재 사용된 단량체 MAA의 경우 여러 개의 단량체가 중합되면, plasminogen 활성의 억제를 통해, PrPSc 형성을 억제함으로써, 항 prion 효과를 나타낼 수 있을 것으로 추측되며, prion 치료제로서의 가능성도 타진 할 수 있을 것이다.
흥미롭게도 처리된 MAA에 따라 PrPSc의 증식에 대한 효과가 다르게 나타났으며, porphyra-334에 의한 PrPSc의 증식이 가장 활발히 진행된 것으로 확인되었다(Fig. 2).
그러므로 shinorine과 porphyra-334는 mycosporine-glycine에는 존재하지 않는 serine 또는 이의 유도체가 존재한다. PrPSc의 축적이 향상되는 현상은 shinorine과 porphyra-334 처리시 나타났으며, 유사현상은 mycosporine-glycine 처리시에는 최소화 되었다. 그러므로 shinorine과 porphyra-334에 공통으로 존재하는 serine 또는 이의 유도체 첨가생성물이 PrPSc 축적에 영향을 미치며, 특히 porphyra-334의 serine 부분에 존재하는 메칠기가 PrPSc 축적에 보다 효과적으로 작용한다 고 사료된다.

Prion 질병에서 PrPSc 축적 역시 세포 내 산화적 스트레스를 발생시키는 직접적인 원인이 되며, 증가된 활성 산소에 의해 세포 사멸이 진행되는 것으로 보고되고 있다[26]. 본 실험에서는 MAA의 처리 시, prion 단백질 축적이 오히려 세포 내에서 증가하는 것이 관찰되었다. 그러나 MAAs 처리에 의한 PrPSc 축적 결과는, 오히려 MAA가 활성산소를 제거함으로써 이로 인한 세포 내 산화적 스트레스를 억제 한 결과로도 해석이 가능하다.
본 실험에서는 MAA의 처리 시, prion 단백질 축적이 오히려 세포 내에서 증가하는 것이 관찰되었다. 그러나 MAAs 처리에 의한 PrPSc 축적 결과는, 오히려 MAA가 활성산소를 제거함으로써 이로 인한 세포 내 산화적 스트레스를 억제 한 결과로도 해석이 가능하다. 따라서, 세포 내에서 prion 단백질 축적 관 련 세포사멸 억제에 대한 MAA의 효과는 독립적인 연구에서 수행될 것으로 기대된다.
후속연구
그러나 MAAs 처리에 의한 PrPSc 축적 결과는, 오히려 MAA가 활성산소를 제거함으로써 이로 인한 세포 내 산화적 스트레스를 억제 한 결과로도 해석이 가능하다. 따라서, 세포 내에서 prion 단백질 축적 관 련 세포사멸 억제에 대한 MAA의 효과는 독립적인 연구에서 수행될 것으로 기대된다.
