천연물 처리 여부에 따른 세포에서 분비되는 Nitric oxide (NO)의 생산량의 차이를 측정하기 위해서 96well plate에 BV2 세포를 well 당 1.0×104개를 분주하고 24시간 뒤 라이브러리 상의 천연물을 2.5 μg/ml가 되도록 처리하였다. 1시간 뒤 Lipopolysaccharide (LPS, 1 μg/ml)를 처리한 뒤 24시간 배양 후 각각을 상등액 100 μl를 원심분리(700g, 5min)하여 부유한 세포를 제거한 이후 새로운 96well plate에 옮겨 분주하였다.
각 well의 배지에 NO detection kit (iNtRon, Seoul, Korea)의 N1 buffer를 각 well에 50 μl씩 첨가 후 실온에서 10분 동안 반응 후 50 μl의 N2 buffer를 첨가하고 다시 10분간 반응시킨 후 550 nm에서 흡광도를 측정하였다. 측정한 값은 동일한 실험에서 수행한 NO의 표준 정량곡선과 비교하여 배지에 포함된 NO의 양을 확인하였다.
대상 데이터
본 실험에 사용된 천연물 라이브러리는 한국한방산업진흥원(KOTMIN)에서 제공받았으며, 총 240개의 물질을 dimethyl sulfoxide (DMSO)를 용매로 하여 500 μg/ml의 농도로 제공되었으며, 각 세포에 적용된 최종 농도는 2.5 μg/ml로 처리되었다.
본 연구에 사용된 천연물은 한국한방산업진흥원에서 분양받아 사용하였으며, 한국연구재단 이공분야 기초연구사업 (NRF-2014R1A2A1A11054095) 및 한국생명공학연구원 주요 사업(KGM4611411)의 지원에 의해 수행되었음.

5μg/ml의 농도의 천연물을 각각 처리한 후 glutamate를 처리하여 그 변화를 MTT assay를 통하여 확인하였다. 그 결과 glutamate 단독 처리군의 세포 생존율은 44.30±11.36%로 나타났으며(Fig. 1.) glutamate 처리에도 불구하고 85%이상의 생존율을 나타낸 천연물은 고삼에서 추출한 (-)-Maackiain (90.9±4.7%), 천근초에서 추출한 1, 3, 6-trihy- droxy-2-methyl-9, 10-anthraquinon (87.5±5.7%), 적하수오에서 추출한 2, 3, 5, 4'-Tetrahydroxystilbene-2-O-β-D-glucoside (89.9±9.5%), 승마에서 추출한 27-deoxyactein (94.3±3.8%), 고금에서 추출한 Baicalein (96.5±7.9%), 선복화에서 추출한 Bri- tanin (89.0±7.0%), 대황에서 추출한 Chrysophanol (95.7± 4.0%), 오미자에서 추출한 Gomisin J (89.9±7.4%), 강진 향에서 추출한 Latifolin (99.9±11.8%), 후박에서 추출한 Magnolol (94.9±1.1%), 수련에서 추출한 Nuciferine (99.8±12.3%), 현호색으로부터 추출한 Tetrahydrocoptisine (94.0±10.2%)으로 나타났으며 이는 Fig. 1과 Table. 1에 정리하였다.

2). 또한, 세포 생존율 측정(MTT assay) 한결과에서는 비처리 대조군 세포의 생존율(100±2.5%)에 비해 LPS 단독 처리군의 생존율은 72.31±6.4%로 나타났다(Fig. 3.). 이러한 차이는 LPS에 의한 자극이 BV2세포에서 정상적으로 유도된 것으로 판단되며, LPS 단독처리군의 생존율과 천연물을 처리한 군의 비교를 통해 처리한 천연물의 세포독성이 없음을 확인하였고, NO의 분비량을 감소하게 한 22개의 물질 중 세포의 생존율의 변화가 LPS 단독 처리군과 비교하여 감소치가 20% 미만인 물질은 총 13가지 물질이 검출되었으며, 이는 우방자에서 추출한 Arctiin, 울금에서 추출한 Bisdemethox- ycurcumin, 대황에서 추출한 Chrysophanol, 산초나무잎에서 추출한 Collinin, 슬모초에서 추출한 Imperatorin, 보골지에서추출한 Isopsoralen, 감초에 추출한 Licoricidin과 Lindenenyl acetate, 삼백초의 뿌리에서 추출한 Manassantin A와 Manas- santin B, 고금에서 추출한 Wogonin, 신선초에서 추출한 4- Hydroxy-3-methoxycinnamal dehyde, 천심련에서 추출한 andrographolide이며, 이는 Table.
본 연구결과를 종합하여 보면 천연물 라이브러리를 이용하여 신경세포와 미세아교세포에서 glutamate와 LPS에 의한 신경세포 사멸과 뇌염증을 모두 억제할 수 있는 물질을 검색한 결과 대황에서 추출한 Chrysophanol이 검출되었으며, 더욱이 검출된 Chrysophanol은 각 세포에서 glutamate와 LPS에 의한 산화적 스트레스로 과도한 ROS 생성을 제거하는 것으로 판단된다.
후속연구
이러한 결과를 바탕으로 Chrysophanol에 대한 보다 깊은 추가 연구를 통하여 산화적 스트레스 유도 신경세포 사멸과 뇌의 면역을 담당하는 미세아교세포의 과잉 활성화에 따른 뇌 염증을 동시에 억제하는 약물로의 개발을 앞으로 하고자 한다.
