본 연구에서는 C. pubescens Burkill 에탄올 추출물(CPEE)의 항산화능과 항염증 생리활성을 DPPH 라디칼 소거능, ROS 생성 억제능, NO 소거 활성 등을 통해 분석하였다. 먼저 CPEE의 항산화능을 DPPH 라디칼 소거능으로 분석한 결과 강한 소거능을 보였으며, 좀 더 자세한 항산화능 작용기작을 알아보기 위해 ROS 생성 억제능으로 분석한 결과 농도 의존적으로 강한 ROS 소거능을 보였다. 뿐만 아니라 대표적인 항산화 효소인 HO-1 및 그 전사 인자인 Nrf2의 단백질 발현에 미치는 영향을 분석한 결과 CPEE에 의해 HO-1 및 Nrf2의 발현이 증가됨을 보였다. 한편 CPEE가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 농도의 존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이러한 결과를 통해 CPEE의 높은 항산화능과 항염증 활성을 확인하였으며 향후 잠재적인 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다.

Furthermore, CPEE induced the expression of the anti-oxidative enzyme heme oxygenase 1 and its upstream transcription factor, nuclear factor-E2-related factor 2, in RAW 264.7 cells. CPEE was associated with a reduction in NO production, which was induced by lipopolysaccharide treatment in a dose-dependent manner. The expression of inducible nitric oxide synthase (iNOS), an upstream regulator of NO production, was also inhibited. Taken together, these results suggest that CPEE has anti-oxidative and anti-inflammatory activities and could be useful as a potential anti-oxidant and antiinflammatory agent.

이에 본 연구에서는 천연에서 유래한 생리활성 보유 신소재 개발의 일환으로 C. pubescens Burkill의 에탄올 추출물이 보유한 항산화 및 항염증 활성을 시험관 분석법과 세포 실험 모델계를 이용하여 분석함으로써 기능성 소재로서의 활용 가능성을 확인해 보고자 하였다.
제안 방법

항산화 물질의 가장 특징적인 기작은 유리기와 반응하는것으로 라디칼 소거 작용은 활성라디칼에 전자를 공여하여 항산화 효과나 인체 노화를 억제하는 척도로 이용되고 있다[1]. CPEE의 항산화능을 DPPH 라디칼 소거 활성을 통해 분석하였다. 그 결과 CPEE의 농도 증가에 따라 강한 라디칼 소거능을 보여 0.
DPPH 라디칼 소거 활성에 의해 CPEE가 보유한 높은 항산화능이 확인됨에 따라 그 작용 기전을 좀 더 자세히 알아보기 위해 먼저 RAW 264.7 세포주에 대표적인 산화적 스트레스 유도인자인 H2O2를 처리하여 CPEE에 의한 ROS 소거능을 분석하였다. 세포 내에서 활성산소가 발생되면 DCF-DA가 에스터라제 또는 산화적 가수분해에 DCFH로 탈아세틸화 되고 비형광성인 DCFH는 활성 산소에 의해 산화되어 2',7'-dichloro-fluorescein (DCF)로 전환되고 이는 강한 형광을 나타낸다[23].
CPEE가 강한 항산화 활성을 보유하고 있음이 상기의 실험을 통해 밝혀졌다. 따라서 CPEE가 항염증 활성 또한 나타내는지 알아보기 위해 NO 생성 저해능을 분석하였다. 먼저 CPEE가 LPS로 자극을 유도한 RAW 264.
H2O2는 대표적인 ROS 중 하나로 소재의 항산화능을 규명하기 위한 많은 연구에서 ROS 유도제로 사용되고 있다[28, 30, 33]. 본 연구에서는 CPEE가 보유한 항산화능을 H2O2로 유도한 ROS 생성에 시료가 미치는 영향을 통해 분석하였다. 이를 위해 RAW 264.
반응성 질소종(reactive nitrogen species)의 하나이며, 최근 염증반응의 중요한 작용인자로 알려진 NO는 신경독성, 신호전달 및 체내방어 등의 생리기능을 갖고 있으며, 염증과 암 발생에 관여해 병리적으로 중요한 역할을 한다[34, 36]. 이러한 NO 생성 억제능의 분석은 Park 등[28]의 방법을 변형하여 수행하였다. RAW 264.
활성 분석 수행 전 시료가 지닌 세포독성의 유무 확인과 함께 이후 실험 농도의 결정을 위해 RAW 264.7 세포주를 24-well tissue culture plate에 well 당 3.0 × 105개씩 분주하여 부착시킨 후 CPEE에 의한 세포 독성 유발 유무를 WST assay를 통해 분석하였다. CPEE 처리 24시간 후 WST 시약(Daeil Lab Service, South Korea)이 든 배지로 교체하여 한 시간 동안 반응시키고 multi-plate reader를 이용하여 450 nm에서 흡광도를 측정하였다.
대상 데이터
본 연구에서 사용한 C. pubescens 에탄올 추출물(이하 CPEE)은 한국생명공학연구원 해외생물소재허브센터에서 구입(분양번호 FBM123-019)하여 사용하였으며 추출과정은 다음과 같다. 건조 및 분쇄한 CPEE를 95% 에탄올을 이용하여 45℃에서 15분간 초음파 추출 후 2시간 동안 정치시키는 과정을 하루 10회 반복하여 총 3일간 추출을 수행한다.
항산화 및 항염증 활성의 세포 실험 모델계로 쥐 대식세포주인 RAW 264.7을 American Type Culture Collection (ATCC, TIB-71TM, VA, USA)로부터 구입하여 10% 우태아 혈청(fetal bovine serum, FBS, Invitrogen Corporation, CA, USA)과 1% 페니실린/스트렙토마이신(Invitrogen)이 포함된 DMEM (Invitrogen) 배지를 사용하여 37℃, 5% CO2조건 하에서 배양하였다.
이론/모형
CPEE의 항산화 활성 기전을 알아보기 위해 대표적인 항산화 효소인 HO-1과 그 전사인자인 Nrf2의 시료 처리에 의한 단백질 발현 변화를 Western blot hybridization으로 분석하였다. HO-1의 일차항체는 Cell Signaling Technology (MA, USA)로부터 구입하였고, Nrf2와 actin의 일차항체와 anti-goat와 anti-rabbit 등의 이차항체는 Santa Cruz Biotechnology (CA, USA)에서 구입하여 사용하였다.
iNOS는 평소에는 세포 내에 존재하지 않으나 일단 유도되면 장시간 동안 다량의 NO를 생성하며, 생성된 NO는 부종 등의 염증반응을 촉진시키고 염증 매개체의 생합성을 촉진하여 염증을 심화시키는 것으로 알려져 있다[17, 40]. CPEE의 항염증 활성 기전을 밝히기 위해 NO 생성의 핵심 단백질인 iNOS의 단백질 발현을 Western blot hybridization으로 분석하였다. iNOS의 일차항체는 Cell Signaling Technology (MA, USA)로부터 구입하였고, Actin의 일차항체와 anti-goat와 anti-rabbit 등의 이차항체는 Santa Cruz Biotechnology (CA, USA)에서 구입하여 사용하였다.
정상적인 상태에서 Nrf2는 Keap1에 의해 비활성화 상태로 세포질에 존재하지만, 산화적 자극을 받으면 Keap1과 해리되어 핵 내로 이동하여 ARE에 결합함으로써 HO-1과 같은 항산화 효소의 발현을 조절한다[27]. 상기 실험에서 CPEE에 의해 ROS가 감소되는 것이 확인됨에 따라 CPEE가 보유한 항산화능의 작용기작을 알아보기 위해 HO-1 및 Nrf2의 발현 정도를 Western blot hybridization을 통해 분석하였다. 그 결과 10−50 μg/ml의 시료 처리에 의해 HO-1 단백질 발현이 증가되었으며 상위전사 인자인 Nrf2의 단백질 발현 또한 증가되었다(Fig.
성능/효과
7 세포 생존율에 미치는 영향을 살펴보았다. CPEE를 농도별(0, 10, 25, 50 μg/ml)로 24시간 처리한 결과, 모든 농도에서 세포 독성을 유발하지 않는 것을 확인하였다(Fig. 1).
상기 실험에서 CPEE에 의해 ROS가 감소되는 것이 확인됨에 따라 CPEE가 보유한 항산화능의 작용기작을 알아보기 위해 HO-1 및 Nrf2의 발현 정도를 Western blot hybridization을 통해 분석하였다. 그 결과 10−50 μg/ml의 시료 처리에 의해 HO-1 단백질 발현이 증가되었으며 상위전사 인자인 Nrf2의 단백질 발현 또한 증가되었다(Fig. 3).
CPEE의 항산화능을 DPPH 라디칼 소거 활성을 통해 분석하였다. 그 결과 CPEE의 농도 증가에 따라 강한 라디칼 소거능을 보여 0.1024, 0.512, 2.56, 12.8 μg/ml의 시료 처리에 의해 DPPH 라디칼 소거능이 각각 21.85, 37.04, 86.99, 99.85%로 나타나 50% 소거 농도를 나타내는 IC50 값이 1.04 μg/ml로 양성 대조군으로 사용한 아스코르브산, 즉 비타민 C의 IC50 값인 1.19 μg/ml과 유사한 정도의 높은 활성을 보여 매우 강한 항산화능을 보유함을 확인하였다(Table 1).
4A). 다음으로 LPS로 자극을 유도한 RAW 264.7 세포주에서 농도별 CPEE의 처리에 따른 NO 생성과 iNOS 발현에 미치는 영향을 분석한 결과 10−200 μg/ml의 시료 처리에 의해 농도의존적인 NO 생성 저해활성과 함께 iNOS 발현이 저해됨을 확인하였다(Fig. 4B, 4C).
3). 따라서 CPEE는 RAW 264.7 세포주에서 Nrf2를 전사시켜 HO-1 단백질 발현을 유도함으로써 산화적 스트레스로부터 항산화 활성을 나타낸다는 사실을 확인하였다.
