Objectives This experimental study was designed to investigate the effect of Phaseolus angularis shell on metabolic syndrome. Methods Each 5 C57BL/6J mice were randomly assigned to normal diet group, high-fat diet(HFD) control group, high-fat diet plus 15.6 mg/kg/day of Orlistat(HFD-Orlistat) group, high-fat diet plus 100mg/kg/day of Phaseolus angularis shell extract(HFD-PAS_E) group. Weight, the blood chemical and hematologic parameter was med. The mRNA expression was assayed through Reverse transcriptase polymerase chain reaction(RT-PCR). Results In HFD-PAS_E group, the body weight gain, weight of liver, and the level of LDL-Cholesterol were significantly decreased and the level of HDL-Cholesterol were significantly increased. The size of adipocyte in HFD-PAS_E group was smaller than HFD group's. In HFD-PAS_E group, the expression of leptin, PPAR-${\gamma}$, AP2/FABP4 mRNA in liver adipocyte tissue was decreased, the expression of Adiponectin, UCP-2 mRNA in liver adipocyte tissue was increased and the expression of Leptin, C/EBP-a, AP2/FABP4 mRNA in epididymal adipocyte tissue was decreased. Conclusion These results suggest that Phaseolus angularis shell has inhibitory effects on metabolic syndrome by reducing the body weight and the levels of lipid contents in high-fat-diet induced obese mice.

적소두 외피의 간독성을 알아보기 위해 혈중 AST, ALT, ALP, LDH를 측정하였다. AST는 심근, 간, 골격근, 신장에 분포하고, ALT는 간, 신장, 심근, 골격근에 분포하며 혈중에는 미량만 존재한다.
적소두 외피의 신독성을 알아보기 위해 혈중 Uric acid와 함께 신장 기능의 지표가 되는 Creatinine의 농도를 측정하였다. Uric acid는 질소의 최종 대사산물로신장으로 이동하여 소변으로 배출된다.

대상 데이터

C57bl/6 마우스 수컷을 대한바이오링크 (Eumseong, Korea)로부터 분양하여 사용하였다. 분양 받은 6주령 실험동물은 2주동안 물과 기본사료(AIN-76A diet)를 자유롭게 공급하여 실험실 환경에 적응시킨 후, 약 23.
본 실험에서 사용한 팥껍질(Phaseolus angularis shell extract, 이하 PAS_E)은 대표적 한약공급업체인 옴니허브 (http://www.omniherb.com. Korea)에서 구입하였으며, 200g 정량 후 1.5ℓ의 80%에탄올에 30분 동안 sonication 하였다. 3mm 여과지 (Whatman, Maidstone, England)를 이용하여 감압 여과한 후, 여과된 에탄올 추출물은 감압농축기(Eyela, Japan)를 이용하여 농축한 후, 동결건조(Freezedryer, Matsushita, Japan)하여 얻은 분말을 70℃ 초저온냉동고에 보관 사용하였다.

데이터처리

모든 실험결과는 SPSS 통계 프로그램을 이용하여 분석하였으며 그 결과는 평균 ± 표준오차 (mean±SE)로 표시하였다. 정산군과 대조군간의 유의성은 정상군 (ND)과 비교하였고, 그리고 각 실험군간의 통계적 유의성을 대조군(HFD-CTL)과 비교하여 Student t-test를 통해 P < 0.

본 실험에서 유의성 있는 결과를 종합하여 분석한 결과, 赤小豆外皮 추출물이 간조직의 중량, 복부 피하지방과 창자주변 지방조직의 무게를 유의적으로 감소시키며 체중감소효과가 있었고 신장과 간에 유해하지 않은 것으로 생각되며 혈중 Adiponectin 수치를 높이면서 insulin 저항성을 감소시키고, 민감도를 증가시키는 경향성을 나타내었다. 지방분화 과정과 에너지 소비에 관여하는 간조직내 PPAR-γ, AP2/FABP4, AMPK-α2, UCP2 mRNA 발현과 부고환 조직내 Leptin, C/EBP-α mRNA 발현을 조절하였다.

후속연구

본 연구에서는 비만대조군(HFD군)과 비교하여 실험군(HFD-PAS_E군)의 간 조직내 AMPK α1 mRNA 발현이 증가하였으나 유의성이 없고 AMPK-α2 mRNA 발현은 유의하게 감소하였다. AMPK α1 mRNA, AMPK α2 mRNA의 AMPK mRNA 총량에 대한 기여도 분석이 부족하여 AMPK mRNA 총량이 늘었는지에 대해서는 정확히 알 수 없는 상태로 팥껍질 추출물의 AMPK mRNA 발현에 대한 영향은 추가적인 연구가 필요할 것으로 사료된다.

그러나 동물 모델을 이용하여 사람의 비만을 동반한 대사증후군에 발생하는 다른 유전적 요인을 고려하지 못했다는 측면에서 사람의 비만을 동반한 대사증후군 치료에 직접적으로 반영하기에는 어려움이 있다. 따라서 실제 한방치료 요법 중 하나로 사용되기 전에 비만을 동반한 대사증후군 환자를 대상으로 추가적인 임상연구가 필요할 것으로 사료된다.
