효소의 한계는? | Sumner가 1926년 urease를 작두콩(jack bean)에서 정제 및 결정화하여 효소 단백질의 구조를 이해하기 시작한 이래로[9,10], 효소는 구조적 특성인 활성 자리 (active site)에서 유래한 기질(substrate) 및 생성물(product)에 대한 고도의 선택성과 뛰어난 효율성으로, 생명공학을 포함한 다양한 의학및 산업적 분야에서 폭넓게 이용되어왔다. 그러나 생체 유래 물질인 만큼 발현과 정제 과정이 복잡하여 제조단가가 높고, 생체 물질의 특성상 활성을 보이는 조건이 화학 촉매제에 비해 제한적이며, 외부 환경에 의해 3차원 구조가 변성(denaturation)되기 쉬워 안정성이 낮다는 단점들로 일반적인 유무기 화학 물질에 비교해 한계를 가진다. |
