Methods:Articles and documents published from 1990 through 2007, containing words like ageing mechanism, ageing concept, successful ageing and longevity were searched via internet and reviewed. Results:1) Definition of ageing : biological ageing can be defined as the progressive loss of function to decrease fertility and to increase mortality, according to the increase of age. The WHO insisted active ageing must be devoid of disease and disability. 2) Mechanism of ageing : Various mechanism of ageing such as molecular, cellular, system, evolutionary and molecular inflammatory theories were reported. 3) Update issues on ageing : As the universal ageing mechanism, theories including oxidative damage, synthesis of advanced glycation end-products, change of insulin-IGF axis, and change of energy balance were suggested. At present, results of animal experiments showed that genetic pathways related to the calorie restriction in the yeasts, nematodes, fruit flies and mice might be associated with the control of life span. Otherwise, the theories of ageing can be supported by such clues as the identification of gene abnormality in relation with human progeria, telomere shortening and search for the longevity genes.
