갯벌 내에는 다양한 종류의 미생물들이 서식하며 이들 미생물의 활동은 유기탄소와 금속이온의 순환에 영향을 미치는 것으로 알려져 있다. 이 연구는 순천만 갯벌 내 금속 환원 미생물의 종 다양성을 밝히고, 갯벌 퇴적물로부터 농화배양한 금속 환원 미생물을 이용한 산화환원에 민감한 금속의 환원과 생광물화작용을 연구하는데 목적이 있다. 순천만 갯벌 내에 서식하는 금속 환원 미생물의 종은 16S rRNA분석을 통해 확인하였고, 미생물에 의한 금속 환원은 3가 철을 포함한 akaganeite (β-FeOOH), 6가 크롬(potassium chromate, K2CrO4그리고 6가 셀레늄(sodium selenate, Na2SeO4)을 전자수용체로 이용하여 연구하였다. 미생물에 의한 생광물화작용을 통해 형성된 광물과 금속 환원 작용은 XRD, TEM-EDX, UV분석을 통해 확인되었다. 연구 결과, 순천만 갯벌 퇴적물 내에는 금속 환원 미생물로 잘 알려진 Shewanella sp., Clostridium sp., Vibrio sp. 등이 존재하는 것으로 확인되었고, 이러한 미생물은 전자수용체로 이용된 akaganeite, 6가 크롬 그리고 6가 셀레늄을 모두 환원시켰다. UV분석 결과, 미생물 생체량이 증가함에 따라 Cr(Ⅵ)농도가 현저하게 감소함으로써 미생물 활동에 의한 6가 크롬에서 3가 크롬으로의 환원작용이 확인되었다. 또한 육안관찰, XRD, TEM-EDX 분석결과, 갯벌 금속 환원 미생물은 유기물(glucose)의 산화와 더불어 akaganeite를 환원시켜 자철석을 형성하며 6가 셀레늄을 환원시켜 셀레늄 금속을 침전시켰다. 따라서 갯벌에는 다양한 종류의 금속 환원 미생물이 서식하며, 이들 금속 환원 미생물은 금속이온을 환원시켜 탄소와 금속의 생지화학적 순환에 영향을 줄 뿐만 아니라 이러한 과정을 통해 광물 합성 및 중금속 오염 정화에 중요한 역할을 하는 것으로 사료된다.

The objective of this study was to investigate microbial diversity and metal reduction as well as biomineralization by the metal-reducing bacteria from inter tidal flat sediments of Suncheon Bay, Jeonnam province, Korea. 16S rRNA analysis was used to identify microbial diversity of the metal-reducing bacteria. Metal reduction and biomineralization experiments using the metal-reducing bacteria enriched from the intertidal flat sediments were conducted using akaganeite (β-FeOOH), potassium chromate (K2CrO4) and sodium selenate (Na2SeO4) as electron acceptors and glucose as an electron donor. XRD and TEM-EDX analyses were used for mineralogical, geochemical, and morphological characteristics of the formed minerals and precipitated metals. UV-vis analysis was used to figure out the correlation between changes of microbial biomass and Cr(Ⅵ) concentrations. The metal-reducing bacteria enriched from intertidal flat sediments of Suncheon Bay mainly consisted of Shewanella sp., Clostridium sp., and Vibrio sp. These bacteria reduced the diverse metals such as akaganeite (β-FeOOH) to magnetite (Fe3O4), sodium selenate (Na2SeO4) to elemental selenium (Se0), and potassium chromate (K2CrO4) to chromium hydroxide (Cr(OH)3), with changes of color and turbidity. In case of Cr(Ⅵ) reduction, decreasing of Cr(Ⅵ) concentration was accompanied by increasing of microbial biomass. These results indicate that metal-reducing bacteria in the intertidal flat sediment may play one of important roles for bioremediation of heavy metals and cycles of organic matter and metals in the subsurface environments.
