Lactoferrin은 인체 여러 조직 및 세포에서 분비되는 80KD의 단백질로, 여러 생리활성을 갖고 있다. 이 활성 중 하나가 항균 활성으로 이 반응은 철이 유리된 상태에서 활성이 나타난다. Lactoferrin의 pepsin 가수분해산물인 lactoferricin 은 Lactoferrin에 비해 항균 활성이 더 강한 것으로 보고되어 졌다. 따라서 본 연구에서는 합성 펩타이드 기술과 lactoferricin 의 구조를 이용하여 항균활성은 더 높아지고 용혈활성으로 나타내는 세포독성은 감소시킨 lactoferricin 유래의 유사체 펩타이드를 생산하기 위한 기초적인 연구를 수행하고자 한다. 그리고 곤충, 개구리 및 포유동물을 포함한 자연계에는 그 외의 항균 펩타이드가 널리 분포하고 있다. 이들 항균펩타이드는 곤충의 면역계 또는 숙주방어에 중요한 역할을 담당하는 것으로 알려져 있다. 또한 이들 항균펩타이드들은 여러 개의 염기성 아미노산으로 포함하므로 강한 염기성을 띠고 인지질 이중막에서는 양쪽 친매성을 띠는 공통적 구조적 특정을 가지고 있다. 이들의 펩타이드의 염기성은 표적세포의 세포막과의 결합에 관련되어 있으며, 이들의 양쪽 친매적 성질은 세포막의 막전위의 변화 혹은 구조의 파괴에 의한 세포의 죽음에 이르게 하는 것과 관련되어져 있다. 나방으로부터 분리된 cecropin은 그람-음성균에는 강하지만, 그람-양성균에는 다소 약하다. 개구리의 피층으로부터 분리된 magainin 또는 bombinin(BO)는 그람-음성균 및 그람-양성균 모두에 대하여 다소 약한 활성을 가진다. 벌독인 melittin (ME)은 그람-음성균 및 그람-양성균 모두에 대하여 매우 강한 항균활성을 나타내지만 진핵세포에 대하여 세포독성을 가진다. 자연형의 많은 항균 펩타이드들을 아미노산 치환에 의하여 많은 유사체들을 합성하였으며, 이들 유사체들은 항균활성이 증가함에 따라서 적혈구세포에 대한 용혈활성도 동시에 증가함을 보였다. Cecropin A(CA)와 melittin(ME)의 아미노 말단의 서열을 접합시킨 접합 펩타이드는 이와는 다른 결과를 나타내었다. 이들 접합 펩타이드 중에 cecropin A(1-8)과 melittin(1-12)를 접합시킨 CA(1-8)-ME(1-12), 접합 펩타이드는 CA 보다 강력한 항균활성을 가지며, 적혈구 세포에 대한 낮은 용혈활성을 나타낸다는 것이 알려져 있다. 그러나 본 연구에서 CA(1-8)-ME(1-12)는 용액법(solution method)을 이용한 생리적 조건에서는 적혈구세포에 대한 다소의 용혈활성을 나타내었다. 따라서 CA(1-8)-ME(1-12)보다 약한 용혈활성을 나타내며, 강한 항균, 항암 또는 항진균 활성을 가지는 합성 펩타이드를 얻기위하여, 본 연구에서는 CA의 아미노말단의 서열인 CA(1-8)을 기준으로 한 CA(1-8)-ME(1-12), CA(1-8)-MA(1-12) 및 CA(1-8)-BO(1-12)을 설계하고 합성하였으며, ME의 아미노말단의 서열인 ME(1-12)을 기준으로 한 MA(10-17)-ME(1-12), BO(11-18)-ME(1-12) 및 LF(20-29)-ME(142)을 설계하고 합성하였다. 또한 CA(1-8)-ME(1-12) 및 CA(1-8)-MA(1-12)의 α-helical wheel diagram을 바탕으로 하여 특정부위의 아미노산을 다른 아미노산으로 치환시킨 유사체를 설계하고 합성하였다. 모든 펩타이드는 Fmoc-chemistry를 이용한 고상법에 의하여 합성하였다. 각 합성 펩타이드의 항균 항암 및 항진균 활성은 CA(1-8)-ME(1-12) 및 CA(1-8)-MA(1-12)의 α -helical wheel diagrm을 바탕으로 하여 이들의 유사체 펩타이드를 설계하고 합성하였다. 항생활성은 세균 소세포 폐암세포 및 진균에 대한 증식억제능에 의하여 결정되었다. 또한 합성 펩타이드의 용혈활성은 인간의 적혈구 세포의 lysis에 의하여 측정하였다. 또한 항균 작용의 기작을 규명하기 위하여 인지질로 만든 리포좀을 model system으로 이용하여 인지질막내에 형광성 물질을 포집시키고 항생활성을 갖는 펩타이드를 처리하여 막의 파괴에 따른 형광물질의 방출 정도를 측정하였다. 리포좀의 막 구성 성분에 따라서 그리고 포집된 형광물질의 크기에 따른 방출 정도를 측정함으로써 항균 펩타이드와 인지질막과의 상호작용 및 항균활성을 나타내는 기작을 규명할 수 있을 것으로 기대한다. Ⅲ. 연구개발의 내용 및 범위 1. 소의 lactoferricin 아미노산서열로부터 크기가 작지만 항균 활성이 높아질 것으로 기대되는 lactoferricin 유래 유사체 펩타이드를 고안, 합성, 정제하였다. 2. CA(1-8)-ME(1-12), CA(1-8)-MA(1-12) CA(1-8)-BO(1-12), MA(10-17)-ME(1-12), BO(11-18)-ME(1-12), 및 LF(20-29)-ME(1-12)는 Fmoc-chemistry를 이용한 고상법에 의하여 합성하였다. 3. CA(1-8)-ME(1-12) 및 CA(1-8)-MA(1-12)의 α -helical wheel diagram 및 이들 펩타이드의 일차구조로부터 8종류의 아미노산 치환 유사체들을 설계하고 합성하였다. 4. 합성 펩타이드의 아미노산 조성과 분자량은 아미노산 분석 및 MALDI 정량 분석에 의하여 결정되었다. 5. 항균 활성은 대장균 (E. coli) 과 고초균 (B. subtilis) 에 대한 최소생육저지 농도(MIC) 측정에 의해 조사하고 세포 독성은 사람의 적혈구를 이용한 용혈활성으로 나타내었다. 6. 합성 펩타이드의 항균활성은 그람-음성균 및 그람-양성균에 대한 중식 억제활성은 agar hole method 및 solution method에 의하여 측정되었다. 7. 합성 펩타이드의 항암활성은 소세포 폐암세포(SCLC)에 대한증식 억제활성에 의하여 측정되었다. 8. 합성 펩타이드의 항진균 작용의 기작을 연구하기 위하여 fluorescence activated fIow cytometry 및 confocal laser scanning microscopy를 수행하였다. 9. Melittin Hybrid 합성펩타이드 즉 ME, CA(1-8)ME(1-12) 및 MA(10-17)ME(1-12)가 Fusarium oxysporum의 성장에 미치는 저해효과를 확인하였다. 10. Magainin Hybrid 합성펩타이드가 Aspergillus fumigatus의 성장에 미치는 저해효과를 확인하고 이러한 합성 펩타이드가 (1,3)-β-D-glucan synthase 활성에 미치는 영향을 조사하였다. 11. CA(1-8)ME(1-12)를 주형으로 변형시킨 analogue peptide합성하여 T. beigelii에 대한 항진균성 활성을 증대시키는 펩타이드를 설계하고 작용기작을 조사하였다. 12. 펩타이드의 항균활성과 2차구조와의 상관관계를 알아보기 위하여 anionic lipid를 모사하는 SDS조건에서의 CD spectra를 측정하였다. 13. 펩타이드의 인지질막과의 상호작용과 파괴 능력을 리포좀을 model membrane system으로 이용하여 조사하였다. IV. 연구개발의 결과 1. 모든 합성 펩타이드는 분석용 역상 HPLC의 용출 패턴으로부터 높은 purity를 지님을 확인하였으며, 그들의 정확한 아미노산 조성과 분자량은 아미노산 분석 및 MALDI 질량분석에 의하여 확인하였다. 2. Lactoferricin B (B1)의 최저생육저지 농도는 E. coli 에 대하여 22.5 μg/ml, B. subtilis 에 대해 6.3 μg/ml 값을 나타내었다. B1의 cysteine 잔기를 serine 으로 치환시킨 B2는 항균 활성이 약간 낮게 나타났다. B2에서 C-말단 12 잔기를 제거하고 N-말??데, 이 결파는 전하의 크기가 같아도 side group의 크기가 항균 활성을 나타내는데 중요함을 시사해준다. 세포 독성을 나타내는 용혈활성은 B1, B2에 비해 펩타이드의 크기가 줄어들면서 낮아지는 것으로 나타났다. 3. Lactoferricin 유래 유사체 펩타이드는 세포막뿐만아니라 세포외막을 파괴한다는 사실을 periplasmic 효소인 β-lactamase 가 펩타이드의 농도와 처리시간에 따라 방출되는 양상으로 확인하였다. 4. CA(1-8)-ME(1-12) 뿐만 아니라 CA(1-8)-MA(1-12), CA(1-8)-BO(1-12), MA(10-17)-ME(1-12), LF(20-29)-ME(1-12)는 천연형의 항균 펩타이드 보다 크기가 작음에도 불구하고 agar hole method 을 이용한 인간 적혈구 세포에 있어서 용혈활성을 가지지 않으며, 그람 음성 및 그람 양성균에 대하여 강한 항균활성을 가졌다. 5. CA(1-8)-ME(1-12), CA(1-8)-MA(1-12) 및 이들의 유사체의 16번, 18번, 19번의 아미노산 잔기가 소수성(Leu, Val) , 염기성(Lys), 염기성(Lys)일때 강한 항균 및 항암을 나타내었다. 6. CA(1-8)-MA(1-12)의 12번째의 아미노산인 Ser이 Leu으로 치환된 유사체가 최소한의 적혈구 용혈활성을 가지며, 강한 항균, 항암 및 항진균 활성을 나타내었다. 7. 펩타이드의 양쪽 친매성의 증가는 항균, 항암 및 항진균 활성보다는 적혈구 용혈활성에 보다 큰 영향을 미쳤다. 8. Melittin Hybrid 합성펩타이드가 Fusarium oxysporum의 성장에 미치는 저해효과를 확인한 결과, CA(1-8)ME(1-l2) 및 MA(10-17)ME(1-12)가 강한 항진균활성을 나타내었으며 agar hole method에서 clear zone이 생성되는 것을 확인하였다. 그리고 이러한 합성펩타이드가 (1,3)-β-D-glucan synthase 활성에 미치는 영향을 조사해 본 결과, 1μg/ml의 농도에서 ME, CA(1-8)ME(1-12) 및 MA(10-17) ME(1-12)는 각각 30.4%, 32.8%, 44.7%의 활성저해률을 나타내었다. 9. 수용액 조건하에서는 각 접합 펩타이드는 구조를 취하지 않으나, 50 % TFE 혹은 30 mM SDS조건에서는 높은 α-helical 구조를 취하였다. 10. 30 mM SDS조건에서의 접합 펩타이드의 α-helicity는 항균, 항암 및 항진균활성 보다는 적혈구 용혈활성에 긴밀한 관련성이 있었다. 11. Magainin Hybrid 합성펩타이드가 Aspergillus fumigatus의 성장에 미치는 저해효과를 조사해 본 결과, MA-inv 및 MA(10-47) ME(1-12)가 강한 항진균활성을 나타내었으며 MA(10-17) ME(1-12)와 같은 경우는 F. oxysporum에서도 강한 항진균활성을 나타내었다. F. oxysporum에서와 마찬가지로 항진균 활성을 가지는 ME, MA, MA-inv 및 MA(10-17) ME(1-12)가 (1,3)-β-D-glucan synthase의 활성에 미치는 영향을 조사해 본 결과, 펩타이드 농도가 10μg/ml에서 MA-inv가 87%의 강한 활성저해률을 나타내었다. 12. 항진균활성을 가지는 것으로 확인한 CA(1-8)ME(1-12)접합 펩타이드를 주형으로 항진균활성을 증진시킬려고 아날로그 펩타이드를 합성, 설계하여 그 활성을 확인한 결과, 18, 19번 위치의 Thr을 Lys으로 치환한 유도펩타이드(A1)가 CA-ME접합펩타이드 보다 높은 항진균활성을 나타내었다. 항진균활성이 가장 높은 유도펩타이드의 작용기작을 FACScan analysis를 통해 확인한 결과 T. beigelii가 이 유도 펠타이드에 의해 melittin과 거의 비슷하게 확실히 사멸시킨다는 결과를 얻을 수가 있었다. 이러한 사실을 Confocal microscopy로 확인한 결과, 유도펩타이드가 세포막을 통해 내부로 들어감으로서 세포막을 파괴시키는 것을 확인할 수가 있었다. 13. 펩타이드의 항균활성과 2차구조와의 상관관계를 알아보기 위하여 anionic lipid를 모사하는 SDS조건에서의 CD spectra를 측정한 결과, 11개의 아미노산으로 이루어진 truncated peptide (B3, B4, B5, B6, B7)들이 SDS 조건에서 50% TFE 조건에서 충분히 α-helical conformation을 이루고 있음을 알았다. 14. CA-MA 유사체 펩타이드의 인지질막의 파괴 능력을 phosphatidylcholine 또는 phosphatidylcholine과 phosphatidylserine의 혼합물 리포좀을 이용하여 조사하였다. V. 연구개발결과의 활용계획 1. Lactoferricin 유래 유사체 펩타이드에 관한 연구를 통하여 항균효과는 좀더 좋아진 반면 세포독성은 낮아진 짧은 길이의 펩타이드가 선정되었다. 이 결과를 이용하여 항균 펩타이드로서의 적용 가능성을 타진하는 지속적인 연구가 필요한 것으로 사료된다. 2. 본 연구에서 합성한 접합 펩타이드들은 항균 펩타이드의 구조와 항생작용과의 상관관계를 이해하는데 적용 될 수 있다. 3. 본 연구에서 합성한 접합 펩타이드들은 항균 펩타이드의 항균, 항암, 항진균작용의 기작을 이해하는데 적용 될 수 있다. 4. 본 연구에서 합성한 접합 펩타이드들은 지금까지 보고된 항암펩타이드들보다 비슷하거나 약한 적혈구 용혈활성을 가지므로 항암제 개발에 유용하게 이용될 수 있다. 5. 본 연구에서 합성한 접합 펩타이드들은 그램-음성균, 그램-양성균, 소세포 폐암세포, 진균 및 적혈구세포에 대하여 각각 다른 활성을 나타내므로 항균 펩타이드의 표적세포의 특이성을 밝히는데 유용하게 이용될 수 있다.

I. Title Development of improved synthetic antimicrobiaI peptides using peptide engineering technologies II. Goal and importance The peptides are the important bioactive materials acting as hormones, growth factors, and antimicrobial properties. The possibilities are suggested that the peptides use as vaccine material in immune reaction and as diagnostic reagents. Lactoferrin, found 1n most exocrrine secretions of mamma1s, is a 80 kDa-sized protein and possess various bioactive functions. Of these biological functions, the antimicrobial activities of lactoferrin display as apolactoferrin, released ferrous ion. A pepsin hydrolysate of lactoferrin, named as lactoferricin, has more potent antimicrobial activity than undigested lactoferrin. Therefore we are willing to develope the lactoferricin-derived ana1ogues, possessing the higher antimicrobial activity and lower cellular toxicity by the synthetic technology of peptides and the examination study of peptidesꡑs structure. Antibacterial peptides are widely distributed in nature including insects, horseshoe crabs, frogs and mamma1s. These peptides have been known to play important roles in insect immunity and host defense. Most of them have a common feature of being highly basic due to multiple basic amino acid residues, and of forming amphipathic structures in phospholipid bilayers. Their basic natures are related to the interaction with negativeIy charged membranes of the target celIs and their amphipathic character which allow them to be incorporated into the membrane, ultimateIy disrupting membrane potential and/or structure, and cell death. Cecropins isolated from cecropia moths are very active against Gram-negative bacteria, but less active against Gram-positive bacteria. Magainin-2 (MA) purified form frog skin and bombinin (BO) have weak antibacterial activity against both Gram-positive and negative bacteria. Melittin (ME), a bee venom, has good antibacterial potency against both Gram-negative and Gram-positive bacteria, but is also cytotoxic for eukaroytic cell. Many analogues of native antibacterial peptides designed by amino acid substitution with improved antibacterial activity showed an increased hemoIytic activity. A notable exceptions are the CA-ME hybrid peptides composed from the N-terminal sequences of CA and ME. Among these hybrid peptides, CA(1-8)-ME(1-12) composed from CA(1-8) and ME(1-12) has known to have more powerful antibacterial activity without hemolytic activity in the agar hole method than CA. However, in our study, CA(1-8)-ME(1-12) showed a significant hemolytic activity in physiological condition using the solution method. In order to obtain synthetic peptides having potent more antibacterial, antitumor or antifungal activities with lower hemolytic activity than CA(1-8)-ME(1-12), in this study, CA(1-8)-ME(1-12), CA(1-8)-MA(1-12) and CA(1-8)-BO(1-12) based on the N-terminal sequence CA and MA(10-17)-ME(1-12), BO(11-18)-ME(1-12) and LF(20-29)-ME(1-12) based the N-terminal sequence of ME were designed and synthesized. Also, several analogues were designed from the α-helical wheel diagrams of CA(1-8)-ME(1-12) and CA(1-8)-MA(1-12). All peptides were synthesized by the solid phase method using Fmoc-chemishy. The antibacterial, antitumor and antifungal activities of synthetic peptides were determined by the growth inhibition against Gram-positive and negative bacteria, small cell lung cancer(SCLC) cells and several fungal strains, respectively. Also, their hemolytic activites were assessed by cell lysis against human red blood cells(h-RBCs). To examine the antimicrobial mechanisms of the antimicrobial peptides, the induced release of liposome-entrapped f1uorescence probes by antimicrobiaI peptides was determined using the liposome as model membrane system. We expect that it is possible to explain the nature of peptide-phospholipid membrane interaction and to examine the mechanisms of antimicrobial acitivities by measurements of release of probes according to phospholipid compositions of liposome and size distinctions of entrapped probes. Also, the aim of this study is the development of shortened peptides with potent antifungal activity without cytotoxicity-The desined peptides showed stronger antifungal activity than their native peptides MA, CA and ME but did not have hemolytic activity. Therefore these hybrid peptides can be potentially used as model peptides to develope the powerful antifungal drug for treating the fungal infection, and be also used for understanding the structure-antifungal activity relationships. III. Contents and Scope 1. The truncated peptides with higher antimicrobial activities composed of 11 amino acid residues derived from bovine lactoferricin were desinged, synthesized and purified. 2. CA(1-8)-ME(1-12), CA(1-8)-MA(1-12), CA(1-8)-BO(1-12) MA(10-17)-ME(1-12), BO(11-18)-ME(1-12), and LF(20-29)-ME(1-12) were designed and synthesized by the solid phase method using Fmoc-chemistry. 3. Eight analogues were designed from the α-helical wheel diagrams and primary structures of CA(1-8)-ME(1-12) and CA(1-8)-MA(1-12) and synthesized by the solid phase method using Fmoc-chemistry. 4. The amino acid compositions and molecular weights of synthetic peptides were determined by amino acid analysis and matrix-assisted laser desorption ionization (MALDI) mass spectrometry. 5. The antimicrobial activities against E. coli and B. subtilis were assayed by the determination of minimal inhibitory concentration (MIC) and the toxicities were measured by hemolytic assay. 6. Antibacterial activity of hybrid synthetic peptides were determined by the growth inhibition against Gram-positive and negative bacteria using either the agar hole method or MIC value measurement. 7. Antitumor activity of synthetic peptides were determined by the growth inhibition against small cell lung cancer (SCLC) cells. 8. In order to design peptides with powerful antifungal activity without cytotoxic property of ME. The hybrid peptides CA(1-8)ME(1-12) and MA(10-17)ME(1-12) were synthesized by solid phase method. MA(10-17)ME(1-12) showed potent antifungal activity comparable to ME against Fusarium oxysporum with no hemolytic activity and strong inhibition of (1,3)-β-D-glucan synthase activity. 9. Two peptides, MA-inv and MA-ME, with the sequence 10-17 of MA at their N-termini were designed and synthesized. The peptides had higher antifungal activities against Aspergillus fumigatus. MA-inv showed up to 82% inhibition of (1,3)-β-D-glucan synthase activity. 10. In order to design synthetic peptides with potent antifungal activity, several CA(1-8)-ME(1-12) analogues were sythesized Substitution of Thr for Lys at position 18 and 19 of CA(1-8)ME(1-12) showed potent antifungal activity. To study the antifungal mechanism of this peptide, FACScan analysis and Confocal microscopy were perform. The results showed that this analogue peptide acts by pore formation in the cell membrane. 11. To study the antifungal mechanism of synthetic peptides were performed by both fluorescence activated flow cytometry and confocal laser scanning microscopy were performed. 12. To investigate the structure-antimicrobial activity relationships, the cicular dichroism (CD) spectra in 30mM SDS mimicking anionic lipid membrane were measured. 13. The nature of peptide-phospholipid membrane interaction and membrane disruption properties were examined using liposome as model system. IV. Results 1. All synthetic peptides were confirmed to be homogeneous from an elution peak by analytical RP-HPLC and their correct amino acid compositions and molecular weights were determined by both amino acid analysis and MALDI mass spectrometry. 2. The MIC of lactoferricin (B1) was 22.5 μg/ml for E. coli and 6.3 μg/ml against B. subtilis and the antimicrobial activities of B2, substituted cysteine of B1 to serine, were the lower than B1. B3 and its subsituted peptides (B4-B7) also had similar antimicrobial activity compared to the long peptides (B1 and B2). B4, substituted all arginine of B3 with lysine appeared the lower antimicrobial activity. The results indicated that the side chain of arginine may be more effective than those of lysine as basic amino acids on antimicrobial activities. Although the short peptides (B3-B7) composed of 11-residues had similar antimicrobial activity with the long peptides (B1 and B2), the peptides showed much less hemolytic activities. 3. The disruption of the outer membrane of E. coli was monitored by measuring the periplasmic enzyme, β-lactamase, activity. Lactoferricin-derived peptide induced the disruption of the outer membrane in dose-dependent and time-dependent manners, indicating the peptide could have disrupted the outer membrane by release of LPS. 4. Although hybrid peptides, CA(1-8)-MA(1-12), CA(1-8)-BO(142), MA(10-17)-ME(1-12), LF(20-29)-ME(l-12) as well as CA(1-8)-ME(1-12) are shorter in size than their parental peptides, they have potent antibacterial activities against both Gram-positive and Gram-negative bacteria without lytic properties against human red blood cells in the agar hole assay. 5. Greater antitumor and antibacterial activities were observed when the residues 16, 18 and 19 of CA(1-8)-ME(1-12), CA(1-8)-MA(1-12) and their analogues are hydrophobic(Leu or Val), basic(Lys) and basic(Lys), respectively. 6. An analogue in which Ser at position 12 in CA(1-8)-MA(1-12) was substituted with Leu showed potent antitumor and antibacterial activity with mimimal hemolytic activity (0.7% hemolysis at the 200 μg/ml peptide). 7. MA-inv and MA-ME, with the sequence 10-17 of MA at their N-termini had higher antifungal activities against Aspergillus fumigatus. MA-inv showed up to 82% inhibition of (1,3)-β-D-glucan synthase activity. 10. To study the antifungal mechanism of this peptide, FACScan analysis and Confocal microscopy were perform. The results showed that this analogue peptide acts by pore formation in the cell membrane. 11. To investigate whether the antimicrobial activity of the lactoferricin-derived peptides is related to their secondary stucture, CD analysis were performed in 30mM SDS mimicking anionic lipid membrane. The overall CD spectra of the short peptides (B3-B7) dissolved in 30mM SDS showed similar patterns in 50% TFE, indicating that most portions of the short peptides were involved in interaction with the SDS molecule and induced to α-helical conformation. 12. In aqueous solution, none of the peptides showed any defined conformation, but the peptides adopted the α-helical structure in 50% trifluoroehthanol(TFE) and 30mM sodium dodecylsulfate(SDS). 13. The α-helicity of the peptides in a 30 mM sodium dodecylsulfate(SDS) solution is more closely related hemolytic activity than antitumor, antibacterial and antifungal activity. 14. The disrupting properties of CA-MA analogue peptides were determined by the leakage of fIuorescent probe of phosphatidylcholine vesicle or phosphatidylcholine phosphatidylserine (4:1) mixture vesicles. V. AppIications 1. The 11-residue truncated peptides derived from bovine lactoferricin, possessing the higher antimicrobial activity but lower cellular toxicity, was selected by the basic studies of lactoferricin-derived ana1ogues. In further research, we will apply these results to the developements of the antimicrobial peptides as therapeutic antibiotics. 2. Hybrid peptides synthesized in our study could be applied in understanding the structure-antibiotic function relationships of antibacterial peptides. 3. Hybrid peptides synthesized in our study could be applied in understanding the mechanism of antibateria1, antitumor, and antifungal actions of antimicrobial peptides. 4. As hybrid peptides synthesized in our study had higher antitumor activity than other antitumor peptides reported previously, but showed similar or lower hemolytic activity, these peptides could be applied in developing antitumor drugs. 5. As hybrid peptides synthesized in our study acted on Gram-positive and negative bacteria, small cell lung cancer cells, fungi, and human blood red cells, respectively, these peptides could be applied in elucidating the details of target cell specficity.
