본 연구의 주목적은 유기합성에 유용한 유기반응을 중심으로 천연효소와 인공효소에 의한 촉매를 자유롭게 개발하는데에 필요한 기초 학술정보의 발굴, 실험기법의 개발, 고급인력의 배출에 있다. 이에 따라 유기반응에 대한 천연효소, 수정효소, 항체효소, 합성효소, 효소모형의 촉매작용에 관련된 유기합성, 반응속도론, 효소화학, 면역학적인 연구가 본 총괄 연구과제에 포함되어 있다. 이러한 총괄연구과제의 효율적인 수행을 위하여 4개의 세부연구과제를 계획한 바, 제 1세부과제에서는 효소 및 효소모형반응 메카니즘연구, 항원고안 및 항체효소의 특성연구
본 연구의 주목적은 유기합성에 유용한 유기반응을 중심으로 천연효소와 인공효소에 의한 촉매를 자유롭게 개발하는데에 필요한 기초 학술정보의 발굴, 실험기법의 개발, 고급인력의 배출에 있다. 이에 따라 유기반응에 대한 천연효소, 수정효소, 항체효소, 합성효소, 효소모형의 촉매작용에 관련된 유기합성, 반응속도론, 효소화학, 면역학적인 연구가 본 총괄 연구과제에 포함되어 있다. 이러한 총괄연구과제의 효율적인 수행을 위하여 4개의 세부연구과제를 계획한 바, 제 1세부과제에서는 효소 및 효소모형반응 메카니즘연구, 항원고안 및 항체효소의 특성연구, 고분자를 이용한 수정효소 및 합성효소의 개발등을 분담하였고, 제 2세부 과제에서는 모노클로날 항체의 제작 및 정제방법의 개선, 병원균 분해 효소작용을 가진 항체에 의한 면역효과, 모노클로날 항체의 부동화등을 분담하였고, 제 3 세부과제에서는 고리펩타이드 합성, 펩타이드의 촉매작용 등을 분담하였고, 제 4세부 과제에서는 효소를 이용한 유기합성을 분담하였다. 이러한 세부과제의 구성의 결과로 각 세부과제의 고유한 문제점의 해결에 대하여 타세부과제가 도움을 줄 뿐 아니라 유기합성에 대한 천연 및 인공효소의 개발과 응용이라는 공통된 목표를 위하여 모든 연구진이 협동할 수 있었다.
제 1세부 과제에서는 천연효소, 효소모형, 항체효소, 반합성효소 등에 관한 연구를 수행하였다. 첫째로, 여러가지 아미노산의 α-(벤조일아미노) 신나모일 유도체의 가수분해 반응에 대한 카르복시펩티다제 A 촉매작용의 반응속도론적 연구 (Bioorg. Chem. 18, 276-282 (1990)에 발표)에서는 천연 효소인 카르복시페티다제 A 활성자리의 기하학적 구조에 관한 정보를 기질의 구조를 조정함으로써 수집하였다. 둘째로, 카르복시펩티다제 A 의 활성자리의 분광학적 적정 (J. Biochem. Biophy. Meth. 22, 167-170 (1991)에 게재)에서는 천연효소인 카르복시펩티다제 A 촉매 반응에서 중간체를 정량적으로 축적시키고 아울러 활성자리를 분광학적으로 적정할 수 있는 방법을 발견 하였다. 세째로, 카르복시펩티다제 A의 모형연구 (Bioorg. Chem. 18, 345-360 (1990)에 게재) 에서는 이 효소의 특성 다수를 소형 유기분자로 재현하는데 성공하였으며, 이효소의 메카니즘에 관한 중요한 정보를 수집하였다. 네째로, m-(2-이미다졸릴아조)페닐 p-톨루엔설포네이트의 가수분해 반응에서의 히드록소구리 (Ⅱ) 이온에 의한 일반염기 촉매작용과 첨가 중간체의 존재(J.Org. Chem. 56, 4364-4370 (1991)에 게재)에서는 설폰산에스테르의 가수분해를 소재로 하여 금속에 배위된 수산화이온이 일반염기로 작용할 수 있다는 금속의 새로운 촉매요인을 발굴하였다. 다섯째로, 금속이온을 루이스 산 촉매로 포함하는 금속효소의 모형 연구 (Acc. Chem. Res. 25, 273-279에 게재)에서는 루이스산 촉배로 작용하는 금속이온의 새로운 촉매역할을 발굴하는 작업, 목표 효소인 카르복시펩피다제 A의 촉매특성을 가능한 한 많이 재현하는 모형을 제작하는 작업, 효소와 같은 원리로 작동하는 합성촉매를 고안하는 인공금속효소의 설계에 관한 작업을 수행하였다. 여섯째로 L-만델산에 대한 항체. 모노클론 항체의 속도론적 분석과 항체를 사용한 광학 분할 (Bull. Korean Chem. Soc. 12, 352-354 (1991)에 게재) 에서는 만델산 환원항체효소를 선별하는 새로운 방법을 도입하였고, 붕소수소화물 환원제의 반응에서 항체를 촉매로 사용하는 새로운 개념을 최초로 시험하였고, 만델산의 라세미화합물을 광학분할하는데에도 항체를 사용하였다. 이 과제는 제 2세부 과제와 공동으로 수행하였다. 일곱번째로, 폴리(알릴아민) 과의 결합을 통한 키모트립신의 내구성 제고 (Bioorg. Chem.에 게재예정) 에서는 α-키모트립신을 여러가지 폴리(알릴아민) 유도체와 공유결합으로 연결하여 변성제 및 열에 의한 불활성화에 대한 저항을 두드러지게 증가시킬 수 있었다.
제 2세부 과제에서는 촉매작용을 가지는 모노클로날 항체를 개발하고 이들을 효과적으로 이용할 수 있도록 정제하는 방법의 개선에 목표로 두고 연구를 수행하였다. 일차적으로는 효소작용을 대신하는 항체를 개발하고자 하였으며 (제 1 세부 과제 결과 참조), 두번째로는 모노클로날 항체의 개발 방법의 개선에 중점을 두었고, 세번째로는 여러가지 물질에 대하여 효소항체를 개발하고 그 효소의 작용을 이해하기 위한 기본적인 모델 연구에 소요되는 모노클로날 항체의 개발 및 그 정제 방법의 개선에 대하여 연구하여 왔다. 이제까지 알려진 대개의 효소항체들이 작용하고 있는 구조적인 특성에 기초하여 리그닌과 같은 물질의 분해에 효소항체가 사용될 수 있고 그러한 연구결과는 자원의 활용에 이용될 수 있다고 판단하여, 리그닌 및 이들의 분해에 중요한 역할을 하고 있는 laccase라는 효소의 작용을 이해하기 위한 실험 모델의 정립을 목표로 하여 느타리 버섯에서 생산된 laccase 대한 여러 종류의 모노클로날 항체를 만들고 그 작용 양상을 연구하였다.
제 3세부 과제에서는 고분자 지지체내의 전하 이동상호 작용(charge-transfer interaction)을 이용하여 고체상 고리 펩티즈를 합성할 때의 일반적인 반응 최적조건을 찾아보고 고리화 반응의 동력학적거동을 고찰하여 보았다. N-말단기에 donor로서 쓰일 수 있는 Met나 Trp을 도입하고 이들의 고리화 반응을 여러가지 용매조건하에서 고찰하여 보았다. 전반적으로 반응속도는 용매 ET 값을 낮을 수록 빨라지며, 고분자 지지체에서 부터의 거리가 짧아질수록 반응이 빨라졌다. 4-Nitorooxime 수지와 3-nitrooxime 수지의 반응성을 비교하여 볼때 3-nitrooxime 수지에서의 고리화 반응이 빨랐으며, 온도에 따른 속도의 변화는 온도가 낮을 수록 빨라졌다. 이러한 결과들로 부터, 고체상 고리 펩티드의 고리화 반응에서 전하 이동착물 (charge-transfer complex)을 효과적으로 이용할 수 있음을 확인할 수 있었다. 고리화 반응시 고분자 지지체의 경직성의 정도가 반응에 큰 변수라 생각되어 여러가지 펩티드를 고분자 지지체에 도입한 후 고리화 반응전후의 Tg (glass transition temperature) 변화를 측정하여 보았다. 고분자 내에서의 가교에 의한 Tg의 변화는 가교의 양이 1% 인 Bio-beads S-X1의 경우 116℃로 측정되었고, 가교의 양이 약 20%를 상회하는 것으로 알려진 macroreticular polystyrene의 Tg가 127℃인 것으로 보아 가교의 양이 1%에서 20%로 증가도 Tg의 차이는 별로 보이지 않았다. 그러나 펩티드 사슬로 graft를 시켰을 경우 Tg의 변화는 매우 컸으며, 고리화 반응전후의 Tg 변화로 부터 3-nitobenzophenone oxime수지 만이 고체상 고리펩티드 반응에 가장 적합한 수지임을 알 수 있었다. 이와 같은 결과들을 바탕으로 효소 모델계인 cyclo (Phe-Asp)?) 와 cyclo(Gly-Ser-Gly-His-Gly-Glu)를 효율적으로 합성할 수 있었다.
제 4 세부 과제에서는 효소 촉매를 이용한 새로운 유기합성반응의 개발이라는 목적하에 기질가수 분해효소, D-젖산탈수소효소, 및 다효소시스템에 대해 연구하였다. 지질가수분해효소 연구에서는 지단백질가수분해효소 (lipoprotein lipase, LPL)의 특이성을 규명하였으며, 이를 기초로 기질 모델을 제시하였다. 이 모델은 새로운 기질을 디자인하거나, 반응의 입체선택성을 예측하는데 유용하게 사용될 수 있다. 또한 기질가수분해효소를 이용하여 광학활성을 갖는 페로센 화합물을 합성하였다. 젖산탈수소효소 연구에서는 D-젖산탈수소효소를 이용하여 네 종류의 D-2-hydroxy acid를 고순도, 고수율로 합성하였다. 그리고, 이들 중 (R)-cyclopropaneglycolic acid로 부터 세 단계의 화학반응을 거쳐 (R)-cylcopropyloxirane을 합성하였다. 다효소시스템의 연구에서는 네 가지 효소를 동시에 사용하는 다효소반응을 이용하여 D-프룩토오스-1,6-이인산을 합성할 수 있었다. 이상의 연구를 통하여 화학적으로 합성하기 어려우나, 유기합성에 유용한 키랄분자 및 탄수화물을 합

The main objective of this project is discovery of basic scientific principles, development of experimental methodology, and tranining of advanced brains needed for design of versatile catalysts for organic reactions based natural enzymes and artificial enzymes. Accordingly, organic synthesis, kinetics, enzyme chemistry, and immunology related to catalysis by natural enzymes, modified enzymes, antibody enzymes, synthetic enzymes, and enzyme models are included in this project. In order to carry out the objectives, the project consists of 4 subprojects. Subproject 1 includes studies on mechanisms of enzymes and enzyme models, design of antigens, characterization of catalytic antibodies, and development of modified enzymes and synthetic enzymes based on polymers. Subproject 2 includes improvement of pruification of monoclonal antibodies, immunological studies on antibodies, and immobilization of antibodies. Subproject 3 in cludes production of cyclic peptides and their catalytic applications. Subproject 4 includes organic synthesis utilizing enzymes. Each of these subprojects are designed to cooperate with one another in achievment of the main objective of this project.
In subproject 1, studies were carried out on natural enzymes, enzyme models, catalytic antibodies, and semisynthetic enzymes. In the first paper published in Bioorg. Chem. 18, 276-282 (1990), kinetics of carboxypeptidase A-catalyzed hydrolysis of a-(benzoylamino) cinnamoyl derivatives of various amino acids was investigated. Here, information on the geometry of the active site of carboxypeptidase A was obtained by adjusting substrate structures. In the second paper published in J. Biochem. Biophy. Meth. 22, 167-170 (1991), quantitative accumulation of intermediates was accomplished in carboxypeptidase A-catalyzed reactions and a method was discovered to titrate the active site spectrophotometrically. In the third paper published in Bioorg. Chem. 18, 345-360 (1990), models of carboxypeptidase A was studied, successfully reproducing many catalytic features of the enzymes with small molecules and obtaining important mechanistic clues for the enzyme. In the fourth paper published in J. Org. Chem. 56, 4364-4370 (1991), general base catalysis by hydroxoCu(Ⅱ) ion and existence of addition intermediates were demonstrated in the hydrolysis of m-(2-imidazolylazo) phenylp-toluenesulfonate. In the fifth paper published in Acc. Chem. Res. 25, 273-279, model studies of metalloenzymes involving metal ions as Lewis acid catalysts were reported. Here, discovery of novel catalytic roles of metal ions acting as Lewis acid catalysts, reproduction of many catalytic characteristics of a target enzyme with small molecules, and design of synthetic catalysts mimicking enzymes were included. In the sixth paper published in Bull. Korean Chem. Soc. 12, 352-354 (1991), monoclonal antibodies for L-mandelate were prepared. In this study, kinetic assay of monoclonal antibodies, use of antibody catalysts in borohydride reductions, and optical resolution with antibodies were included. This work was carried out in collaboration with subproject 2. In the seventh paper to be published in Bioorg. Chem. (1992), enhancement of durability of chymotrypsin, such as resistance to denaturing agents and thermonicactivation, was accomplished by cross-linking with poly (allylamine).
In subproject 2, we have tried to develop monoclonal antibodies with catalytic activities, which can replace enzymes in various steps of biological metabolic pathways, and to improve the moethods of purifying the antibodies without any activity loss. Monoclonal antibodies were, in the first place, developed with catalytic activities against several transient state substrates (see the results of subproject 1), and then the way how to develop monoclonal antibodies has been tried to be improved for better production with intact activities. Finally, we exteneded our effort to the related proteins to use the monoclonal antibodies for modelling and understanding of their roles in the metabolic pathways. Since most of the monoclonal catalytic antibodies were developed against the substrates with phenol ring, it should be possible to develop monoclonal antibodies against lignin which is an important component of wood, and can be used for the improvement of form\est resources. Laccase, in addition, was used for monoclonal antibody development for future modelling of this enzyme's bidirectional activity in polymerization and degradation.
In subproject 3, cyclization reactions of model peptides were conducted on nitrobenaophenone oxime resins. Four model peptides with different chain lengths which contain N-terminal Met or Trp residue as an electron donor were prepared on the oxime resins. Cyclization reactions of the model peptides were monitered by reversed phase HPLC. Pseudo first order rate constants were obtained in 6 different organic solvents with various polarity. The results showed that cyclization rate constants in THF were the greatest and the one in DMF were the smallest in four model peptide cases. Good correlations were obtained between the rate constants and the E? values of the solvents. Interestingly, the rate constants were decreased with increasing temperatures, which clearly implicated that the cyclization process proceeded via charge-transfer complex formation. The effect of grafted peptide moedty on the rigidity of the polymer acyl carriers was estimated by measuring the Tg/s of various pertide-oxime resin esters using differential scanning calorimetry (DSC). The degree of crosslinking on the polymer affected very little on the rigidity of the polymer, but graftization with oxime group and peptides largely affected the rigidity of the polymer. Especially, the size of the first amino acids bound to the polymers mainly affected the rigidity of the polymer and the cyclizational behaviours. From the results of the Tg study, we rationalized that 3-nitrobenzophenone oxime resin should be very effective in cyclization reaction of peptide. On the basis of these findings, cyclo (PheAsp)?) and rationalized cyclization cyclo (Gly-His-Gly-Glu) which are hte models of enzymes were synthesized effectively.
The fourth subproject consists of three parts: 1) organic synthesis using lipases, 2) organic synthesis using D-lactate dehydrogenase, and 3) organic synthesis using multi-enzymes. In the lat part, we established the specificity of lipoprotein lipase (LPL) and proposed the substrate model for predicting and interpreting the enantioselectivity in LPL-catalyzed reactions. In addition, the efficient resolutions of ferrocene-containing substrates have achieved by using lipase-catalyzed hydrolysis and transesterfication. In the 2nd part, we successfully employed D-lactate dehydrogenase (Staphylococcus epidermis) as the catalyst in the synthesis of four different D-2-hydroxy acids, which provided the products in high yields and excellent optical purities. To show th esynthetic utility of the enzymatically reduced products, (R)-cyclopropaneglycolic acid was converted via three chemical steps into (R)-cyclopropyloxirane. In the last part, we syntheiszed D-fructose-1,6-diphosphate by using a four-enzyme system (glycerokinase, pyruvate kinase, triosphosphate isomerase, and FDP aldolase). This system included dihydroxyacetone as the starting material, ATP as the phosphorylation agent, and phosphoenolpyruvate/pyruvate kinase as the ATP-regeneration system. The products were isolated as the barium salts in 60% yield.
