Alpha-amylase는 동·식물에 널리 분포되어 있으므로 기원이 서로 다르다 해도 작용기가 유사할 것이라는 가정하에 본 연구팀은 Bacillus, Streptomyces 및 벼로부터 α-amylase를 coding하는 유전자를 찾아 DNA 염기서열을 비교 검토하였다. 각 연구진에서는 유전자를 찾았고 일부는 DNA 염기서열을 결정하였고 일부는 현재 진행중에 있다. 본 실험에서의 성가는 유전자를 발견한 것으로 학문적으로 중요한 기초가 될 것이며 또한 streptomyces는 maltotetraose를 특이적으로 다량 생산하여 산업적으로 이용이 가능할 것으로 기대된다. 연구팀 상호간의 협력에 의해 연구가 원만이 진행되었으며 연구결과를 요약하면 다음과 같다.
벼 α-amylase 유전자는 벼 cDNA library로부터 α-amylase cDNA 유전자를 분리하는 실험으로 수행하였다. 먼저 gene bank 를 이용하여 α-amylase의 염기서열을 탐색하여 conserved region 의 염기서열을 갖는 21-mer, 23-mer, 16-mer의 deoxyoligonucleotide를 각각 합성하여 정제하고 이를 probe DNA로 이용하였다. 예비처럼 결과 벼의 호분층을 10 ppm 지베렐린, 20ppm CaCl?가 포함된 배지에서 36 시간 동안 배양하였을 때 α-amylase 역가가 가장 높았으며 이 조건에서 호분층을 배양하여 total RNA 를 분리하였다. 이로부터 mRNA 를 분리하고 0.3 kb-2.5 kb크기의 cDNA 를 합성한 후 λgtll vector에 삽입하여 5 X 10? pfu/㎍ RNA 크기의 cDNA 유전자은행을 작성하였다. 합성된 deoxyoligonucleotide를 probe로 하여 plaque hybridization 방법으로 cDNA 유전자 은행으로 부터 6 개의 α-amylase clone 을 선발하였다. 또한 PCR screening 방법으로 1 kb 이상의 insert를 포함하는 2개의 clone 을 얻었으며 이들을 다시 plaque hybridization 방법으로 검색해 본 결가, 두 clone 모두에 probe가 강하게 binding하는 것을 관찰할 수 있었다. 이 clone을 사용하여 Northern blot analysis 를 실시한 결과 이를 coding 하는 mRNA가 호분층에서만 발현되는 것으로 확인되었다. Genomic Southern blot analysis 결과 다수의 band 가 나타나는 것으로 보아 α-amylase의 유전자를 분리하고 5' 부위의 염기서열을 분석하였다. 그 결과 분리한 clone 이 지금까지 보고된 α-amylase 유전자와 80-90%의 상동성을 지니고 있었으며, 특히 벼의 α-amylase와는 95%의 유사성을 보임으로써 분리해낸 유전자가 α-amylase 유전자임이 확인되었다. 염기서열 분석결과 분석된 부분의 염기서열 내에 두개의 ATG codon이 15 bp의 간격을 두고 발견되며 intron/exon boundary도 발견된다.
Bacillus licheniformis가 분비하는 내열성 α-amylase 유전자로 부터 효소를 생산하고 antibody를 얻어 효소정제용 chromatography에 이용할 수 있었고 유전자의 DNA 를 분석하여 amylase의 1차 구조와 작용기전에 대하여 연구하였다. Antibody 를 이용한 immunoaffinity chromatography 는 높은 수율로 α-amylase를 정제할 수 있었다. DNA 염기서열로부터 추정한 아미노산 서열을 비교할 때 α-amylase의 4개 conserved region 간에는 상동성이 매우 높았다. 내열성 α-amylase의 구조적 특징은 없었으나 G + G 함량이 높았다. 유전자 조작에 의한 재조합체 α-amylase는 특정 product를 생산할 수 있었으며 내열성의 변화도 유도할 수 있었다.
퇴비와 토양에서 순수 분리?을 측정하고 조효소액으로 amylase 활성과 maltotetraose 생산성을 고려하여 KSM 35 균주를 최종 선방하였다. KSM 35는 형태학적 특성으로 보아 방선균임을 알 수 있었으며 전균체성분 분석과 세포벽 구성성분 분석결고, (sub)(LL)-DAP와 glycine을 함유한는 type 1의 세포벽을 가지는 것으로 분류되며, Bacillus의 amylase와 같은 endo 형의 효소라고 판단되었고, Streptomyces KSM-35의 amylase 활성 최적 pH는 6부근이었다. Streptomyces KSM-35의 amylase의 pI는 4.3이고 분자량은 50,500 정도로 추산된다. streptomyces KSM-35의 amylase는 호화된 전분의 분해력에 비하여 생전분 분해력은 매우 약하였으며 전분 가수분해 산물을 분석한 결가 2.5% 분용액에서 가수분해 주산물인 maltotetraose가 0.56%, 그리고 maltotriose가 0.16%, maltose가 0.20% 생기었으며 그 외에도 glucose, maltopentaose, maltohexaose등이 미량 검출되었다. 이 균주를 이용하여 shot gun 방법으로 Streptomyces sp. KSM-35에서 amylase를 coding하는 유전자를 cloning 하였다. 또한 형질전환된 E. coli amylase 유전자 내부에 amylase 분비에 필요한 signal sequence를 가지고 있음을 추측할 수 있었다. Ca?? 이온 10mM 존재시 각각의 온도에서 30분 예열 한 후 잔존 효소 역가를 측정한 결과, 60℃에서는 최대 역가의 80%가 나타났다. TLC와 HPLC 를 이용하여 분석에 의하면 maltotetraose가 주산물로 생산되었으며, maltose와 maltotriose 순으로 생산되었다.

Each team isolated the gene, and the nucleotide sequencing is either completed or in progree. This and other related results provide us with an important basis for future academic investigation. Akso Streptomyces is expected to be of industrial use found to specifically produce maltotetraose in mass. Research progess was made satisfactoring through mutual coorperation among the teams. The research results are summarized as follow.
The rise α-amylase cDNA cloning experiment was carried by using germinating rice seed as starting material. The conserved regions DNA sequence of the α-amylase gene of plant were searched out by Gene Bank program.
Three oligonucleotides, 21-mer, 23-mer, and 16-mer were synthesized and used for polymerase chain reaction. Fro mRNA isolation, aleurone layers were prepared from the rice seeds and incubated in the media containing 20 ppm CaCl? , 10 ppm GA? with mold agitation. The polyadenylated mRNA was isolated from them by phenol extraction method and oligo (dT) cellulose chromatography. double strand cDNAs with 0.3 -2.5 kb range from the polyadenylated mRNA were synthesized and The cDNA expression library was constructed into λgt11 expression vector. The aleurone layer cDNA library was screened with synthetic deoxyoligonucleotide,. As a result 6 clones for α-amylase were obtained out of which 2 clones having more than 1 Kb insert were selected by PCR screening method. To obtain a full length rice α-amylase gene clone, PCR was carried out, and an 1.8 kb α-amylase DNA was obtained cloned into pUC19 plasmid DNA. It was confirmed by Northern blot analysis that this gene(pJY 19) is expressed only in the aleurone layers. Multiple bands were observed by genomic Southern blot analysis, suggesting that α-amylase is encoded by multiple closely related genes. The 5' region nucleotide sequence of the gene was determined. It was composed of intron and exon sequences. The sequence simularity of this region with α-amylase genes of other plants was 80-95%.
The gene encoding a thermostable α-amylase of Bacillus lickeniformis (ATCC27811) has been cloned and expressed in an E. coli strain HB101. α- amylase produced by the transformant was purified by using ion-exchange chromatography, Sephadex gel filtration, and preparative electrophoresis. Anti-α-amylase antibody was prepared from the serum of the rabbits immunized with purfied α-amylase. Immunoadsorbent was prepared by immobilizing the anti-α-amylase IgG on CNBr-activated sepharose and used for the immunoaffinity chromatography. The complete nucleotide sequence of the gene and its franking region was determined to reveal one open reading frame (ORF) of 1,536 bp which was preceeded by a putative promoter and a putative ribosome binding site. The ORF contained a putative signal sequence of 29 amino acids with Ser-Ala-Ala at the putative cleavage site. The deduced amino acid sequence of the amylase was composed of 438 amino acids and its molecular weight was calculated to be 55,200 daltons. Comparison of the amino acid sequence with those of various starch degrading enzymes revealed four conserved domains. It was found that the distance between the first and second domains is an important factor in determining the major reaction products. Delection of the nucleotide sequence between the first and second domains of the thermostable α-amylase led to alteration in the properties of the enzymer, especially in thermostability and the major reaction products.
A bacterial strain KSM-35 which yield maltotetraose as the main product from starch, was isolated from a compost. The strain KSM-35 was identifed as Streptomyces according to its morphological. cultural, and physiological characteristics. The amylase preparation obtained from Streptomyces sp. KSM-35 was purified by ammonium sulfate fractionation, DEAE-Toyo pearl ion-exchange chromatography and Sephadex G-100 filtration. The 45 fold purfied amylase preparation which showe a single band on SDS-PAGE was used for its enzymic characterization. The optimum temperature and pH for the reaction was 55℃ and pH 6.0, respectively. This enzyme was stable for 2 hours at 60℃. The stability was maintained in the pH range between 6.0 and 9.0. The enzyme activity was stimulated by Na? but inhibited by Cu??, Hg??, Fe???, EDTA and concanvalin A. The enzyme was a 50,500 dalton protein and its isoelectric point was 4.3. The endo type reaction of this enzyme yielded ca. 55% Maltotetraose as the main product from starch. The gene encoding the amylase was cloned and expressed in E. coli starch. The gene encoding the amylase was cloned and expressed in E. coli using the high-copy-number plasmid pBSG18, a derivative of pUC18, as a vector. The recombinant plasmid pSJ1 contained the 5.7 kb BamH1 DNA fragment originated from Streptomyce sp. KSM-35 chromosomal DNA. The starch hydrolysing activity of E. coli containing the recombinant plasmid pSJ1 was stronger than that of Streptomyces sp. KSM-35 chromosomal DNA. The starch hydrolysing activity of E. coli containing the recombinant plamid psJ1 was stronger than that of Streptomyces sp. KSM-35. The amylase obtained from E. coli with pSJ1 was identical to that obtained from Streptomyces KSM-35 in molecular weight, optimum temperature and pH value for reaction, stability against heat and pH, and the starch hydrolysing manner.
