최근 유전공학 기술의 발전은 재조합 세포를 이용한 각종 의약용 단잭질 생산을 비롯하여 기존 방법에 의해 생산되던 효소류나 항생물질 등을 보다 경제적으로 생산할 수 있는 새로운 수단을 제공하였다. 본 연구에서는 재조합 세포배양에 필요한 여러 공정설계 변수들에 대한 총체적인 연구를 수행하여 유전자 재조합 세포를 이용한 제품생산에 필요한 생물공정 관련 knowledge base를 확립하고 재조합 세포배양의 해석에 필요한 생물공정 속도론의 확립 및 재조합 세포 배양의 최적화 방안을 체계화 하고자 하였다. 한편 재조합 세포를 이용한
최근 유전공학 기술의 발전은 재조합 세포를 이용한 각종 의약용 단잭질 생산을 비롯하여 기존 방법에 의해 생산되던 효소류나 항생물질 등을 보다 경제적으로 생산할 수 있는 새로운 수단을 제공하였다. 본 연구에서는 재조합 세포배양에 필요한 여러 공정설계 변수들에 대한 총체적인 연구를 수행하여 유전자 재조합 세포를 이용한 제품생산에 필요한 생물공정 관련 knowledge base를 확립하고 재조합 세포배양의 해석에 필요한 생물공정 속도론의 확립 및 재조합 세포 배양의 최적화 방안을 체계화 하고자 하였다. 한편 재조합 세포를 이용한 유전공학 제품 생산의 경우 이제는 한 제품의 생산에도 여러 종류의 미생물 세포의 사용이 가능하며 동, 식물세포의 사용까지도 가능하게 되고 있다. 이렇게 점점 다양해져 가는 재조합 세포배양공정을 좀 더 체계적으로 분석하기 위하여 본 연구에서는 대장균을 비롯 효모, 방선균 그리고 식물세포 등 각각의 경우에 대한 재조합 세포 배양공정의 해석 및 최적화 연구를 수행하여 각 공정의 특성을 조사하였다. 그동안 수행된 연구결과는 주요내용을 세부과제별로 요약하면 다음과 같다.
제 1 세부과제에서는 서로 특성이 다른 플라스미드를 함유하는 재조합 대장균을 사용하여 재조합 미생물의 발효특성에 미치는 유전인자 및 환경인자의 영향을 고찰하였다. 플라즈미도의 replication origin은 동일하나 각기 그 특성이 조금씩 다른 pBR 322, pASl, pNK-2, pNKM-21 pASPT2를 함유하고 있는 재조합 대장균을 사용하여 유전형질의 변화가 재조합 균주의 배양특성에 미치는 영향을 집중적으로 연구하였는바 본 연구에서는 여러 발효공정 변수 중 특히 배양온도와 용존산소의 변화에 따른 유전자 발현이 재조합 균체의 성장, 세포 활성도, 재조합 세포의 안정성 및 생성물의 생산성에 미치는 영향을 종합적으로 분석 검토하여 실험결과를 토대로 유전인자와 환경인자와의 상호관계를 규명하고자 하였다.
재조합 단백질 분리 방법을 연구해보고자 수용성 이상계(aqueous two-phase system)를 이용한 방법의 사용가능성을 검토하였다. 여러가지 수용성 이상계를 검토한 결과 PEG 8000과 dextran 38800을 이용한 경우가 가장 효율적인 것으로 나타났으며 이들을 이용하여 순수정제된 인터루킨-2, inclusion body로부터의 인터루킨-2 분리, 그리고 재조합 대장균으로부터 직접적인 인터루킨-2 분리 효능에 대해 각각 조사하였다. 실험결과 inclusion body나 재조합 대장균 자체로부터의 인터루킨-2 회수를 위하여는 sodium dodecylsulfate(SDS)첨가가 매우 효율적인 것으로 나타났으며 인터루킨-2 함량 대비 SDS의 비율이 증가할수록 분배계수 및 회수율이 증가되었다. 최적조건에서의 겉보기 분배계수 및 회수율은 각각 3.0 및 80% 이상으로 나타나 기존의 다단계공정에 비해 일단계 공정으로 더 높은 분리 정제 효율을 얻을 수 있는 것으로 확인되었다.
또한 본 연구에서는 소위 Lee & Bailey 모델을 기초로 하여 cloned gene expression의 조절을 최적화 하기 위한 여러가지 유전자 재조합 DNA의 설계에 따른 효율성을 비교 검토하였다. 이미 그 타당성이 입증된 lac promoter 및 lambda pR promoter의 유전학적 구조모델을 이용하여 single 또는 dual-repressor control인 경우에 대하여 이론적 해석을 하였으며 시뮬레이션 결과 multicopy plasmid 에 cross-regulation 형태로 유전자 발현을 조절하는 것이 가장 효과적인 것으로 나타났다. 이러한 시스템은 지금까지 발표된 기존의 유전자 발현 조절기능에 비해 정교한 제어가 가능한 것으로 나타났는바 입증될 경우 유전자 발현 조절을 더욱 정교하게 할 수 있는 것으로 기대된다.
제 2 세부과제는 유전자 조작된 효모세포 배양기의 해석, 최적화 및 제어에 관한 연구로 재조합 효모를 이용하??의 효과를 실험적으로 고찰하고 실험결과의 modeling을 통하여 최적화 발효공정을 개발하는 것이 본 연구의 목적이다.
SUC2 유전자를 가진 재조합효모 발효공정의 최적화를 위하여 필요한 세포생육속도, 균체수율 및 invertase생성속도를 포도당 농도의 함수로 표시한 수식을 개발하고, 관련된 속도상수를 실험결과로부터 결정하였다. 이를 위해 일련의 회분식 및 유가배양식 실험을 행하였으며 또한 포도당 대사과정의 전환현상을 고려하기 위하여 cybemetic 개념을 도입한 세포 성장식을 개발하였다. 개발된 세포 성장식은 computer simulation을 통하여 실험결과와 잘 일치함을 증명하였다.
재조합미생물발효공정의 최적화를 위하여 conjugate gradient 방법에 의거한 새로운 최적화 방법을 개발하였다. 재조합효모 발효공정의 경우, 목적합수인 invertase농도를 최대로 하는 최적의 포도당 주입 전략을 구하였다. Pontryagin의 초적화 이론과 conjugate gradient 방법으로 구한 최적의 발효전략은 초기에 균체농도를 증가시키며, 후기에 invertase 생성을 유도하는 것이다. 10시간동안의 유가식 발효공정의 경우, 최저화 발효전략은 배양초기 6시간동안 포도당 농도를 2.0 g/1 이상으로 유지하여 invertase생성을 억제하면서 균체농도를 증가시켰고, 6시간에서 8시간까지는 주입을 멈추어 포도단 농도를 invertase 발현의 최적농도인 0.225 g/1로 감소시켜 invertase 발현을 유도하였다. 발효시간에 관한 영향을 살펴본 결과 발효시간은 12시간이 최적임을 알 수 있었다.
인체에서 유래한 lysozyme 유전자 (HLZ) 발현특성을 조사한 결과, 높은 포도당 농도(20 g/1이상)에서는 ADHl promoter에 의해 220 units/ml의 lysozyme이 생성되었으며, 생성된 lysozyme은 MF α1신호부위에 의해 90% 이상이 분비되어 발효액 중에 존재하였다. 혐기성 조건에서의 lysozyme 생성량은 호기성 조건에 비하여 최종 농도는 1.7배 향상되었다. 인체 lysozyme 분비를 관장하는 분비신호를 비교한 결과 MF α1 분비신호는 mouse α-amylase보다 세포 성장속도는 1.2배, lysozyme 분비속도는 2.3배, lysozyme 생산량은 1.5배 높게 나타나 효모유래의 분비신호가 유리함을 알 수 있었다.
제 3 세부과제에서는 항생물질인 cephamycin C 생합성 유전자가 재조합된 방선균을 구축하여 여러 발효특성을 조사하는 것으로 목적으로 하였다. 본 세부과제에서는 대장균/방선균 shuttle cosmid vector인 pKC505 내에 Str. clavuligerus ATCC27064의 genomic library를 제조하고, 환생성효소 (isopenicillin N synthetase; IPNS) 유전자중 homology가 높은 영역에 해당하는 oligonucleotide probe를 이용한 in situ hybridization 으로 cephamycin C를 생합성하는 유전자 뭉치를 선별하였으나, 이 유전자 뭉치 중 일부만을 선별할 수 있었다. 이에 선별된 유전자를 template로 하여 중합효소 연쇄반응으로 IPNS 유전자만을 증폭하여 대장균 운반체 pUC18에 삽입하므로서 E.coli DH5 내에서 IPNS 유전자를 발현시키는데 성공하였다. 이렇게 확보된 IPNS 유전자를 방성균 운반체인 pIJ 702에 이식한 후 Str. lividans 에 도입하여 IPNS 유전자를 발현시켰고, 또한 cephamycin C의 원 생산균주인 Str. clavuligerus에 도입하여 항생물질 생산능력이 향상된 방선균을 구축하였다.
이렇게 재조합된 방선균을 이용하여 여러가지 발효 변수들, 즉 plasmid 안정성, 탄소원, 질소원, 황원, 인산염 등의 영향을 조사하였다. 재조합 방선균내에 도입된 plasmid는 대장균이나 효모에 비해 다소 불안정한 양상을 보였으나, 원 균주와 달리 IPNS 유전자가 대수기부터 생산되어 idiophase에도 효소 역가를 계속 유지할 수 있었다. 그리고, 재조합 방선균에서도 탄소원이나?? 있었으며, 인산염에 의해서도 항생물질 생산이 억제되었다. 그러나, glucose나 인산염에 의해 IPNS의 생산이 억제되지 않는 것으로 보아, 이러한 억제 현상은 IPNS 유전자의 발현 조절에 의해서 일어나는 것은 아니라는 사실을 알 수 있었다. 한편, 재조합 미생물은 원 균주와 마찬가지로 무기황원을 이용하였으며, 특히 cysteine에 의해 IPNS 유전자 발현이 억제되어짐을 관찰할 수 있었다.
제 4 세부과제에서는 식물세포내로 외부유전자를 도입하여 안정적인 외래단백질의 생산을 시도하였다. 우선 model system으로 E.coli의 유전자인 GUS(β-glucuronidase)를 담배세포로 도입하여 세포배양에 따른 세포내 안정성을 조사하였다. 6개월 이상의 장기적인 배양에 있어 도입된 GUS는 지속적인 selection pressure(Kanamycin의 첨가)을 유지하여 주었을 경우 매우 안정된 발현이 유지되었음을 확인하였으나 아무런 selection pressure을 주지 않은 상태로 장기간 배양을 하였을 경우 비록 완전히 도입된 유전자의 발현이 사라지지는 않으나 selection pressure를 지속한 culture에 비해 현저히 낮은 유전자의 발현 즉 안정성이 떨어짐을 알 수 있었다.

Recent developments and advances in the area of genetic engineering have provided new tools for the economic production of valuable products such as enzymes and antibiotics. In this research, a comprehensive study on the bioprocess design parameters for genetically engineered recombinant cell cultivation, bioprocess-related knowledge bases, the bioprocess kinetics, and optimization strategies required for the production of gene products have been established. Since it is now possible to use various kinds of cells from microbial cells to plant and animal cells, E.coli, yeast, Streptomyces, and plant cells were employed for the analysis of recombinant cell culture processes. The major achievements of each research subject are as follow.
The first subject concerns the investigation of the effect of genetic and environmental factors on recombinant E.coli fermentation. Using a homologous series of plasmids which have the same replication origin, the influence of the nature of plasmids properties and culture conditions on fermentation process parameters were studied. The effect of induction temperature and the dissolved oxygen level on cell growth, cell viability, plasmid stability, and cloned-gene productivity have been intensively investigated. It was found that the optimal temperature for maximizing target protein production could be changed depending on the interaction between the host-cell metabolism and the cloned-gene expression.
The partitioning of recombinant human interleukin-2 in PEG 8000-dextran 38800 aqueous two-phase system has been investigsted in order to improve the recovery yield from the recombionant cells. The addition of sodium dodecyl sulfate singificantly enhanced the partition coefficient and recovery yield. The results of this work indicate that PEG-dextran two-phase partitioning might provide a simple way for sthe recovery and partial purification of other recombinant proteins.
Molecular level models based on Lee & Bailey model have been used to evaluate the effectiveness of several different configurations of control system of the cloned gene product expression. Both single and dual-repressor situations were considered, employing genetically structured models for the lac and lambda pR promoter. In the second research subject, the optimization and control of recombinant yeast fermentation processes were investigated using recombinant S. cerevisiae strains producing invertase(SUC2) and human lysozyme(HLZ). THe rate of cell growth and product synthesis was analyzed as a function of glucose concentration and the rate constants were determined from a series of batch and fed-batch experiments. The cell growth model based on cybemetic concepts were developed. Simulation results of the proposed model equation was found to fit very well with the experimental data.
In order to optimize the recombinant yeast fermentation processes, a novel optimization method was developed using a conjugate gradient method the strategies for maximizing the objective function, in this case invertase production, were obtained. The best strategy was the increase of cell growth in the early stage followed by the induction of the invertase production.
Lysozyme production was also investigated. At higher glucose concentration, the production level of lysozyme from ADH1 promoter was 220 units/ml and more than 90% of the produced lysozyme was secreted into the fermentation broth. Under anaerobic conditions, the lysozyme level was 1.7 time higher than that obtained under aerobic conditions. When the efficiency of secretion signal sequences was compared, it was found that cell growth rate, lysozyme secretion rate, and lysozyme production with MF α1 were 1.2-, 2.3-, and 1.50 fold higher than that obtained from mouse αed. A part of gene cluster for cephamycin C synthesis has been cloned from the genomic library of Str. clavuligerus ATCC27064 in pKC505, E.coli/Streptomyces shuttle cosmid vector, which was screened by oligonucleotide probe of the homologous region in isopenicillin N synthetase (IPNS) gene. Using the cloned gene as template, IPNS gene was amplified by polymerase chain reaction, and then inserted into the polycloning site of E.coli plasmid pUC18. It was confirmed that the cloned IPNS gene was efficiently expressed in E. coli DH5 host cell. In order to construct the recombinant Streptomyces, IPNS gene was transferred into pIJ702, a high copy plasmid for Streptomyces. The recombinant Str. lividans TK24 with IPNS gene showed the enzyme activity, and the Str. clavuligerus recombinated with this gene could produce antibiotic 3 times more than the original strain.
In fermentative studies using the recombinant Str. clavuligerus, it was found that these strains could not maintain the plasmid stably differently from other host microorganisms, but it was observed that IPNS was produced from logarithmic phase and its activity was maintained during idiophase. The catabolite repression by carbon sources and nitrogen sources was observed in this recombinant Str. clavuligerus, and inorganic phosphate could also inhibit the antibiotic production. However, IPNS production was not repressed by glucose and inorganic phosphate. The recombinant strain could also utilize inorganic sulfur rather than organic sulfur, and the repression of IPNS synthesis by cysteine was observed.
In the fourth research, we investigated the possibility of production of animal proteins by recombinant DNA tobacco cell culture. We have established transgenic tobacco cell lines carring β-glucuronidase(GUS) of E.coli as a model system and its long term stability in tobacco cell culture was investigated. The GUS gene introduced into tobacco cells were stably maintained during the long term cultivation (over 6months) under the consistent selection pressure of kanamycin. While the cells under no selection pressure showed lowered gene expression of GUS that is less stable in long term cultivation.
From these results, human superoxide dismutase(SOD) and mouse transforming growth factor(TGF-β) were cloned into the plant overexpression vector pGA 643 which were later introduced into Agrobacterium tumefaciens by freezing -thaw method, The NPT Ⅱ, Wouthern, Northen analyses and activity staining confirmed the expression of human SOD gene and the biological activity of SOD. However, we could not confirm the production of the mouse TGF-β by transgenic tobacco cell culture. Southern and Northern analyses showed expression of TGF-β upto RNA level but Western blot and biological activity test failed to confirm the production of the TGF-β required posttranslation and modifiaction to be biologically active. It has been oru final goal to establish recombinant DNA plant cell culture system for the production of animal proteins replacing the expensive animal cell culture system. However, it was found that complicated proteins such as TGF-β need some modification in gene to be properly expressed in plant cells and special expression systems.
