HSA의 경우fluorescamine을 이용한 형광분석법에 의하여공유결합도를 정량적으로 분석하 였고전기영동을 통하여 공유결합물의 분자량을 측정하였다. 실험결과 HSA의 경우 단 백질내 Iysine기와 활성화 PEG의 몰비율은 1:5가 최적이었고, pH는 9-11에서 공유결합도가 높았으며 최적반응시간은 50분으로 나타났다. 반응도중 (10-20분)에 가역적인 중간체 (intermediate)가 형성됨을 형광분석을 통해 관찰할 수 있었다. β-galactosidase의 경우 활성과 PEG와의 결합을 통해 역가가 상실됨을 볼 수 있었다. 20-6O℃의 은도변화에 따른 안정성을 40℃에서 공유결합물이 다소높은 안정성을 보일 뿐 큰 차이는 없었고, PH 3-10 사이의 효소 안정성은 활성화 PEG와의 공유결합체가 다소 높게 평가되었다. ATPS조건에서 일반 β-galactosidase는Na2s04가 주성분인 하층으로 이동하였으나 공유결합체는 상층으로 이동하였고 β-galactosidase와 활성화 PEG와의 반응 몰비를 중가시킬수록 90%이상의 효소를PEG층으로 이동시킬 수 있었다. 이러한 변형 단백질의 생성, 분석 및 이용 기술은 기존 단백질의성능 향상과 생물공정에의 응용이 가능할 것으로 사료된다.

In this study, using SC(succinmnidyl carbonate)-PEG as an activated PEGand HSA (human serum albumin)and β-galactosidase as model proteins, theconjugation characteristics were investigatedand the feasibility of applying the reactionto a bioseparation process was evaluated.With HSA the optimum reaction condition interms of pH, molar ratio of SC-PEG andIysine residue, and reaction time wasdetermined from the degree of modiflcation datadeduced from fluorescence assay andmolecular weight estimation using SDS-PAGE. The result indicated that pH 9-11, 1:5(Iysine: SC-PEG) molar ratio, and 50 minutes~reaction time was optimum for thehighest degree of modification. The conjugationreaction was fast (t1/2 = ca. 5 min),but at 10 to 20 minutes of the reaction time anintermediate of reversible nature wasseen to form. With β-galactosidase, some loss inenzyme activity was observedafter the conjugation. The loss appeared proportional tothe degree of modification,which suggested the active sites are somehow consisted ofamino group-containingresidues. Compared to those of the intact enzyme,temperature(20-60℃) and pH(3-10) stability of the conjugate were not as muchimproved. However, when theconjugates were exposed to ALPS (aqueous two phaseseparation) system composedof regular PEG 4000 and Ma2SO4 they showed markedlydifferent partitioningbehavior compared to that of the intact enzyme. The intactenzymes stronglypartitioned to the bottom phase when regular PEG was used as aphase component.However, when SC-PEG was used as a supplement to the regularPEG and allowedto conjugate with the enzyme, the conjugated enzymes reversed thepartitioningdirection to the top phase. The more SC-PEG was used, the higherpartitioncoefficient was resulted, and greater than 90% recovery yield was attained. Insum,the PEG conjugation technique can improve protein properties and be applied toproteinseparation process.
