모든 생물은 주위환경의 다양한 돌연변이원(방사선, UV, 화학물질등)으로부터 항상 노출되어 있으며 이들로부터 손상을 입게 되면 즉각적으로 반응하여 복구하게 된다. 이중에서 UV는 DNA염기에 직접 작용하여 pyrimidine dimer를 형성하여 해당되는 유전자에 손상을 주게 된다. 그러면 이 dimer는 관련된 수리 유전자들에 의해서복구가 되는데, 본 실험에서는 P 전이인자를 이용하여 돌연변이체를 만들고 이 수리기구(repair mechanism)에 직접 또는 간접으로 이상이 있다고 생각되는 계통들을 선별하였

UV 감수성 유전자(P에 의해서 돌연변이가 된 유전자)라는 말은 곧 생물이 UV에 노출되었을 때 생기는 유전자의 손상을 수리하는 유전자이다. 그래서 이 유전자에 돌연변이가 생기면 UV 감수성을 보이게 된다. 특히, 재조합과 DNA수리의 연관성에 비추어 재조합빈도를 알아보기 위해서 여러 가지 유전분석을 하였다. 이들 pus계통은 대조군보다 다소 높은 재조합 빈도를 보였다. 흥미롭게도 pus25 계통에서 수컷이 더 눈색의 침착도가 암컷보다 높았는데 이 말은 수컷에서 이 유전자가 더욱 특이적으로 많이 발현된다는 말이다. 그래서 일반적으로 노랑초파리에서는웅성재조합(male recombination)이 일어나지 않는데, 이 계통(pus25)은 웅성재조합이 적은비율이지만 발생하였다(0.14%). 그리고 P에 의해서 표지된 이 유전자들의 일부를 포함하는 유전자 절편을 plasmid rescue를 이용하여 확보하였다. P의 삽입위치는 pus25, pus248은 3번 염색체이고, pus92, pus181은 X 염색체이다.본 연구를 통해서 4가지의 pus계통이 확보되었으며, 이들 중 pus25는 웅성재조합현상을 보였다. 향후 이 계통을 이용하여 초파리와 같은 고등생물의 DNA수리 기구와 웅성재조합 억제기구에 대한 중요한 실마리를 찾을 수 있으리라 기대한다.

All organisms live in an environment that poses continualthreats to their genetic mareial. Ionizing radiations, utraviolet light from the sun andvarious chemical gents(mutagens) cause alterations in it. In particular, mutations fromthe UV light involve pyrimidine dimers in the DNA sequence affected. In a normalorganism, these dimers are replaced with normal bases via a serious of DNA repairmechanisms. In this study, for the purpose of identifying DNA repair-related genes inD. melanogaster, a wide variety of mutants have been produced by P element- mediatedinsertional mutagenesis. Of these 4 UV-sensitive lines which are thought to bedefective directly or indirectly in UV-repair mechanisms have been screened byUV-sensitivity tests. Several genetic studies involving recombination frequencyanalyses(female), and insertion site determination by molecular techniques as well asmale recombination tests(MRT), have been performed. These lines (called pus lines)showed somewhat different recombination rates from those of the normal lines. Also,plasmid rescue using the P-tagged genes has been performed by use of severalenzymes which cut both an inner part of intact P and Drosophila genome. Theinsertion sites of P in each pus line are 3rd chromosome(pus25, pus248), andX-chromosome(pus92, pus181). The relative UV-sensitivity of each line is0.734(pus25), 0.780(pus92), 0.860(pus181), and 0.611(pus248), respectively. Especially,one of the intriguing features of pus25 is that its male recombination hasoccurred(approximately 0.14%). In addition, recombination rates(female) obtained weresomewhat higher than those of the control groups, by 7.1% (pus25), 3.46%(pus92), and6.09%(pus181), respectively.From this study, we have constructed 4 UV-sensitive lines, pus lines, containing 1male recombination-occurring line. It is expected that these lines may give us a clueto DNA repair mechanisms in D. melanogaster. Espescially, the line pus25 may play akey role in elucidating the suppression mechanism of male recombination. Moreover,several putative clones via plasmid rescue have been obtained which contain fragmentsof Drosophila genome. These clones will show us what the genes and/or the repairmechanism is like in D. melanogaster.
