For the develooment and practical application of antitumor and antimetastatic prescriptions and effective substances from KSBS and its constituents, the antitumor and antimetastatic effects of enami-sojeokbaekchulsan(KSBS) and KSBS-I (Hongsam sojeok bakchulsan) were evaluated experimentally. In antitumor study of KSBS, $10^-3$g/ml of KSBS exerted significant antitumor activity against B16-F0, A549 and SNU-1. Medicinal herbs inhibiting cell growth such as B16-F0, A549 and SNU-1 up to above 30% were Orostachys Herba, Lonicerae Flos, Taraxaci Radix, Pinelliae Rhizoma, Scutellariae Radix, Curcumae Radix, Lithospermi Radix, Oldenlandiae diffusae Herba. Above 50% inhibition of protein kinase C was shown in 125ug/ml of SBS and KSBS, Orostachys Herba, Lonicerae Flos, Lithospermi Radix, Coicis Semen, Aurantii nobilis Pericarpium. KSBS significantly inhibited tumor weight of B16-F0 and lengthened survival time (T/C %) to S-180 to 147% of control and also suppressed the platelet aggregation. KSBS and KSBS-I significantly inhibited topoisomerse-I at 125 ㎍/㎖. In antimetastatic study $10^-3$ of SBS & KSBS, Lonicerae Flos, Taraxaci Radix and Oldenlandiae diffusae Herba inhibited cell adhesion of A549 to 50%. KSBS and KSBS-I significantly inhibited pulmonary colonization by B16-F0 and histopathological metastasis as compared with control data. KSBS significantly inhibited the invasion of B16-F0 especially in water extract, MeOH layer and EA layer. Tanshione ⅡA from Salviae miltriorrhiza Radix, ursolic acid from Oldenlandiae Herba, farneciferol C and galbanic acid from Ferula assafoetida were isolated and their structures were identified. $IC_50$ of tansione ⅡA, ursolic acid, farneciferol C and galbanic acid were approximately 4 ㎍/㎖, 4 ㎍/㎖, 5 ㎍/㎖,10 ㎍/㎖ against cancer cells such as A549, SK-OV-3, SK-MEL-2, XF-498 and HCT-15. Especially ursolic acid, farneciferol C and galbanic acid inhibited angiogenesis to 60% according to CAM assay. KSBS and KSBS-I protected side-effects such as crypt injury in small intestine induced by irradiation of 4Gy liniac. from the above results SBS, KSBS and KSBS-I had antitumor and antimetastatic effects and KSBS-I was more effective, which suggest KSBS and KSBS-I can be applied to the treatment for cancer.
