새로운 구조를 갖는 생분해성 고분자 재료를 합성하여 이들의 구조 및 물성을 확인하여 높은 반응성 및 생체적합성을 요구하는 의료용 소재의 제조방법을 확립한다. 다양한 물성을 통해 의료용 소재로서 커다란 장점을 지닌 삼중블럭 형태의 의료용 소재를 합성하기 위한 기존의 coupling 방법은 반응률이 낮고 toxic한 촉매 혹은 작용기가 잔존하는 단점이 있다. 본 연구에서는 반응성이 향상된 의료용 생분해성 삼중블럭 공중합체 및 그의 제조방법에 관한 것으로 폴리락타이드(혹은 폴리글리콜라이드 혹은 폴리카프로락톤)를 합성한 후, 반응성이 매우 큰 아실할라이드기를 갖는 폴리에틸텐글리콜을 합성하여 이들을 coupling 반응함으로써 높은 반응률을 나타내는 공중합체 제조하는 것이다. 고분자 재료와 세포외기질의 복합화 기술 개발을 통하여 생체적합성 및 생체기능성이 우수하고 안정적인 인공각막 및 결막조직의 재생을 위해 고분자 지지체의 소재 설계, 합성 및 제조하고 각막세포를 추출 및 배양하여 고분자/세포 복합화와 복합체의 체외특성 평가를 실시하고, 지지체를 결막조직에 이식하여 체내 특성 평가를 실시하였다. 먼저 적절한 분해성 고분자인 PLGA 50:50을 이용하여 다공성의 PLGA 지지체를 제조하고, collagen, Hyaluronic acid 및 Amniotic membrane을 이용하여 PLGA/collagen/hyaluronon/amnion hybrid 지지체를 제조하여 높은 기계적 강도와 세포적합성을 가지는 지지체를 제조하였으며, 그 외 우수한 생체적합성을 가지는 Collagen, Hyaluronic acid의 천연 고분자 재료를 이용하여 지지체를 제조한 후 세포점착률 및 증식률, 그리고 세포 배양시 지지체/세포의 형태변화를 주사현미경(SEM) 및 면역세포학적 검사(H&E staining)를 통하여 확인하였다. 일반적으로 조직공학용 고분자 지지체는 우수한 기계적 물성 및 생체적합성, 적절한 생분해성과 특정 세포와만 상호작용 할 수 있는 능력 등의 특성을 가져야 한다. 본 연구에서 먼저 새로운 glycerol 함유한 triacrylated oligolactide기반 고분자 network을 제조하기 위하여 acrylic acid(AA) 및 poly(ethylene glycol) (PEG)-acrylate (PEG-Ac)의 두 개의 화합물을 사용하였다. 제조된 무독성 고분자인 GL-AA 및 GL-PEG 지지체 말단에 세포 특이성을 같은 bioactive 펩타이드 리간드인 RGD를 그라프트함으로서 조직공학을 위한 고분자 지지체를 제조하였다. 이러한 생체적합성 및 생분해성 고분자지지체의 표면, 벌크 특성과 세포와의 상호작용을 연구하였다 멤브레인 표면에서의 콜로이드 상호작용의 영향을 고찰하기 위해 모델 콜로이드인 라텍스의 dead-end 한외여과를 다루었다. 투과플럭스 결과로부터 정전상호작용의 실험적 변수인 용액의 이온화세기가 감소하면, 전기이중층의 확장으로 막표면에 쌓이는 케?층의 두께가 증가하는 사실을 알 수 있었다. 선형 포아슨-볼쯔만(Poisson-Boltzmann) 정전위장에 특이점 해법을 적용하여 먼거리 정전상호작용 에너지를 결정하고, 메트로폴리스 몬테카를로 모사를 시행하여 벌크용액에서 콜로이드 입자의 확산계수를 산출하였다. 서스펜션의 입자농도와 이온화세기에 따른 입자 확산계수의 정량적 거동을 확인하였고, 투과플럭스와 확산계수로부터 농도분극층에서의 입자의 농도분포를 예측하였다. 계면동 전기 효과에 의해 발생되는 흐름전위에서 헤름홀쯔-스몰루쵸스키 식으로부터 막기공에서의 제타전위로 정의되는 멤브레인 전위를 결정하였다. 멤브레인 여과가 진행할수록 정전 반발력이 증가함에 따른 전기이중층의 확장은 투과플럭스의 증가와 함께 멤브레인 전위의 감소를 나타냈다.

Poly(ethylene glycol) (PEG)/polylactide(PL)/poly(ethylene glycol) (PEG) triblock copolymers have been reported as a drug delivery material due to it's biocompatibility, biodegradability, sol-gel properties and so on. But these triblock copolymers have a low reaction yield, toxic functional group and catalyst. We have prepared these triblock copolymers having a high reaction yield and non-toxicity, and investigated their properties. In this research, 2-arm PL with various molecular weight were synthesized as a hard segment. End group of methoxy poly(ethylene glycol)(mPEG) were functionalized to acid halide and used as a soft segment. PEG/PL/PEG triblock copolymers were synthesized by coupling of these two materials in the presence of pyridine. We have prepared these triblock copolymers with various molecular weight and certificated the coupling reaction of end group of PL and mPEG by FT-lR and $^{1}H$-NMR. mPEG/PL/mPEG triblock copolymers had higher thermal degradation temperature than mPEG and PL. $T_m/; and/; {\Delta}H_m$ of mPEG/PL/mPEG decreased as molecular weight of mPEG increased. Hydrophilic property increased as molecular weight of mPEG increased and molecular weight of PL decreased in mPEG/PL/mPEG. Hydrolytic degradation showed that the change of mPEG/PL block ratio of mPEG/PL/mPEG was high when the molecular weight of mPEG was high. Corneal transplants and conjunctival autograft transplantation commonly are performed for the successful surgical treatment of ocular surface diseases, but the demand for the cornea tissue exceeds the supply and the recurrence rate is above 30%. In this study we investigated the effects of the modified PLGA scaffolds with collagen, amniotic membrane and hyaluronic acid for ocular surface regeneration. Porous PLGA (M.W.: 110,000) scaffolds were prepared by a salt casting particulate leaching method with the salt size 300$10^6$cells/ml. These structures were maintained in culture for the following periods: 1, 4, and 7 days. Histology was done to analyze for new tissue formation on our 3D scaffolds. Briefly, fixed cultures on scaffolds were stained with hematoxylin and eosin (H&E) in situ. SEM visualized the morphology of cells growing throughout the scaffolds. All the modified scaffolds enhanced cell adhesion as compared to PLGA control and demonstrated better for cell proliferation. A random distribution of cells throughout the lattice was observed in histological cross-sections of cell-culture scaffolds. Migrating cells on the modified and unmodified scaffolds appeared to be healthy and well adhered to the scaffolds. Collagen/amnion/hyaluronon scaffold showed the highest cell adhesion and proliferation. SEM pictures of the scaffolds showed that epithelial cell and stromal fibroblasts were well attached and spread on the surface. This study was also performed to investigate histopathologically the effect of the modified PLGA scaffolds in ocular surface wound. Conjunctival wound was created in white rabbits bilaterally and applied collagen/hyaluronon PLGA scaffold for 4 weeks. The regeneration of ocular surface tissue was evaluated after treatments in conjunctival wound. At postoperative 4 weeks we observed the proliferation of normal collagen and cells at the wound area. Our results indicate that tissue engineered design of this modified scaffold and in vitro cell culture system may allow it to be formed whole ocular surface tissue. Biodegradable polymers are used in the field of tissue engineering as scaffolds on which cells may grow and form tissue. These polymer materials are relatively biocompatible and appropriately biodegradable, but they lack the ability to interact biospecifically with cells. In this study, we have synthesized three types of glycerol (G)-lactide (L) triols (GL3, GL9, and GLI5) with controlling each molar ratio of G and L and then reacted the GL triol with acryloyl chloride to get GL triacrylate(GL-Ac). Finally photo-crosslinked network films having the GL-Ac and acrylic acid (AA) or poly(acrylic acid) (PAA) were prepared by UV photo-polymerization. The bulk and surface characteristics of GL-AA network and GL-PAA semi-lPN films were evaluated by gel content, water absorption, Tg, Td, carboxyl group analysis, electron spectroscopy for chemical analysis (ESCA), and water contact angle. GL-AA and GL-PAA networks showed good bulk properties and AA-enriched & hydrophilic surface. Meanwhile, poly(ethylene glycol) (PEG) has been employed as a spacer in tissue engineered scaffolds because of its remarkable nonadhesion towards proteins and cells. To induce selective cell adhesion upon such cell-resistant scaffolds, reactive functional pendent groups on the material can be directly employed for grafting peptide ligands(L) such as Arg-Gly-Asp(RGD). Lactide-based PEG polymer network films (GL-PEGs) were first prepared by UV polymerization of a glycerol-lactide triacrylate (GL-Ac) with PEG monoacrylate (PEG-Ac) to use as specific cell-adhesive scaffolds for tissue engineering. Then, activated GL-PEG network films(GL-PEG-SC) were prepared by using N,N'-disuccinimidyl carbonate (SC) for covalent attachment to peptide ligands. Finally, GL-PEG-RGD networks were made from GL-PEG-SC. Surface and bulk properties of the obtained GL-PEG-L networks were examined by ATR-FTIR, ESCA, contact angle, SEM and gel content, water absorption, DSC, TGA, and Instron, respectively. All GL-PEG-RGD networks displayed more adhesion and spreading of fibloblasts than GL-PEG. This is attributed to the incorporation of the bioactive RGD ligand. These new GL-PEG-RGD polymer networks may be useful as scaffolds for tissue engineering for specific cell adhesion. In order to investigate the effects of the colloidal interaction in the membrane filtrations, the dead-end ultrafiltration of latex colloids was conducted with fully retentive membranes. Experimental results concerning the permeate flux during the filtration indicate that the cake layer thickness increased with the decrease of the ionic strength, due to the expanded Debye double layer thickness around the particles. The concentration dependence of the gradient diffusion coefficient of colloidal particles has been examined as a function of solution ionic strength. The Metropolis Monte Carlo method was employed so as to determine the thermodynamic coefficient, and the hydrodynamic coefficient was evaluated from the previously developed relation. The long-range electrostatic interactions between the particles were determined by using a singularity method, which provides accurate solutions to the linearized electrostatic field. We found that both the permeate flux and the particle diffusion are related to determine the concentration distribution above the cake layer. The streaming potential generated by the electrokinetic flow effect within electric double layer of charged channel is applied to analyze the membrane potential by using the Helmholtz-Smoluchowski equation. The influence of physicochemical parameters upon the filtration has been examined with simultaneously monitoring of the membrane potential as well as the permeate flux. With increasing of solution ionic strength, the permeate flux decreases but the membrane potential increases. It is obvious that the increase of solution ionic strength provides a compact cake layer due to the shrinkage of Debye length.
